Abstract

A diagnostic reagent for pregnancy determination similar to that of Specification 1,078,939 contains an anti protein for human chorionic gonadotrophin, (HCG), derived from the blood serum of animals rendered immune to HCG by repeated injections thereof, and extracted by adsorbing the anti-protein on water swollen particles of cross-linked dextran containing random ether linkages, such as Sephadex G; (Registered Trade Mark), and then eluting the anti protein. (see Div. A5-A6). For use the anti-protein is adsorbed on polystyrene latex particles which preferably have an average diameter of from 0.5 to 1.3 microns. The reagent consists of the anti-protein coated polystyrene particles in suspension in an aqueous solution containing 0.45% to 1.5% glycine, 0.2% to 0.45% of the sodium salt of EDTA, 4% to 6% albumin, 0.05% to 0.1% sodium azide, and triethanolamine to adjust the pH to from 8.2 to 8.6.ALSO:A specific anti-protein to human chorionic gonadotrophin (HCG) is isolated from the blood serum of a lower animal containing the anti-protein by adsorbing the serum on water-swollen particles of cross-linked dextran containing random ether linkages to glucose residues in the polysaccharide chains (e.g. Sephadex G; Registered Trade Mark), eluting the anti-protein, and recovering it from the eluate. Elution is suitably effected with an aqueous solution of sodium chloride and tris (hydroxymethyl)-aminomethane of pH 8.0, the same reagent being used to swell the dextran gel. As a serum source, animals such as rabbits may be immunised with a number of injections of a commercial human HCG preparation, the animals being bled when a suitable anti-serum titre has been reached. Recovery of the anti-protein from the eluate is by lyophilisation, re-solution and dialysis. The HCG anti-protein is of use in preparing a pregnancy diagnostic reagent, by adsorption on to polystyrene latex particles (see Div. B1-B2)
