Abstract

1414752 {[(diaminopteridyl)methyl]methylamino}benzoylglutamic acid INSTITUTUL ONCOLOGIC 20 March 1974 12465/74 Heading C2C N - [p - {[(2,4 - diamino - 6 - pteridyl)methylmethylamino}benzoyl]-glutamic acid is prepared by (i) treating a salt of 2,4,5,6-tetraaminopyridine with 1,3-dihydroxyacetone bisulphite in aqueous media with air-bubbling and in the presence of a thio-alcohol as catalyst (preferably cysteine) and an oxidation agent (preferably selenium dixide (ii) reacting 2,4-diamino-6- hydroxymethylpteridine obtained with a halogenation agent preferably thionyl chloride or phosphorus oxychloride) in an inert media (preferably chloroform or methylene chloride) in the presence of a basic catalyst (preferably pyridine or triethylamine) to form the 6-halomethyl derivative and (iii) condensing this compound with N-[p-methylaminobenzoyl]-glutaric acid in buffered aqueous media. The D, L or racemate compound is prepared by using the appropriate glutamic acid in stage (iii).
