Abstract

Carbamate kinase: its preparation and use. The carbamate kinase of Enterococcus (formerly Streptococcus) faecalis which catalyses, in the presence of mg2+, the formation of carbamate and adenosine triphosphate (ATP) from carbamyl phosphate and adenosine diphosphate (ADP) and the reverse reaction from carbamate and ATP to carbamyl phosphate and ADP, is prepared by genetic engineering techniques. A gene coding for the carbamate kinase of the usual source of the enzyme (Enterococcus faecalis) is isolated, sequenced and used to transform a micro-organism and generate a transformant which can be cultured to produce carbamate kinase in high yield. A purification process which can be used to produce essentially pure enzyme is used.
