Abstract
Método para crear, de manera no específica, múltiples copias de RNA partiendo de ácido nucleico que contiene material inicial que comprende un conjunto de mRNAs, comprendiendo cada mRNA un terminal de poli-A, en donde el material se pone simultáneamente en contacto con: S un oligonucleótido que comprende una secuencia oligo- dT en su extremo 3'', la secuencia de un promotor reconocido por una RNA polimerasa en su extremo 5'' y una región de iniciación de transcripción que se localiza entre la secuencia oligo-dT y la secuencia del promotor, en donde el oligonucleótido se bloquea en su extremo 3'', de tal manera que se prohíbe la extensión a partir de ahí, S un enzima que tiene actividad de transcriptasa inversa, S un enzima que tiene actividad de RNasa H, y S un enzima que tiene actividad de RNA polimerasa, S suficiente cantidades de dNTPs y rNTPs, y la mezcla de reacción resultante se mantiene en las condiciones apropiadas durante un periodo de tiempo suficiente para que tengan lugar los procesos enzimáticos.
