Abstract

Tetrahydropyridine derivatives of the formula <FORM:1122942/C2/1> wherein R1 represents a C1- 12 alkyl radical, a C3- 5 alkenyl radical, the cyclopropymethyl radical or a C7- 9 phenylalkyl radical; R2 represents hydrogen or the methyl radical; and R3 represents a C1- 4 alkyl radical, the phenyl radical or a C7- 9 phenylalkyl radical; and their salts with inorganic or organic acids are obtained by partially reducing pyridinium compounds of the formula <FORM:1122942/C2/2> wherein R4 and R5 are each lower alkyl radicals or together represent a lower alkylene radical and X- represents a monovalent anion or the normal equivalent of a polyvalent anion, solvolysing the tetrahydropyridal ketals produced and, if desired, converting the products into salts with inorganic or organic acids. The pyridinium compound used may be obtained by reacting the hydrochloride of a 4-pyridyl ketone with an hydroxy compound, e.g. ethylene glycol, and quaternizing the 4-pyridyl ketal produced with an ester R1X. Therapeutic compositions, having antitussive and analgetic properties, contain as the active ingredient the above tetrahydropyridine derivatives or their pharmaceutically acceptable salts. The analgetic composition may be in a form suitable for oral or parenteral application.
