Abstract
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von cis-Cedrendiol umfassend das Bereitstellen von alpha-Cedren in einem Lösungsmittel umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel, das Zugeben von Kaliumpermanganat und einer Base und, optional, das Entfernen des weiteren protischen Lösungsmittels aus der Reaktionsmischung.
Description
Verfahren zur Herstellung von Cedrendiol
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von c/s-Cedrendiol umfassend das Bereitstellen von alpha-Cedren in einem Lösungsmittel umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel, das Zugeben von Kaliumpermanganat und einer Base und, optional, das Entfernen des weiteren protischen Lösungsmittels aus der Reaktionsmischung.
Cedrendiole sind wichtige Ausgangsstoffe für die Herstellung hochwertiger Duftstoffe. Aufgrund seiner herausragenden Geruchseigenschaften als Acetonid ist das c/s-Cedrendiol von besonderem Interesse. Die Herstellung von c/s-Cedrendiol erfolgt in der Regel aus Cedren in einem zweistufigen Verfahren. Zunächst wird Cedren zum Cedrenepoxid oxidiert und dieses Epoxid anschließend zum Diol geöffnet. Bei der Öffnung des Epoxides bilden sich allerdings ebenfalls Nebenprodukte (Cedralon, frans-Cedrendiol).
Semmler und Hoffmann berichten von der Oxidation von Cedren mit Kaliumpermanganat zum Cedrendiol (F.W. Semmler, A. Hoffmann, Chem. Ber., 1907, 40, 3521). Dabei wird Cedren in wässrigem Aceton gelöst und allmählich fein gepulvertes Kaliumpermanganat eingetragen. Die Aufreinigung erfolgt durch Extraktion mit Ether und anschließendem Abdestillieren des Ethers. Die auf das eingesetzte Cedren bezogene Ausbeute beträgt allerdings lediglich 12-15 % und das beschriebene Verfahren ist daher nicht für eine wirtschaftliche Synthese geeignet.
Es war Aufgabe der vorliegenden Erfindung ein Verfahren zur direkten Darstellung von cis- Cedrendiol aus Cedren ohne eine Zwischenstufe bereitzustellen, bei welchem Ausbeuten erreicht werden, die eine wirtschaftliche Anwendung erlauben.
Die Aufgabe wird gelöst durch ein Verfahren zur Herstellung von c/s-Cedrendiol umfassend die Schritte: i) Bereitstellen von alpha-Cedren in einem Lösungsmittel umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel, bevorzugt ausgewählt aus der Gruppe bestehend aus sekundären und tertiären Alkoholen, besonders bevorzugt ausgewählt aus der Gruppe bestehend aus fe/f-Butanol und Isopropanol, ii) Zugeben von Kaliumpermanganat und einer Base zu der Lösung aus Schritt i), und, optional iii) Entfernen des weiteren protischen Lösungsmittels aus der Reaktionsmischung aus Schritt ii). Es wurde überraschend gefunden, dass der Einsatz einer Mischung umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel und die Zugabe von Kaliumpermanganat zusammen mit einer Base die Ausbeute an c/s-Cedrendiol erheblich steigern konnten. Somit konnten Ausbeuten von über 80 % und sogar 90 % bis zu einem 100 g Maßstab erreicht werden. Dies konnte auch im 150 kg Maßstab reproduziert werden. Die erfindungsgemäße Umsetzung ist in Schema 1 dargestellt.

Die Anwesenheit der Base verhindert die unerwünschte Bildung von Cedralon. Cedrendiol ist unter sauren Bedingungen deutlich instabiler als unter basischen (siehe Schema 2).

Zudem begünstigt die Base die Bildung des Diols, weil sie die Abspaltung des Mangans aus dem Mangan-Cedrendiol-Komplex beschleunigt, wie in Schema 3 gezeigt, und es so nicht zu einerweiteren Oxidation des Diols kommen kann (Wiberg und Saegebarth, J. Am. Chem. Soc., 1957, 79, 11 , 2822-2824).

