NOUVEAUX COMPLEMENTS NUTRITIONNELS OU DIETETIQUES
ANTI-VIEILLISSEMENT A BASE D'ACIDE GRAS POLY-INTURE
NEW NUTRITIONAL OR DIETETIC SUPPLEMENTS
ANTI-AGING BASED ON POLY-INTURE FATTY ACID
The present invention relates to a nutritional supplement based on a polyunsaturated fatty acid and to its use for correcting or repairing dietary errors.
It relates more particularly to preparations based on a polyunsaturated fatty acid to supplement or improve the nutritional needs related to age.
It specifically relates to food or dietetic preparations intended to compensate for the effects of aging, which contain in determined quantities, a polyunsaturated fatty acid, stearidonic acid or octadecatetraenoic acid (C18: 4 n-3) in combination or in combination mixture with diluents, inert materials such as flours, carbohydrates, starches or modified celluloses, in particular chemically.
Stearidonic acid is a polyunsaturated fatty acid whose presence in nature's products is rare and even frequently, undetectable. The main sources are borage seeds, evening primrose, blackcurrant seeds, raspberry seeds and red currant seeds. Another source of stearidonic acid is found in fish oils such as tuna oil, sardine oil or halibut oil and other similar fatty fish.
It is possible to use the oil as it is, but it is generally preferred to use concentrates obtained by vacuum distillation or by fractionation of these oils. It is also possible to obtain concentrates containing mainly or essentially stearidonic acid.
It is also possible to carry out a fractionation or a concentration of the lipids into stearidonic acid by passing over a silver or mercurial resin or over a column of silica gel loaded with silver nitrate and selective elution.
(cf. BREVET
EUROPEEN 092085).However, the most abundant and accessible source is black currant seed oil, raspberry seed oil, gooseberry seed oil or gooseberry seed oil with an acid content. stearidonic ranges from 2.5 to 5%. (cf. PATENT
EUROPEAN 092085).
It is also possible, starting from blackcurrant seed oil, to carry out a fractionation by high performance liquid chromatography in order to obtain a cut in which the stearidonic acid content is found to be appreciably higher compared to what it could be in l 'original oil.
The great interest of stearidonic acid for food, results from its place in the cascade of transformations, undergone by polyunsaturated fatty acids towards the different types of prostaglandins. Prostaglandins of series 1 (PG1) come from dihomo r-linolenic acid, prostaglandins of series 2 (PG2) come from arachidonic acid and prostaglandins from series 3 (PG3) come from eicosapentaenoic acid .
Stearidonic acid also plays an important role in the constitution of cell membranes leading to the formation of phospholipids rich in polyunsaturated acids.
The formation of prostaglandins is a very important element of physiology because they are essential mediators for muscle contraction, brain function and vascularization.
However, this biological whole tends to deteriorate due to aging because in the elderly, the enzymatic activities called desaturasic activities are decreasing. This drop in desaturasic activities leads from linoleic acids (series n-6) and a-linolenic acids (series n-3) to a decrease in the formation of polyunsaturated fatty acids which constitute the essential links of nutrition, respectively r-linolenic acid (C18: 3 n-6) and stearidonic acid (C18: 4 n-3), which give rise, after desaturations and successive elongations, to the synthesis of membrane polyunsaturated fatty acids and prostaglandins by cyclization.
The regulation of this complex biological system takes place through a key enzyme, 86-desaturase. It is the limiting enzyme of the system. It is an enzyme that is highly regulated in particular by insulin and by certain proteins which activate it, adrenaline or glucagon which inhibit it. Likewise, certain fatty acids such as α-linolenic acid inhibit it. We have therefore been able to demonstrate the existence of a transformation coefficient in animals by in vivo experiments. In humans, with age, there is a decrease in the activity of A6-desaturase and the occurrence, in comatic subjects or in patients with severe hepatic impairment, of an abnormal metabolic pathway, A5-desaturase.
It therefore appears, two ways of deriving the metabolism of essential polyunsaturated fatty acids - a toxic way (ss-oxidation) - a way of energy use which constitutes, in a way,
a waste.
The conversion of essential fatty acids into polyunsaturated fatty acids under the influence of A6-desaturase therefore constitutes a bottleneck.
Faced with this drop in desaturasic activity, essential for the formation of physiological mediators, one could conceive the contribution of slightly unsaturated fatty acids to reactivate the desaturasic activity by artificially increasing the concentration of substrate. This method has proven to be ineffective because high concentrations of fatty acids inhibit or reduce desaturasic activity as a result of regulation.
On the contrary, and this is what the invention is based on, the contribution of more unsaturated fatty acids and in particular that of stearidonic acid makes it possible to compensate for the decrease in A6-desaturasic activity and thus, to continue biosynthesis long-chain polyunsaturated fatty acids and in particular the synthesis of eicosapentaenoic acid and finally docosahexaenoic acid or cervonic acid.
Stearidonic acid is the physiological precursor of prostaglandins PG3. We thus re-establish the biosynthesis of essential mediators.
The drop in desaturasic activity leads to a modification of the fatty acid composition of the membranes and in particular of the nervous membranes. Brain lipids and in particular brain phospholipids contain a very high content of long-chain polyunsaturated fatty acids which are formed from essential acids, linoleic acid and o-linolenic acid. They therefore play an important role in the maintenance of nerve activity and for the conservation of normal brain activity.
