EL METODO TIENE POR OBJETO LA DETECCION DEL VIRUS DE LA PESTE PORCINA AFRICANA A TRAVES DEL EMPLEO DE UN ENSAYO INMUNOENZIMATICO, EN EL QUE SE UTILIZA COMO ANTIGENO LA PROTEINA MAYORITARIA DEL VIRUS (P72), OBTENIDA POR EXPRESION DEL GEN QUE LA CODIFICA. EL GEN SE IDENTIFICO POR HIBRIDACION CON SONDAS DE OLIGONUCLEOTIDOS DEDUCIDAS A PARTIR DE INFORMACION ESTRUCTURAL DE LA PROTEINA PURIFICADA. UNA VEZ DETERMINADA SU SECUENCIA DE NUCLEOTIDOS, EL GEN SE INSERTO EN EL VECTOR DE EXPRESION PAR3038, OBTENIENDOSE EL PLASMIDO RECOMBINANTE PS72A. ESTE PLASMIDO SE UTILIZO PARA TRANSFORMAR LA CEPA DE E.COLI BL21 (DE3), QUE TRAS LA INDUCCION CON IPTC, PERMITIO LA PRODUCCION A GRAN ESCALA DE LA PROTEINA P72 RECOMBINANTE. LA PROTEINA SE PURIFICO POR CROMATOGRAFIA LIQUIDA DE ALTA RESOLUCION Y SE UTILIZO DIRECTAMENTE EN ENSAYOS ANTIGENO-ANTICUERPO CON MUESTRAS DE SUERO DE ANIMALES CON OBJETO DE DIAGNOSTICAR LA ENFERMEDAD.