Unter einem protischen Lösungsmittel ist im Zusammenhang mit der vorliegenden Erfindung jedes Lösungsmittel zu verstehen, welches über eine funktionelle Gruppe verfügt, aus der Wasserstoff als Proton abgespalten werden kann. Da Wasser in H+ und OH- dissoziieren kann, stellt es ebenfalls ein protisches Lösungsmittel dar. Weitere protische Lösungsmittel sind primäre, sekundäre und tertiäre Alkohole sowie Carbonsäuren. Alkane, Ether, Ester, Ketone und Aldehyde dagegen stellen keine protischen Lösungsmittel im Sinne der vorliegenden Erfindung dar.
Unter einer Base ist im Zusammenhang mit der vorliegenden Erfindung eine Verbindung zu verstehen, die in wässriger Lösung Hydroxidionen bildet. Geeignete Basen für den Einsatz in Schritt ii) werden weiter unten beschrieben.
Als vorteilhaft haben sich sekundäre und tertiäre Alkohole und ganz besonders fe/f-Butanol und Isopropanol herausgestellt. Sekundäre und tertiäre Alkohole lösen Cedren gut und es kommt zu keiner Phasentrennung, wenn eine wässrige Lösung von Kaliumpermanganat zugegeben wird. Diese Lösungsmittel sind auch unter den Reaktionsbedingungen weitgehend inert, so dass sie nicht durch das Kaliumpermanganat oxidiert werden und somit die Notwendigkeit einer größeren Menge Oxidationsmittel vermieden werden kann. Dies trifft insbesondere für tertiäre Alkohole wie tert-Butanol zu, für Isopropanol jedoch nur bedingt, da eine Oxidation zu Aceton möglich ist. Allerdings ist Isopropanol noch vollständig mit Wasser mischbar, so dass immer noch gute Ausbeuten erhalten werden. Die genannten Lösungsmittel lassen sich auch leicht durch Destillation entfernen, da sie flüchtiger als Wasser sind.
In einer bevorzugten Ausführungsform umfasst das oben beschriebene Verfahren zusätzlich die Schritte: iv) Extrahieren des entstandenen c/s-Cedrendiols aus der Reaktionsmischung aus Schritt iii), und v) Entfernen des Extraktionsmittels aus Schritt iv) und Erhalten von cis- Cedrendiol.
Das entstandene c/s-Cedrendiol lässt sich durch Extraktion erhalten. In diesem Zusammenhang ist es wichtig, dass in Schritt iii) das weitere protische Lösungsmittel möglichst vollständig entfernt wird, damit dieses die Phasentrennung bei der anschließenden Extraktion nicht stört. Geeignete Extraktionsmittel werden weiter unten beschrieben.
Weiter bevorzugt ist ein Verfahren wie oben beschrieben, wobei in Schritt i) das alpha- Cedren eine Reinheit von mindestens 70 %, bevorzugt mindestens 80 %, besonders bevorzugt mindestens 90 % aufweist und/oder wobei das alpha-Cedren vor Schritt i) destilliert wird.
In Schritt i) kann beispielsweise Zedernöl verwendet werden, in welchem alpha-Cedren zu ca. 70 % vorliegt. Wenn das alpha-Cedren, z.B. in Form von Zedernöl, vor Schritt i) destilliert wird, kann eine Reinheit von über 90 % erreicht werden. Falls weniger reines alpha-Cedren eingesetzt wird, wird mehr Kaliumpermanganat und Base benötigt und es muss bei tieferer Temperatur dosiert werden.
In einer weiteren bevorzugten Ausführungsform des oben beschriebenen Verfahrens wird in Schritt i) das alpha-Cedren in einem Gewichtsverhältnis von 1 : 5 bis 1 : 10 alpha-Cedren zu dem weiteren protischen Lösungsmittel bereitgestellt, bevorzugt in einem Gewichtsverhältnis von 1 : 6 bis 1 : 9 alpha-Cedren zu dem weiteren protischen Lösungsmittel, besonders bevorzugt in einem Gewichtsverhältnis von 1 : 7 bis 1 : 8 alpha- Cedren zu dem weiteren protischen Lösungsmittel und/oder in Schritt i) wird das alpha- Cedren in einem Gewichtsverhältnis von 1 : 1 bis 1 : 6 alpha-Cedren zu Wasser bereitgestellt, bevorzugt in einem Gewichtsverhältnis von 1 : 2 bis 1 : 5 alpha-Cedren zu Wasser, besonders bevorzugt in einem Gewichtsverhältnis von 1 : 3 bis 1 : 4 alpha-Cedren zu Wasser.
