The neck is threaded (8), the filter capsule (2) is screwed tightly on the neck, by screwing the neck supports the grooved joint (9). The pressure accumulator (4) is joined to the pressure regulator assembly (3) employing a pressurizing cock (5) and a depressurizing valve (6). The filter capsule (2) comprises a pressure-resistant filter support (10), a filtration membrane (11), a splined joint on its outermost ring (9), a screw thread (12), a plug having a thread (13) adapted to the thread (12), a plug (14) adapted to the emptying zone (15) and a support (16) adapted to the liquid trash used during the filtrations. The pressure regulator (17) receives the pressure accumulator (18) through a thread, a low flow needle (1 9) pierces the pressure accumulator during screwing. The valve (5) allows the gas to escape to the pressure reducer (20) and to pressurize the tank through the low pressure port (21). An example of use of the new device is given below. diagnosis of Legionella pneumophilla by gene enzymatic amplification technique. Diagnostic techniques using the methods of amplification of the genetic material of certain microorganisms have appeared in many laboratories. These methods are sensitive, specific and allow to obtain reliable results in a few hours. In addition, these amplification techniques are now used in many analysis laboratories, they have the advantage of detecting bacteria that are possibly not cultivable because of their physiology or their state.

Cette méthode a par ailleurs été utilisée avec succès dans le diagnostic de Legionelle (Wellinghausen et al. PCR. Appl Environ Microbiol. Sep;67(9):3985-93. (2001)). Real-time quantitative PCR is a typical example of these methods. This technique is used in many fields, it has the advantage of being very sensitive, specific and to allow a thorough automation of laboratory procedures and results reporting. by avoiding the possibility of contaminations, this technique has been described in particular in patents WO9202638 and US5723591. This method has also been used successfully in the diagnosis of Legionella (Wellinghausen et al., PCR Appl Environ Microbiol., Sep; 67 (9): 3985-93 (2001)).
Chaque récipient supérieur est rempli par un litre d'eau chaude sanitaire à 60 C chargé par (10, 100, 1000, 10000, 100000, 1000000) unités de Légionelles.. Chaque cartouche compacte de filtration équipée d'un filtre de 47mm de diamètre ayant des pores de 0,44tm est vissée sur le goulot du récipient supérieur, l'ensemble est retourné et placé sur une poubelle à liquide. Une cartouche de 1 6g de C02 est vissée sur son support et le robinet est ouvert pour alimenter le détendeur qui délivre une pression de 13 104 Pa. La filtration totale des prélèvements demande entre 3 et 7 minutes suivant les prélèvements. Le robinet est alors fermé et le récipient dépressuriser en utilisant la valve. La cartouche compacte de filtration est hermétiquement fermée puis envoyée au laboratoire dans une enveloppe. La membrane est libérer par simple extraction du joint cannelé au laboratoire. La membrane est traitée comme décrit dans la référence (1) sauf que les amorces (a) 5'tggcgctcaattggcttt-3' ; Seq ID No. 1; et (b) 5'-atctttctgaaacttgttaagaacgtctt-3' ; Seq ID No. 2; utilisée sont décrites dans la présente invention et que la sonde marquée correspond à (c)5'-accgaacagcaaatg-3'; Seq ID No. 3 de la présente invention. Les échantillons sont soumis à 45 cycles d'amplification. The membrane is treated as described in reference (1) except that the primers (a) 5'tggcgctcaattggcttt-3 '; Seq ID No. 1; and (b) 5'-atctttctgaaacttgttaagaacgtctt-3 '; Seq ID No. 2; used are described in the present invention and that the labeled probe corresponds to (c) 5'-accgaacagcaaatg-3 '; Seq ID No. 3 of the present invention. The samples are subjected to 45 amplification cycles.
