Abstract

1442677 Microbial lipase TAKEDA YAKUHIN KOGYO KK 19 Sept 1973 [20 Sept 1972] 43943/73 Heading C3H [Also in Divisions A5 C5 and G1] A microbial lipase, which accumulates in the culture medium during cultivation of microorganisms of the genus Phycomyces, particularly P. nitens and P. blakesleeanus, and whose enzyme activity is enhanced in the presence of bile, is characterized as follows: maximum ultraviolet absorption spectrum at 275-280 millimicrons, significant infra-red absorption spectra at 3À0, 3À38, 6À05, 6À55, 6À88, 7À15, 8À12, and 9À3 microns, soluble in water and aqueous salt solution of 0À01 to 0À1 of ionic strength, insoluble in alcohols, acetone and ether, optional activity between pH 5 and 8 and 35-41‹ C., stable over pH range 4 to 10 at 23‹ C., stable at temperatures up to 40‹ C. but inactivated by 30% at 45‹ C. and almost completely inactivated at 50‹ C. at pH 7, not inhibited by diisopropyl fluorophosphate, EDTA or p-chloromercuribenzoate (specified conditions) and exhibits an at least 50% increase in lipase activity on olive oil in the presence of dog bile diluted to 1/200. Characteristics from amino acid analysis: no cysteine residue, two S-S bonds, two trytophan residues, no saccharide and no lipid. The enzyme is recovered from the culture medium and purified by conventional methods. The Specification discloses pharmaceutical preparations (see Division A5) including in combination (1) the lipase and bovine bile powder and (2) the lipase, microbial acid protease, amylase and cellulase, and pancreatin.