Es hat sich gezeigt, dass mit weniger weiterem protischem Lösungsmittel im Verhältnis zu alpha-Cedren der Umsatz unvollständig ist, weil Cedren ausölt und sich abscheidet. Wenn weniger Wasser im Verhältnis zu Cedren eingesetzt wird, komm es zwar zu einem vollständigen Umsatz aber mit weniger Ausbeute. Werden die angegebenen Gewichtsverhältnisse eingehalten, ist der Umsatz vollständig und die Ausbeute gut.
Bevorzugt ist auch ein Verfahren wie oben beschrieben, bei dem das Zugeben in Schritt ii) über einen Zeitraum von 30 Minuten bis 10 Stunden, bevorzugt 3 bis 7 Stunden, besonders bevorzugt 4 bis 6 Stunden stattfindet.
Bei dem erfindungsgemäßen Verfahren kann ein guter Umsatz und gute Ausbeuten bereits nach wenigen Stunden erreicht werden. Wird allerdings schneller dosiert, sinkt die GC- Reinheit (Reinheit gemäß Gaschromatographie) des Endproduktes. Gute Ergebnisse werden ab 3 bis 4 Stunden Dosierungszeit erhalten. Die Dosierung kann in dem erfindungsgemäßen Verfahren aber auch über einen Zeitraum länger als 10 Stunden erfolgen, z.B. über 15 oder 20 Stunden. In einer weiteren bevorzugten Ausführungsform des oben beschriebenen Verfahrens ist die Base eine anorganische Base, bevorzugt ausgewählt aus der Gruppe der Alkali- und Erdalkalihydroxide und Alkali- und Erdalkalicarbonate, besonders bevorzugt ausgewählt aus NaOH, KOH, K2CO3 und CaCOs.
Weiter bevorzugt ist auch ein Verfahren wie oben beschrieben, bei dem in Schritt ii) das Kaliumpermanganat in einer Menge von 0.8 bis 5 Äquivalenten bezogen auf das in Schritt i) bereitgestellte alpha-Cedren zugegeben wird, bevorzugt in einer Menge von 1 bis 3 Äquivalenten, besonders bevorzugt in einer Menge von 1 .2 bis 1 .5 Äquivalenten und/oder
wobei die Base in einer Menge von 0.5 bis 3 Äquivalenten bezogen auf das in Schritt i) bereitgestellte alpha-Cedren zugegeben wird, bevorzugt in einer Menge von 1 bis 1.5 Äquivalenten, besonders bevorzugt in einer Menge von 1 .2 bis 1.3 Äquivalenten.
Unter Einhaltung der oben spezifizierten molaren Verhältnisse der Reagenzien, d. h. Kaliumpermanganat und Base zu eingesetztem alpha-Cedren wurden besonders gute Ausbeuten erzielt.
In einerweiteren bevorzugten Ausführungsform des oben beschriebenen Verfahrens wird in Schritt ii) eine Mischung enthaltend oder bestehend aus Kaliumpermanganat, Base und Wasser zu der Lösung aus Schritt i) gegeben, bevorzugt wird dabei die Mischung während des Zugebens ständig gerührt und/oder es wird ein Gas durch die Mischung durchgeleitet.
Die Zugabe von Kaliumpermanganat in wässriger Lösung und nicht als Feststoff hat den Vorteil, dass die Konzentration an Kaliumpermanganat lokal nicht so stark erhöht wird. Zudem baut sich die lokal erhöhte Konzentration bei Zugabe von bereits gelöstem Kaliumpermanganat auch schneller wieder ab, als wenn Kaliumpermanganat als Feststoff zugegeben wird. Durch eine lokal erhöhte Konzentration können Nebenreaktionen begünstigt werden, aus denen höher oxidierte Produkte resultieren.
