Abstract

1,269,531. 6,6-Difluoro steroids. SYNTEX CORP. 21 March, 1969, No. 15091/69. Addition to 1,248,938. Heading C2U. The invention comprises compounds of formula wherein R<SP>1</SP> is F or Cl; R<SP>2</SP> is H or a free, esterified or etherified hydroxy group; R<SP>3</SP> is H, OH or Me, with the proviso that R<SP>3</SP> is Me when R<SP>1</SP> is F and R<SP>2</SP> is H or a free or esterified hydroxy group; or R<SP>2</SP> and R<SP>3</SP> together represent the group wherein R<SP>4</SP> and <SP>R6</SP> each represent H, C 1-6 alkyl, C 1-6 haloalkyl, C 3-6 cycloalkyl (C 3-6 cycloalkyl)- (C 1-6 alkyl), " aryl," " aryl "-(C 1-6 alkyl), monocyclic heterocyclyl or (monocyclic heterocyclyl)- (C 1-6 alkyl), or R<SP>4</SP> and R<SP>5</SP> together with the intervening carbon atom represent an optionally esterified or salified (C 2-8 carboxyalkylidene) group, or a monocyclic cycloalkylidene or monocyclic heterocyclic ylidene group; and Z is a single or double carbon-carbon bond. The term " aryl " signifies phenyl optionally substituted with 1-3 F, Cl, OH, C 1-6 alkyl, C 1-6 alkoxy or acylamino groups. Any heterocyclic radicals have 5 or 6 ring atoms including 1 or 2 O, N or S atoms. Preparation of the above is from the corresponding #<SP>4</SP> - 6(α or #) - monofluoro steroids by enol etherification to 3 - alkoxy - #<SP>3,5</SP> - 6 - fluoro steroids which are then treated with perchloryl fluoride to yield the #<SP>4</SP> inventive compounds which may then be dehydrogenated. The compounds of Formula I are said to possess progestational activity, and may be made up with carriers into pharmaceutical compositions for oral and parenteral administration.
