SELECTION OF TRANSGENIC EMBRYOS.
La présente Invention est relative à un procédé de sélection d'embryons transgéniques. The present invention relates to a process for selecting transgenic embryos.
La transgénèse est largement utilisée chez la souris, mais encore peu développée chez d'autres animaux, tels que les animaux de ferme. Cette situation s'explique par le coût important de cette technologie liée principalement au faible taux d'intégration d'un transgène dans le noyau transcriptionnellement actif de l'oeuf : on estime que chez la plupart des espèces domestiques, moins de 1% des embryons injectés qui se développent à terme après transfert dans une femelle receveuse sont transgéniques [MURRAY et al. (1989)], alors que ce taux est d'environ 10% chez la souris. Cette situation implique chez ces espèces de recourir à un nombre d'autant plus élevé d'embryons et de femelles receveuses. De ce fait, le coût du premier veau transgénique produit a été estimé à environ 3 millions de francs [SEIDEL, (1993)]. (1989)], whereas this rate is around 10% in mice. This situation implies in these species to resort to an even higher number of embryos and recipient females. As a result, the cost of the first transgenic calf produced was estimated at around 3 million francs [SEIDEL, (1993)].
Pour diminuer ce coût, il a été proposé d'augmenter le taux d'intégration de l'ADN exogène en recherchant de nouveaux vecteurs s'intégrant plus facilement dans le génome de l'embryon. En effet, chez plusieurs espèces, ce n'est pas tant le coût de production de ces embryons qui constitue l'obstacle économique principal, que celui lié à l'accès à un grand nombre de femelles receveuses une fois l'injection réalisée : il faut par exemple chez les bovins, l'équivalent d'environ 150 receveuses pour produire un animal transgénique fondateur [HILL et al., (1992)]. Dans ces conditions, le coût technique de production de blastocystes microinjectés ne représente que seulement 10% du coût d'entretien du troupeau de receveuses.
