Abstract

Method for the enzyme synthesis of deoxyoligonucleotides on a solid substrate, which comprises performing the synthesis on a solid substrate in the (5') to (3') direction using terminal transferase, a DNA polymerase regardless of type which preferentially catalyses the incorporation of deoxyribonucleoside (5') triphosphates to single chain DNA terminal (3') hydroxyl and uses deoxyribonucleotides with their (3') hydroxyls temporarily protected in order to add a single nucleotide in each cycle.
