1,218,602. Determining antigens. BEHRINGWERKE A.G. 5 Jan., 1968 [5 Jan., 1967], No. 844/68. Heading B1X. Antigens are determined in an airtight container formed by a bottom plate with a raised side wall and a cover, the bottom plate being subdivided into inner and outer compartments by a continuous raised inner wall, by a method comprising filling the outer compartment with a gel vehicle containing an antiserum to the antigen being determined, filling the inner compartment with a wet sponge-like material which will provide a humid atmosphere in the closed container, forming holes in the gel vehicle and putting into them a known volume of the antigen solution to be determined and known volumes of solutions of the antigen of different known concentrations, closing the container allowing the reaction to take place by diffusion of antigen into the gel so forming opaque rings around the holes and comparing the diameters of the opaque rings formed by the known and unknown solutions, the measured diameters being appropriately adjusted if the various solutions have been used. The gel vehicle is preferably agar gel 1% to 3% w/w containing 1% to 20% v/v serum.
