Abstract

1,166,364. N-(tert-aminoalkyl) amide derivatives. AMERICAN CYANAMID CO. 13 Oct., 1967 [25 Nov., 1966 (2)], No. 46843/67. Heading C2C. Novel N-(tert.-aminoalkyl) amide derivatives of the general Formula III wherein R 2 is hydrogen, C 1-6 alkyl or C 2-6 alkenyl; n is 2, 3 or 4; A is a thiophone radical of the formula or a phenyl radical of the formula R and R 1 are hydrogen, C 1-6 alkyl, or halogen; and B is 1-aryl.4-p-perazinyl, 4-arylpiperidino, or 4 - aryl - 3,6 - dihydro - 1(2H) - pyridyl, with the proviso when B is 1 .aryl.4.piperazinyl, A may not be phenyl or substituted phenyl and the pharmaceutically acceptable acid addition salts thereof are prepared by any of the following methods: (1) Reacting a compound A-Y with an amine of the formula B-C n H 2n -(H)R 2 , where Y is a reactive moiety being an acid halide, an ester, an ahydride or an amide. (2) Reacting a compound of formula with a compound H-B, wherein X is halogen, C 1-6 alkylsulphonyloxy, or monocyclic arylsulphonyloxy. Compounds of the above general Formula III in which A is a thiophene radical may also be prepared by the following methods: (3) Reading a compound of the formula ACON(R 2 )H with a compound of the formula X-C n H 2n B ; (4) Reacting an acid of the formula ACOOH with N,N<SP>1</SP>-carbonyldhmidazole and treating the thus obtained ketone with an amine of the formula B-C n H 2n N(R 2 )H ; (5) Reacting an acid of the formula ACOOH with an amine of the formula in the presence of a carbodiimide. N - (2 - Bromoethyl) - and N - (3 - bromopropyl) - 2 - thiopheneearboxamides are obtained by reacting 2-phenoyl chloride with 2-bromoethylamine and 3-bromopropylamine respectively. N - (2 - Bromoethyl) - and N - (2 - chloroethyl) N - methyl - benzamides are prepared by reacting benzoyl chloride with 2-bromoethylamine and 2-chloroethylmethylamine respectively. Pharmaceutical compositions, suitable for oral administration and having analgesic, antiinflammatory, diuretic and anti-depressant properties, contain the above novel compounds or their non-toxic pharmaceutically acceptable acid addition salts as the active ingredient and a pharmaceutically acceptable carrier or diluent.
