Abstract
1,263,481. Automatic liquid chromatography apparatus. BECKMAN INSTRUMENTS Inc. 26 March, 1969 [2 April, 1968], No. 15815/69. Heading B1X. A high pressure automatic liquid chromatography system suitable for amino acid analysis comprises a sequentially stepped rotatable sample-carrying table 25 and two independent ion-exchange columns 10, 11. As shown, a sample is being drawn into sample valve 23 under the influence of pump 26 and eluent from any of reservoirs 14-17 is passing by pump 12 through a by-pass line in valve 23 into and through long column 10 thereby eluting a previously delivered sample into reactor-detector 22, while at the same time a sample previously drawn up into sample valve 24 is being eluted into short column 11 by means of eluent from any of reservoirs 15-18 passed by pump 13. At the appropriate points in time, table 25 is stepped and valves 19, 20, 21, 23, 24 and 27 switched by means of a programmer to reverse the functions of the two column systems. The sample reservoirs into which the sample is loaded suitably comprises a helically-wound tube mounted around a spool, in which tube the sample is held by means of surface tension (see Fig. 2, not shown). Alternatively, spirally-wound sample reservoirs (see Figs. 6a, b, not shown) may be used. The details of valves 23, 24 are the subject of Specification 1,263,483.