It has thus been found in rats that a defect in α-linolenic acid in the diet very significantly modifies the fatty acid composition in various organs and in particular the brain. There is, in fact, a modification of the enzymatic activity of the cerebral membrane, a reduction in the amplitude of the electrophysiological parameters such as for example the electro-retinogram, a modification of the resistance to toxic substances of the central nervous system and above all a reduction aptitude for learning.
Given this considerable decrease in nerve and brain functions, it appeared that the introduction of stearidonic acid into the diet was capable of compensating for this drop in enzyme activity and restoring the possibility for the body to '' synthesizing polyunsaturated fatty acids such as docosahexaenoic acid or cervonic acid (C22: n-3) and / or eicosapentaenoic acid (C20: 5 n-3) tl has therefore been written that the decrease Q6-desaturasic activity was a key factor in the phenomena of aging. The addition of stearidonic acid in food or dietetic preparations according to the invention makes it possible to reduce or delay the influence of this drop in enzymatic activity.
The nutritional or dietetic preparations according to the invention consist of stearidonic acid, mixed or supplemented with diluent or a vehicle allowing use by the digestive route.
For this use, use will be made of a formulation in the form of soft capsules, dragees or tablets, tablets, sachets of flavored or sweetened powder, cookies, pancakes or confectionery, possibly combined with other active ingredients. (vitamins, citroflavonoids, free radical scavengers, trace elements, flavonoids derivatives).
Stearidonic acid can also be incorporated into a fluid vehicle such as a jelly, a vegetable mucilage or a suspension or else in the form of microgranules or else microencapsulated in gelatin to form a powder.
For use in the elderly, presentation in the form of soft capsules or in the form of sachets of powder is most convenient.
In general, the nutritional or dietetic preparations according to the invention contain from 0.100 to 0.500 g of pure stearidonic acid or in the form of a concentrate titrated in stearidonic acid, and preferably from 0.100 to 0.250 g.
These preparations can be administered several times a day at a dose of 0.100 to 0.500 g per unit.
The daily administration administration will be a function of the other lipid contributions of the food and in particular of the food of the old subject. If it is poor in source of lipids capable of providing polyunsaturated fatty acids such as peanut oil or hydrogenated fats, it will be necessary to provide larger amounts of stearidonic acid in supplement form nutritional. On the contrary, if the diet already provides fats likely to contain small amounts of poly-unsaturated fatty acids such as fish oils, the food requirement for stearidonic acid will be more reduced and the compositions according to the invention will therefore constitute a dietary supplement.
The exemplary embodiments of nutritional or dietetic compositions illustrate the invention without, however, limiting it.
PHARMACOLOGICAL STUDY OF STEARIDONIC ACID ON THE ACTIVITY OF
DESATURASE IN RATS AND MICE
In the present study, 86-desaturasic activity was measured in mice and found linear up to 4 mg of protein per ml of incubation medium at all ages with brain homogenates. With liver homogenates, the activity was linear up to 2.5 mg protein / ml for the first 5 days after birth. Then it was linear up to 1.5 mgykl.
Figure 1 shows that the desaturasic activity of the liver increases approximately 7 times from the 18th day of gestation to the 11th day until weaning in mice and remains constant for 9 months.
In a very interesting way, t6-desaturase decreases by 4 times during aging. Basically, this scheme is similar to that found for the desaturation of linoleic acid, but the desaturasic activity is 2 times greater with alpha-linolenic acid than with linoleic acid. The modification of the desaturasic activity is more marked with α-linolenic acid (4 times) than with linoleic acid.
In rat liver microsomes, desaturasic activity decreases up to 9 months, although the animal develops continuously throughout its existence.
In the mouse brain, desaturasic activity decreases dramatically during pre- or post-natal development (4 times) until weaning. The clearest activity was found during the perinatal period. Early brain development requires large amounts of polyunsaturated fatty acids for the synthesis of membranes.
Desaturasic activity # 6 in rats decreases by 2 between weaning and the age of 9 months and then presents a low and stable level. This raises the question of whether this low level of activity is sufficient to provide the polyunsaturated fatty acids necessary for the physiological metabolism of all brain membranes.
As desaturasic activity also decreases during aging, in the liver, if the concentration in the brain is too low, the liver cannot compensate for this deficiency. This is why, during the aging period, it seems particularly advantageous to supply a quantity of stearidonic acid in the diet so as to bypass the decrease in desaturasic activity and again to provide a substrate for the elongation of polyunsaturated fatty acid chains.
Figure I shows the evolution of A6-desaturasic activity during development and during aging in the brain of mice and in liver homogenates.
Les cercles i indiquent la synthèse complète d'acide gras poly-insaturés à partir d'acide o-linolénique (principalement l'acide stéaridonique (C18:4 n-3) avec de petites quantités d'acide eicosapentaénoique (C20:5 n-3) et d'acide docosahexaénoique (C22:6 n-3).Circles i indicate the complete synthesis of polyunsaturated fatty acid from o-linolenic acid (mainly stearidonic acid (C18: 4 n-3) with small amounts of eicosapentaenoic acid (C20: 5 n- 3) and docosahexaenoic acid (C22: 6 n-3).
