Abstract

1356346 Immunological testing PHARMACIA AB 26 May 1971 [27 May 1970] 17118/71 Heading G1B A reagent-combination for use in carrying out tests on samples of human origin utilizing an immunological reaction between an antigen (1) and an antibody (2), one of which is of ovine or bovine origin comprises either antigen (1) or antibody (2), and non-human animal gamma globulin (3) for reaction with heterophile antibodies present in the sample under test, the non-human animal gamma globulin (3) being contained separately from (1) or (2) or being mixed with (1) or (2). The antigen (1) or antibody (2) may be chemically or physically bound to carrier particles (for example co-polymers of dextran and epichlorhydrin, or to microcrystalline cellulose or cellulose in the form of a paper disc or to a tube of plastics material) and may be radio-actively labelled or labelled with colour producing atom or group, for example with fluorescein. The non-human gamma globulin (3) may be horse, sheep or bovine. Examples relate to determination of reagin-Ig directed against cow epithelium where the reagents are allergen bound to dextranepichlorhydrin co-polymer resin treated with cyanogen bromide or to microcrystalline cellulose, sheep anti-reagin epsilon fragments, I<125> labelled reagin, with bovine serum to provide component (3); determination of reagin-Ig directed against horse epithelium using allergen bound to cross-linked dextran substituted with isothiocyanotophenoxyhydroxypropyl groups and sheep anti-reagin Fc fragments labelled with I<125>, with horse or sheep gamma globulin as component (3); pregnancy tests using latex particles coated with sheep anti-HCG and bovine or sheep gamma globulin as component (3); determination of IgE using sheep-anti IgE bound to dextran-epichlorhydrin copolymer resin treated with cyanogen bromide, and IgE labelled with I<125>, with sheep gamma globulin as component (3).
