Abstract
Procedimiento de unión covalente de ligandos para la preparación de reactivos inmunológicos para utilización en inmunoanálisis homogéneos que emplean micropartículas (MPs) y su aplicación a la medida de ferritina y del receptor soluble de la transferrina, en suero o plasma sanguíneos. Procedimiento consistente en un reactivo para la medida de sustancias en muestras biológicas que incluye el uso de micropartículas (MPs) en suspensión acuosa recubiertas de anticuerpo (Ac) mediante un puente ligando-contraligando. En este reactivo las MPs tienen fijado en su superficie un contraligando como avidina o estreptavidina y los anticuerpos específicos están marcados con un ligando como biotina. Además, la presente invención describe un procedimiento específico para la unión covalente de ligandos sobre la fracción Fc de los anticuerpos que permite una disposición espacial de los mismos sobre las MPs muy favorable a la inmunoagregación. Como resultado de esta disposición espacial aumenta el número de anticuerpos activos y por lo tanto se reduce la cantidad de anticuerpo necesario para la preparación de los reactivos. También, y debido a esa disposición espacial favorable, se eliminan las reacciones inespecíficas debidas al factor reumatoide y se mejora la estabilidad de los reactivos. El invento se aplica a la evaluación del estado de las reservas de hierro mediante la determinación turbidimétrica/nefelométrica de la ferritina y de los receptores de transferrina en suero o plasma sanguíneos.Covalent ligand binding procedure for the preparation of immunological reagents for use in homogeneous immunoassays using microparticles (MPs) and their application to the measure of ferritin and the soluble transferrin receptor, in blood serum or plasma. Procedure consisting of a reagent for the measurement of substances in biological samples that includes the use of microparticles (MPs) in aqueous suspension coated with antibody (Ac) by means of a ligand-counter-ligand bridge. In this reagent, the MPs have a counterlink such as avidin or streptavidin fixed on their surface and the specific antibodies are labeled with a ligand such as biotin. Furthermore, the present invention describes a specific procedure for the covalent binding of ligands on the Fc fraction of the antibodies that allows a spatial arrangement of the same on the MPs very favorable to immunoaggregation. As a result of this spatial arrangement the number of active antibodies increases and therefore the amount of antibody required for the preparation of the reagents is reduced. Also, and due to this favorable spatial arrangement, non-specific reactions due to rheumatoid factor are eliminated and the stability of the reagents is improved. The invention is applied to the evaluation of the state of iron reserves by means of the turbidimetric / nephelometric determination of ferritin and of the transferrin receptors in blood serum or plasma.