Bevorzugt liegt in der Mischung, die in Schritt ii) zu der Lösung aus Schritt i) gegeben wird, das Wasser in einem Gewichtsverhältnis von 1 : 5 bis 1 : 20 Kaliumpermanganat zu Wasser vor, bevorzugt 1 : 10 bis 1 : 15 Kaliumpermanganat zu Wasser, besonders bevorzugt 1 : 12 bis 1 : 14 Kaliumpermanganat zu Wasser.
Da sich die Mischung an der Löslichkeitsgrenze des Kaliumpermanganats befindet, ist es von Vorteil die Mischung während des Zugebens ständig zu rühren, oder ein Gas, z.B. Stickstoff, durch die Mischung zu leiten. Durch das Rühren bzw. die Gasblasen, wird die Mischung in Bewegung gehalten und das Kaliumpermanganat bleibt in Lösung. In einer Ausführungsform des oben beschriebenen Verfahrens wird in Schritt i) das alpha- Cedren in dem Lösungsmittel bei einer Temperatur im Bereich von 5 bis 40 °C bereitgestellt, bevorzugt 15 bis 30 °C, besonders bevorzugt 18 bis 22 °C und/oder in Schritt ii) das Kaliumpermanganat und die Base mit einer Temperatur von 10 bis 40 °C zugegeben, bevorzugt 15 bis 30 °C, besonders bevorzugt 20 bis 24 °C.
Das erfindungsgemäße Verfahren kann in einem relativ großen Temperaturbereich durchgeführt werden, insbesondere auch bei Temperaturen um Raumtemperatur, so dass keine Heizung oder Kühlung notwendig ist. Wird bei höheren Temperaturen dosiert, sinkt die GC-Reinheit des Endprodukts. Eine Dosierung bei tieferer Temperatur (ca. 10 °C) funktioniert ebenfalls, verbessert jedoch nicht das Ergebnis der Reaktion.
Gemäß einerweiteren bevorzugten Ausführungsform des oben beschriebenen Verfahrens erfolgt das Entfernen des weiteren protischen Lösungsmittels in Schritt iii) mittels Destillation.
Wie bereits weiter oben erwähnt, ist es für die Aufreinigung des Produkts mittels Extraktion wichtig, dass das weitere protische Lösungsmittel in Schritt iii) möglichst vollständig entfernt wird, damit die Phasentrennung nicht gestört wird. Die Entfernung mittels Destillation erzielt besonders gute Ergebnisse. Wird fe/f-Butanol als weiteres protisches Lösungsmittel eingesetzt, wird es bevorzugt als Azeotrop mit einem Anteil von 10 bis 30 % Wasser, besonders bevorzugt als Azeotrop mit einem Anteil von 15 bis 25 % Wasser abdestilliert.
Nach der Destillation ist es vorteilhaft den Destillationsrückstand auf etwa 40 °C abzukühlen bevor das Extraktionsmittel zugegeben wird. Dadurch wird gewährleistet, dass das Cedrendiol nicht teilweise wieder zerstört wird (Bildung von Cedralon), wenn im folgenden Schritt das Gemisch angesäuert wird. Die Dosierung der Schwefelsäure und insbesondere der Natriumhydrogensulfitlösung sind auch leicht exotherm.
In einer weiteren Ausführungsform des erfindungsgemäßen Verfahrens wird als Extraktionsmittel in Schritt iv) ein aprotisches Extraktionsmittel verwendet, bevorzugt ausgewählt aus der Gruppe bestehend aus Ethern und Estern, besonders bevorzugt ausgewählt aus der Gruppe bestehend aus Ethylacetat und Methyl-fe/f-butylether. Unter einem aprotischen Lösungsmittel ist im Kontext der vorliegenden Erfindung ein Lösungsmittel zu verstehen, welches nicht über eine funktionelle Gruppe verfügt, aus der Wasserstoff als Proton abgespalten werden kann. Aprotische Lösungsmittel sind beispielsweise aliphatische oder aromatische Alkane oder Alkene, Ester, Ether, Ketone oder Aldehyde. Insbesondere in Ethylacetat ist die Löslichkeit von Cedrendiol sehr gut, so dass eine hohe Raum-Zeit-Ausbeute erhalten wird.
