1,208,435. Preserving micro-organisms. NATIONAL RESEARCH DEVELOPMENT CORP. 29 Oct., 1968 [1 Nov., 1967], No. 496 79/67. Heading A2D. [Also in Division F4] A method of preserving a specimen (e.g. micro-organisms) comprises drying the specimen under substantially isothermal conditions by heating the specimen in a receptable, partially evacuating the receptacle and controlling the total pressure in the receptacle in such a manner that the rate of loss of heat from the specimen is substantially equal to the rate at which heat is supplied to the specimen, evacuating the receptacle to a higher vacuum after the specimen has been dried and then hermetically sealing the receptacle. In the apparatus shown, ampoules 1 and 7 each containing a specimen, are mounted on manifold 4 which is connected to a gasballasted rotary vacuum pump 5 and aircooled oil diffusion pump 6, heated in hot water bath 11 or a heated fluidized sand bed and partially evacuated. The difference in the temperature of control specimen in ampoule 7 and the water bath is sensed by a thermocouple 9 and this difference is maintained at a substantially constant predetermined value during drying, by controllably feeding air via adjustable needle valve 15 into the evacuating system. The valve 15 is automatically operated in response to temperature difference sensed by the thermocouple 9. When the specimens have been dried the temperature of the specimens rises towards that of the bath and the valve 15 is closed. The ampoules are then automatically raised from the bath and evacuated to a high vacuum. The evacuated ampoules are healed by a high temperature flame.
