Abstract
1529097 Modified hGH SCRIPPS CLINIC & RESEARCH FOUNDATION 6 Sept 1976 [8 Sept 1975] 36893/76 Heading C3H A method of producing modified human growth hormone (hGH) of increased biological potency comprises digesting hGH with a proteinase to remove amino acid residues 137 to at most 149 and to at least 147 inclusive, and recovering the modified hGH. Preferably the proteinase is of bacterial origin, e.g. subtilisin. In an example hGH is digested in a solution of sodium phosphate containing subtilism. The reaction is terminated by addition of diisopropylfluorphosphate and the solution is then dialysed. The dialysed digest is then fractionated on DEAE-cellulose and three hGH fractions result.
