Abstract

Molecular diagnostic methods for the detection of Brucella abortus and differentiation from vaccine strain B19. A series of molecular diagnostic methods which can be used to differentiate between the Brucella abortus B19 vaccine lineage and all other lineages of the genus Brucella are described. In one case, amplification of the genome is applied using oligonucleotides derived from the region containing the genes coding for the erythritol utilization route as indicators. Another group of methods is based on the hybridization of genome DNA using test oligonucleotides from the eri region mentioned. In this case the hybridization technique known as the Southern type or alternatively the "dot blot" procedure may be used. Finally these methods are applied to the identification of micro-organisms of the genus Brucella in any biological sample. The new procedures have advantages over conventional immunological and culture methods providing greater specificity and speed.
