Abstract

The invention comprises compounds of the general formula <FORM:0822055/IV (a)/1> (wherein either (1) R represents a mono- or dialkylamino or heterocyclic amino group, A a dialkylamino or heterocyclic amino group, R1 a methyl group and R11 hydrogen, or (2) R represents a mono- or di-alkylamino or heterocyclic amino group, A a morpholino group, R1 hydrogen and R11 hydrogen or a methyl group, or (3) R represents a monoethylamino or pyrrolidino group, A a piperidino group and R1 and R11 both hydrogen), and the preparation thereof by (a) (when R is dialkylamino or heterocyclic amino) reacting an aminoalkyl halide Hal-CHR1-CHR11-A with an alkali metal derivative of a diphenylacetamide compound, of the formula (C6H5)2-CMe-CO-R (Me=alkali metal), or (b) (when R11=H) reacting an acid halide A-CH2-CHR1-C(C6H5)2-CO-Hal with a mono- or di-alkylamine or a heterocyclic amine (preferably two molecular proportions thereof) at a temperature below the cyclization temperature of the acid halide. When R1 and R11 are not both hydrogen, process (a) yields a mixture of isomers, which may be separated by making use of the difference in solubility of the bases or their salts in certain solvents, e.g. of the hydrochlorides in acetone or the bases in ether/petroleum ether. Process (b) may be combined with the preparation of the acid halide by treating the corresponding amino acid, or a mineral acid salt thereof, with excess of a halogenating agent (e.g. thionyl chloride or phosphorus trichloride). Examples and a table describe compounds containing various combinations (within the limits of the definitions above) of the following meanings of R and A; R=methylamino, ethylamino, isopropylamino, n-butylamino, dimethylamino, diethylamino, pyrrolidino, piperidino, 2-methyl-, 2 : 6-dimethyl- and 2-methyl-5-ethyl-piperidino, morpholino; A=morpholino, piperidino, dimethylamino, pyrrolidino. The hydrochlorides of some of the products are described. Specification 772,921 is referred to.ALSO:Analgesic compositions contain, in admixture with a physiologically inactive diluent material, one or more compounds of the general formula <FORM:0822055/VI/1> wherein either (1) R represents a mono- or dialkylamino or heterocyclic amino group, A a dialkylamino or heterocyclic amino group, R1 a methyl group and R11 hydrogen, or (2) R represents a mono- or di-alkylamino or heterocyclic amino group, A a morpholino group, R1 hydrogen and R11 hydrogen or a methyl group, or (3) R represents a monoethylamino or pyrrolidino group, A a piperidino group and R1 and R11 both hydrogen. An example describes tablets and a powder containing 1-(4-N-morpholino-4-methyl-2 : 2-diphenylbutyryl)-pyrrolidine.
