1331892 Dibenzoxazepine-N-carboxylic acid hydrazides G D SEARLE & CO 22 March 1972 [23 March 1971] 13339/72 Heading C2C Novel compounds of general Formula I in which Y are Y<SP>1</SP> are hydrogen, halogen, methyl or trifluoro methyl and R is C 7-12 alkanoyl, halo- C 1-6 alkanoyl, carboxy-C 1-6 alkanoyl, 3 - carboxyacryloyl, phenoxy-C 1-6 alkanoyl, halophenoxy-C 1-6 alkanoyl, C 1-6 alkyl sulphonyl or α-toluenesulphonyl are prepared by (1) reaction of a C 1-6 alkyl ester or acid halide of a Y,Y<SP>1</SP>-substituted 10,11-dihydrodibenz- [b,f][1,4]oxazepine - 10 - carboxylic acid with Q-NH-NH 2 in which Q is C 1-6 alkyl sulphonyl, α-toluene sulphonyl, carboxy acryloyl or C 7-12 alkanoyl or is a C 1-6 alkanoyl group substituted by halo, carboxy, phenoxy or chlorophenoxy or (2) by reaction of Y,Y<SP>1</SP>-10,11-dihydrobenz- [b,f][1,4]oxazepine - 10 - carboxylic acid hydrazide with Q-X in which X is hydroxy, halogen, -OC 1-6 alkyl or O-CO-Z (in which Z is -OC 1-6 alkyl or is connected to Q to form a cyclie anhydride). The reactions take place in the presence of an inorganic base such as sodium or potassium bicarbonate or a tertiary amine. Preferred compounds are 1-R-2-(8-chloro-10,11- dihydrodibenz[b,f][1,4]oxazepine - 10 - carbonyl) hydrazines in which R is octanoyl, heptanoyl, decanoyl, 5-chloropentanoyl or 4-chloro-butanoyl. Compounds of Formula I are antagonists of of prostaglandin, 5-hydroxy tryptamine and acetylcholine and are useful as topical antiinflammatory agents, anti-diarrheal agents and agents for reducing sebum secretion. 1-Octanoyl- 2 - (8 - chloro - 10,11 - dihydrobenz[b,f][1,4]- oxazepine - 10 - carbonyl) hydrazine forms with a carrier an anti-inflammatory composition for administration topically and a composition for inhibiting sebum secretion for administration parenterally.
