Abstract

1331568 Detecting white blood cells MOUNT SINAI RESEARCH FOUNDATION 26 Oct 1971 [30 Oct 1970] 49717/71 Heading B1X A process for detecting a type of white blood cell comprises adding a cytological fixing agent to a solution containing white blood cells to kill the white blood cells and to immobilise catalytic enzymes within the cells without seriously damaging their catalytic properties and without precipitating soluble components present in the solution, adding a cytochemical substrate to the solution containing the killed cells to enzymatically stain the type of white blood cell, and detecting the stained cell. The stain-forming substance added depends upon what type of cell it is desired to detect; the reaction is a consequence of the different enzymes present in different types of cell. Eosinophils and neutrophils may be stained with 4-chloro-1-naphthol and monocytes with a naphthyl ester, indexyl ester or 8-hydroquinoline ester. The process is applicable to blood, spinal fluid 'and bone marrow.
