Abstract
1472667 1-Substituted 5-methyl-5-azauracils UPJOHN CO 25 April 1975 [20 May 1974] 17206/75 Headings C2C and C2P Novel 1-substituted 5-methyl-5-azauracils of the general formula wherein each of R and R<SP>1</SP> is a hydrogen atom, or each of R and R<SP>1</SP> is a C 2-18 hydrocarboncarboxylic acyl group, optionally substituted by halo, nitro, hydroxy, amino, cyano or thiocyanato, or C 2-8 alkoxycarbonyl, or one of R and R<SP>1</SP> is hydrogen and the other is phosphono or optionally substituted hydrocarbon-carboxylic acyl or alkoxycarbonyl as defined above, are prepared (a) when each of R and R<SP>1</SP> is a hydrogen atom, by cultivating Streptomyces platensis var. clarensis, having the identifying features of NRRL 8035, in an aqueous nutrient medium containing C and N sources under submerged aerobic conditions until substantial antibiotic activity is imparted to the medium; (b) when neither R nor R<SP>1</SP> is a phosphono group and at least one of them is other than hydrogen, by acylation of the product of (a); (c) when R is hydrogen and R<SP>1</SP> is a phosphono group, by phosphorylation of the product of (a); and (d) when R is a phosphono or acyl group and R<SP>1</SP> is a hydrogen atom, by phosphorylation or acylation of the 51-trityl derivative of the product of (a) and removal of the trityl group by mild acid hydrolysis. Antibacterial compositions comprise, as active ingredient, a 1-substituted 5-methyl-5-azauracil of the above general formula, together with a pharmaceutically acceptable carrier.
