Abstract
Compounds of the general Formula II <FORM:1025564/C2/1> wherein R1 is methoxy or benzyloxy, R2 is a C1 to C5 alkyl, and R3 is hydrogen, carbethoxy, or an acyl group derived from a C1 to C5 aliphatic or aromatic carboxylic acid which is optionally substituted by hydroxy, C1 to C5 alkoxy, aralkoxy, amino or C1 to C5 alkyl, and the hydrochlorides thereof are prepared by reduction with an alkali metal aluminium hydride of a compound of the general Formula <FORM:1025564/C2/2> or a C1 to C5 ester thereof, to the correspondin 2-hydroxymethyl benzofuran, converting the latter into the corresponding 2-chloromethyl benzofuran by reaction with a chlorinating agent, reacting said 2-chloromethyl compound with a metal cyanide to form the corresponding 2-cyanomethyl benzofuran, reducing the 2-cyanomethyl compound with an alkali metal aluminium hydride to produce the corresponding 2 - (b - aminoethyl) - benzofuran, which is optionally isolated as a hydrochloride or the corresponding 2 - (b - acylaminoethyl) - benzofuran of Formula II is prepared by reaction with an aliphatic or aromatic carboxylic acid or the corresponding compound of Formula II, wherein R3 is carbethoxy is prepared by reaction with ethyl chlozoformaIe. The preferred chlorinating agent employed for the formation of 2-chloromethyl benzofuran is thionyl chloride in the presence of pyridine.
