1488469 Lactate determination BOEHRINGER MANNHEIM GmbH 2 July 1976 27658/76 Heading G1B Process for the quantitative determination of L-lactate in a sample by reaction with nicotinamideadenine-dinucleotide (NAD) in the presence of lactate dehydrogenase (LDH) with the formation of reduced nicotinamide-adenine-dinucleotide (NADH) and pyruvate, reaction of the latter with glutamate in the presence of glutamate-pyruvatetransaminose (GPT) with the formation of alanine and an α-ketoglutorate in alkaline buffer solution and measurement of the NADH formed to indicate the amount of L-lactate in the sample, wherein the reaction with the glutamate is carried out in the presence of at least 0.4 mol/litre of carbonate or bicarbonate. A suitable reagent for the above process comprises LDH, GPT, NAD and glutamate and is characterized by a content of 0À40-1À2 mol/litre of a carbonate buffer of pH 9À5-10À5. The reagent may also contain sodium azide. The process may also be used to determine the L-alamine content in a sample.
