Abstract

1,253,328. Gamma-globulin purification. UPJOHN CO. 18 March, 1970 [1 April, 1969; 3 March, 1970], No. 13046/70. Heading A5B. γ-globulin is purified from blood serum or plasma by separating γ-globulin from the other proteins in the serum or plasma by treatment with acrinol and removing acrinol from the resulting aqueous solution of the γ-globulin by contacting the solution with silica gel or a mineral silicate adsorbent and separating the adsorbent from the solution. The mineral silicate may be magnesium silicate or trisilicate, native calcium-magnesium silicate, talc heulandite, stilbite, chabazite, analcite or antrolite. An insoluble portion may be separated from the plasma or serum at a pH of from 7 to 8 by adding 3.5 to 5 volumes of 0.4 to w/v acrinol aqueous solution thereto, the solution recovered is mixed at pH 7 to 8 with human erythrocyte stroma or erythrocytes and from the solution recovered y-globulin is precipitated at pH 7 to 8 by adding acetone or a C 1-6 alkanol to provide an acetone or alkanol concentration of 20 to 30% u/v and the precipitate is recovered; the precipitated y-globulin is dissolved by pH 6.5 to 7.5 in phosphate buffer and the resulting solution is treated with silica gel, the solution not adsorbed upon the silica gel is recovered, passed over diethylaminoethyl cellulose equilibrated with phosphate buffer at pH 6.7, the diethylaminoethyl cellulose is eluted with phosphate buffer at pH 6.7 and the γ-globulin is recovered from the eluate in accordance with measurements of adsorbancies at 280 mÁ. The serum or plasma treated may be that of an animal treated with lymphoid tissue of human origin, from a horse treated with human thymocytes, or of human origin.
