Abstract

The invention comprises pyridyl-(3)-methyl dihydrogen phosphate and alkali metal, alkaline earth metal and ammonium salts thereof. The products may be obtained by phosphorylating pyridyl-(3)-methyl alcohol by treating it with a mixture of phosphorus pentoxide and phosphoric acid at an elevated temperature, hydrolysing any polyphosphates contained in the resulting mixture by heating same in acid solution, passing the hydrolysate through a cationic exchange resin and separating off that fraction of the eluate which contains the required pyridyl-(3)-methyl dihydrogen phosphate and, if desired, treating same with a precipitating agent or, where a salt is required, carrying out the defined process and treating said fraction or the pyridyl-(3)-methyl dihydrogen phosphate precipitated therefrom with an alkali metal hydroxide, alkaline earth metal hydroxide or ammonium hydroxide followed by isolation of the resulting salt by precipitation or concentration. The phosphorylation may suitably be conducted at 60 DEG C. and hydrochloric acid may conveniently be used in the hydrolysis step. Suitable cationic exchange resins are sulphonic acid resins in their acid form. Acetone may be used as the precipitating agent in either case. Before feeding the hydrolysate on to the column containing the cationic-exchange resin it is preferred to remove the bulk of inorganic oaterial contained therein by treating it with phenol and partitioning the phenolic layer between ether and water. The resulting aqueous layer contains the required product and is then fed on to the column. Examples are given for the production of the pyridyl-(3)-methyl dihydrogen phosphate and its mono- and disodium salts. The products are stated to be effective vasodilators.
