Abstract

Determn. of a specific type of group of microorganisms is effected by (a) introducing marker DNA (I) into a host microorganism, where (I) codes for a detectable expression prod. (II) is derived from a eukaryotic or prokaryotic donor and is normally absent in or poorly expressed by the host, and (b) detecting expression of (II) as evidence of the presence of the host microorganism. Determn. of antibiotics is effected by incubating an antibiotics-sensitive microorganism contg. (I) into a medium contg. the test antibiotic(s), detecting expression of (II), and comparing the result with known responses of the microorganism to antibiotics. (I) codes for luciferase and is derived from Vibrio fischeri. It is introduced into the host microorganism by (a) transduction using a bacteriophage vector, esp. L28 (ATCC 40183) or lambda phase L4; (b) transformation using a plasmid vector, esp. pBTK5 or pAChv-1; or (c) conjugation using a bacterial strain contg. (I) in a transferrable chromosome or plasmid.
