Abstract

Procedimiento de cuantificación del gluten en alimentos. Procedimiento que se realiza por espectrometría de masas MALDI-TOF: 160 {u l de extracto etanólico de muestra junto con 32 {u l de octil-{b -D-glucopiranósido 50 mM se mezclan con 100 {u l de disolución saturada de ácido sinapínico en acetonitrilo acuoso al 30% con 0,1% de ácido trifluoroacético, se seca a una alicota de 25 {u l en Speed-Vac hasta reducir su volumen a 3-4 {u l (aprox. 10 min) un de esta mezcla se deposita sobre el pocillo de un portamuestras de acero inoxidable y se deja secar a temperatura ambiente durante 5 min.Quantification procedure for gluten in food. Procedure performed by MALDI-TOF mass spectrometry: 160 {ul of sample ethanolic extract together with 32 {ul of 50 mM octyl- {b -D-glucopyranoside are mixed with 100 {ul of saturated solution of synapinic acid in acetonitrile 30% aqueous with 0.1% trifluoroacetic acid, dry to a 25 {ul aliquot in Speed-Vac until reducing its volume to 3-4 {ul (approx. 10 min) one of this mixture is deposited on the well of a stainless steel specimen holder and allowed to dry at room temperature for 5 min.
