Abstract

1414235 Purifying cyclodextrin-producing enzymes HAYASHIBARA KK S OKADA and S KITAHATA 30 Oct 1973 [30 Oct 1972] 50381/73 Heading C3H A method of purifying a cyclodextrinproducing enzyme involves contacting an aqueous solution of the contaminated enzyme, e.g. the filtrate of a culture broth of Bacillus macerous or Bacillus megaterium, with starch and/or cyclodextrin (cycloheptadextrin, cyclohexadextrin or crude cyclodextrins) and then eluting the enzyme that has been absorbed thereon. In a preferred procedure, there is added to the initial impure solution ammonium sulphate in a concentration of 25-30% saturation or a hydrophilic organic solvent (ethanol, acetone or isopropanol) up to 30% v./v., as a result of which any co-existing protein is rendered turbid and may be filtered off, starch is introduced and the resulting starch suspension is passed through a layer or column of starch including a small amount of diatomaceous earth. The starch layer is washed with an aqueous solution of ammonium sulphate and/or a hydrophilic organic solvent, the washed starch is suspended in dilute aqueous sodium hydrogen phosphate solution and a solution of the purified enzyme is recovered by filtration.
