Claims (21)
1. Menetelmä kemikaalitestausta varten, joka käsittää a) solujen viljelyn ensimmäisessä kasviperäisessä nanofibrilliselluloosa (NFC) hydrogeelissä tai sen päällä in vivo tapaisten solujen saamiseksi; b) kasviperäisen nanofibrilliselluloosa hydrogeelin poistamisen; c) in vivo tapaisten solujen altistamisen testi kemikaalille toisessa kasviperäisessä nanofibrilliselluloosa hydrogeelissä; d) altistetuiden in vivo tapaisten solujen inkuboinnin; e) testi kemikaalin vaikutuksen havaitsemisen in vivo tapaisiin soluihin ainakin yhdellä mittauksella inkuboinnin aikana tai sen jälkeen.
2. Patenttivaatimuksen 1 mukainen menetelmä joka edelleen käsittää kasviperäisen nanofibrilliselluloosa hydrogeelin poistamisen enemmän kuin yhden kerran a) ja e) vaiheen välillä.
3. Jonkin patenttivaatimuksen 1 tai 2 mukainen menetelmä joka edelleen käsittää in vivo tapaisten solujen altistamisen testi kemikaalille vaiheen a) hydrogeelissä.
4. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainitulla toisella nanofibrilliselluloosa hydrogeelillä on samat tai eri ominaisuudet kuin vaiheen a) nanofibrilliselluloosa hydrogeelillä.
5. Patenttivaatimuksen 4 mukainen menetelmä, missä mainitut eri ominaisuudet koskevat yhtä tai useampaa ominaisuuksista jotka on valittu nanofibrilliselluloosa hydrogeelin jäykkyydestä, nanofibrilliselluloosa pitoisuudesta, nanofibrilliselluloosa hydrogeelin nolla leikkaus viskositeetistä, nanofibrilliselluloosa varauksesta, ja nanofibrilliselluloosa hydrogeelin läpinäkyvyydestä.
6. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainittu kemikaalitestaus käsittää myrkyllisyys testauksen; turvallisuus testauksen; lääke ehdokkaiden testauksen; lääke seulonnan; tai aihiolääke kandidaatti testauksen.
7. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainittu testikemikaali on valittu joukosta joka koostuu lääkkeistä; lääke kandidaateista; aihiolääkkeistä; aihiolääke kandidaateista; nanopartikkeleista; solua säätelevistä aineista, ruoasta ja ruoan lisäaineista; kotitalous tuotteista; teollisista kemikaaleista; pakkausmateriaaleista; ilmanraikastimista; kasvin kasvua säätelevistä aineista; ympäristö myrkyistä; tuholaismyrkyistä; henkilökohtaisen hygienian tuotteista; tai niiden kemiallisista ainesosista.
8. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainitut in vivo tapaiset solut ovat kantasoluja, primaarisoluja tai sekundaarisoluja tai mikä tahansa näiden kombinaatio.
9. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä nanofibrilliselluloosa hydrogeeli järjestetään tuen päälle ennen kuin solut viljelyä varten istutetaan hydrogccliin tai sen päälle; tai sen jälkeen kun solut viljelyä varten on ensin istutettu hydrogeeliin.
10. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä kasviperäisen nanofibrilliselluloosan poistaminen käsittää fyysisen, mekaanisen tai kemiallisen poistamisen tai minkä tahansa näiden kombinaation, edullisesti kasviperäinen nanofibrilliselluloosa käsitellään entsymaattisesti sellulaasilla riittävän pitkän ajan jotta solut ainakin osittain vapautuvat.
11. Patenttivaatimuksen 10 mukainen menetelmä, missä sellulaasi on sellulolyyttinen entsyymiseos, valinnaisesti käsittäen hemisellulaasia; kaupallisen sellulaasin; osittain puhdistetun sellulaasin; tai puhdistetun sellulaasin.
12. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä testi kemikaalin vaikutuksen havaitsemisen in vivo tapaisiin soluihin käsittää kvalitatiivisen havaitsemisen, kvantitatiivisen havaitsemisen tai minkä tahansa näiden kombinaation.
13. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä testi kemikaalin vaikutuksen havaitsemisen in vivo tapaisiin soluihin käsittää kromatografisen havannointijärjestelmän, optisen havannointijärjesteinään tai minkä tahansa näiden kombinaation.
14. Patenttivaatimuksen 13 mukainen menetelmä, missä kromatografinen havannointijärjestelmä perustuu kromatografiaan kuten kaasukromatografia, HPLC, affiniteetti-, syrjäytys-, joninvaihto-, koko ekskluusio-, geelisuodatus, kiinteä proteiini neste-, paperi- tai ohutkerroskromatografiaan; tai elektrokromatografiaan kuten geelielektroforeesi, 2D geelielektroforeesi, tai isoelektrinen fokusoiva.
15. Patenttivaatimuksen 13 mukainen menetelmä, missä optinen havannointijärjestelmä perustuu yhteen tai useampaan visuaaliseen tutkimukseen; spektroskopiaan kuten ydinmagneettinen resonanssi (NMR), Raman, IR, UV, näkyvä valo, fluoresenssi, massa spektroskopia (MS); mikroskopiaan kuten optiseen mikroskopiaan (faasikontrasti, käänteisfaasikontrasti, samapolttopisteinen, fluoresenssi) tai electronimikroskopiaan (TEM, SEM) tai atomivoima mikroskopiaan (AFM); fotometriaan; laser- tai virtaussytometriaan, valinnaisesti käyttäen korkea sisältö seulontaa (HCS) ja/tai isotooppi merkintää.
16. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainittu nanofibrilliselluloosa hydrogeeli sisältää 0.05-10 p-%, kuten 0.1-4 p-%, tai 0.12-1.2 p-% nanofibrilliselluloosaa.
17. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainittu nanofibrilliselluloosa hydrogeeli käsittää natiivi nanofibrilliselluloosaa tai anionista nanofibrilliselluloosaa, kuten oksidoitu nanofibrilliselluloosa.
18. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainittu nanofibrilliselluloosa hydrogeeli käsittää nanofibrilliselluloosaa pääasiassa selluloosa Lsestä.
19. Jonkin edellä mainitun patenttivaatimuksen mukainen menetelmä, missä mainitun nanofibrilliselluloosa hydrogeelin jäykkyys on ainakin 4 Pa, edullisesti 4-50000 Pa.
20. Kasviperäisen nanofibrilliselluloossa hydrogeelin käyttö menetelmässä kemikaalitestausta varten, missä nanofibrilliselluloosa hydrogeeli käytetään solujen viljelemiseen hydrogeelissä tai sen päällä in vivo tapaisten solujen saamiseksi; mainittu kasviperäinen nanofibrilliselluloosa poistetaan; mainitut in vivo tapaiset solut altistetaan testi kemikaalille toisessa kasviperäisessä nanofibrilliselluloosa hydrogeelissä; altistetut in vivo tapaiset solut inkuboidaan; ja testi kemikaalin vaikutus in vivo tapaisiin soluihin havaitaan ainakin yhdellä mittauksella inkuboinnin aikana tai sen jälkeen.
21. In vivo tapaisten solujen käyttö kemikaalitestausta varten jotka solut saadaan viljelemällä soluja kasviperäisessä nanofibrilliselluloosa hydrogeelissä tai sen päällä, joka käyttö käsittää kasviperäisen nanofibrilliselluloosa hydrogeelin positamisen ennen testi kemikaalin vaikutuksen in vivo tapaisiin soluihin havaitaan toisessa kasviperäisessä nanofibrilliselluloosa hydrogeelissä.