Bevorzugt wird in dem oben beschriebenen Verfahren in Schritt iv) das Extraktionsmittel in einem Gewichtsverhältnis von 1 : 2 bis 1 : 20 des in Schritt i) bereitgestellten alpha-Cedrens zu Extraktionsmittel verwendet, bevorzugt in einem Gewichtsverhältnis von 1 : 5 bis 1 : 12 des in Schritt i) bereitgestellten alpha-Cedrens zu Extraktionsmittel, besonders bevorzugt in einem Gewichtsverhältnis von 1 : 8 bis 1 : 9.5 des in Schritt i) bereitgestellten alpha- Cedrens zu Extraktionsmittel.
Um das Produkt vollständig zu extrahieren, sind bestimmte Mengen an Extraktionsmittel nötig. Das Extraktionsmittel kann auch in zwei oder mehr Portionen zugegeben und die Extraktionsmittelphase dazwischen abgetrennt werden, um die Raum-Zeit-Ausbeute des Verfahrens zu steigern.
Als weitere Maßnahme zur Verbesserung der Effizienz bei der Extraktion kann Schwefelsäure zudosiert werden. Ist es hierbei allerdings zu warm (> 40 °C), bildet sich etwas mehr Cedralon als Nebenprodukt. Zudem kann auch eine Natriumhydrogensulfitlösung zudosiert werden, welche als Reduktionsmittel fungiert und sämtliches im Gemisch enthaltenes Mangan zu Mangan(ll) reduziert, welches gut wasserlöslich ist. Bei dieser Redox-Reaktion werden Protonen verbraucht, daher läuft sie bevorzugt in saurer Umgebung. Das Sulfit wird dabei zu Sulfat oxidiert. Gegen Ende der Dosierung kann die Temperatur durch eine Begleitheizung auf ca. 50 bis 55 °C angehoben werden. Dadurch entstehen zwei klare Phasen und durch die erhöhte Temperatur kommt es zu einer besseren Phasentrennung. Es kann hierbei bereits etwas Cedrendiol ausfallen, was durch weitere Zugabe von Extraktionsmittel bzw. eine Temperaturerhöhung wieder in Lösung gebracht werden kann. Die wässrige Phase kann dann abgetrennt und die Extraktion mit einer weiteren Portion Extraktionsmittel wiederholt werden. Durch Zugabe von Soda und Wasser können letzte Reste von Schwefelsäure aus der organischen Phase entfernt bzw. neutralisiert werden. Um die Lagerstabilität des Cedrendiols zu erhöhen, sollten keine Säurespuren im Cedrendiol verbleiben. Zusätzlich kann noch mit Natriumchlorid/Wasser gewaschen werden, damit sichergestellt ist, dass keine Säurereste im Cedrendiol verbleiben und die entstehende Cedrendiol-Lösung annährend neutral ist. Durch das Natriumchlorid trennen sich die Phasen besser/schneller und zudem gelangt weniger organische Verbindung ins Abwasser. Das Produkt fällt in der Extraktionsmittelphase beim Abkühlen gegebenenfalls bereits teilweise aus. Durch Entfernen des Extraktionsmittels wird das c/s-Cedrendiols in einer GC-Reinheit von über 90 % und in Ausbeuten von über 70 %, bevorzugt über 80 %, besonders bevorzugt über 90 % erhalten.
Die vorliegende Erfindung betrifft auch die Verwendung einer Mischung umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel, bevorzugt ausgewählt aus der Gruppe bestehend aus sekundären und tertiären Alkoholen, besonders bevorzugt ausgewählt aus der Gruppe bestehend aus fe/f-Butanol und Isopropanol, als Lösungsmittel bei der Oxidation von alpha-Cedren mit Kaliumpermanganat, vorzugsweise in einem Verfahren wie oben beschrieben.
Durch den Einsatz der Mischung umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel konnte die Ausbeute an c/s-Cedrendiol entscheidend gesteigert werden. Dadurch wird es erstmals möglich c/s-Cedrendiol in einem Schritt durch Oxidation mit Kaliumpermanganat in einer Ausbeute zu erhalten, die eine wirtschaftliche Anwendung erlaubt.
Je nach Ausgangsmaterial können bei dem erfindungsgemäßen Verfahren zwei Isomere von cis-Cedrendiol gebildet werden (Verbindungen der Formeln (I) und (II), siehe unten). Dabei liegen die beiden Isomere jeweils in einer charakteristischen Menge vor, wobei die Verbindung (I) deutlich überwiegt. Beispielweise entsteht eine Mischung, in der die Verbindung der Formel (I) zu 70 bis 99 Gew.-% enthalten ist, besonders bevorzugt zu 85 bis 95 Gew.-%, und/oder die Verbindung der Formel (II) zu 0.1 bis 2 Gew.-% enthalten ist, besonders bevorzugt 0.3 bis 1.5 Gew.-%, jeweils bezogen auf das Gesamtgewicht der Mischung.
(l) (II)
Im Folgenden wird die Erfindung anhand von Beispielen weiter charakterisiert, wobei diese Beispiele nicht limitierend zu verstehen sind.
Beispiel 1: Herstellung von c/s-Cedrendiol
1) alpha-Cedren (1.00 g, 1 .07 mL, 4.89 mmol) wird in einer Mischung von tBuOH (10 mL) und H2O (5 mL) gelöst.
2) Die farblose Lösung wird auf 0 °C gekühlt und eine Mischung aus KMnC (1.12 g, 7.09 mmol) und NaOH (0.254 g, 6.36 mmol) in H2O (15 mL) wird langsam (über einen Zeitraum von 45 min) bei 0 °C dazugegeben. Die Reaktionstemperatur wird dabei unter 10 °C gehalten. Nach Zugabe wird die Reaktionsmischung bei 0 °C für 25 min gerührt.
3) Nach vollständiger Umsetzung des alpaha-Cedrens (die Reaktion wird mittels DC (Dünnschichtchromatographie) kontrolliert), wird die Reaktionsmischung durch einen Celitfilter (h = 1.5 cm, d = 5 cm) filtriert. Der Celitfilter wird mit EtOAc (30 mL) gewaschen.
4) Das Filtrat wird mit NaCI gesättigt und mit EtOAc (3x20 mL) extrahiert. Die organische Phase wird mit 10% Na2S203-Lösung (1 x20 mL) gewaschen, über Na2SÜ4 getrocknet und die flüchtigen Komponenten werden verdampft, wobei cis-Cedrendiol als weißer Feststoff (893 mg, 3.75 mmol, 77 %) erhalten wird. Rf (S1O2, CH: EtOAc, 3:1) = 0.18.
Beispiel 2: Herstellung von c/s-Cedrendiol
1) Cedren (Reinheit ca. 93 %, 100 g, 0.46 mmol, 1.0 Äq) wird in fe/f-Butanol (720 g) und Wasser (360 g) vorgelegt und auf 20 °C temperiert.
2) Innerhalb von etwa 5 h wird ein Gemisch aus Kaliumpermanganat (98 g, 0.62 mol, 1.35 Äq), Wasser (1320 g) und NaOH (22.4 g, 0.56 mol, 1.22 Äq) bei einer Innentemperatur von
20-24 °C gleichmäßig dosiert (leicht exotherme Reaktion). Dabei wird durch das Gemisch kontinuierlich Stickstoff geleitet und/oder es wird kontinuierlich gerührt. Es entsteht ein braunes Gemisch mit Niederschlag. 3) Das fe/f-Butanol wird als Azeotrop mit ca. 20 % Wasser bis 140 mbar abdestilliert. Die
Begleitheizung wird dabei auf 80 °C eingestellt. Insgesamt werden ca. 910 g Destillat erhalten. Das dabei erhaltene fe/f-Butanol kann erneut für die Reaktion eingesetzt werden.
4) Anschließend wird auf 40 °C abgekühlt und Ethylacetat (700 g) zugeben.
5) Bei 40°C wird Schwefelsäure (40 %, 200 g) zudosiert.
6) Bei etwa 40°C wird Natriumhydrogensulfit-Lösung (40 %, 215 g) zudosiert und gegen Ende der Dosierung die Begleitheizung bereits auf 55 °C angehoben. Es entstehen zwei klare Phasen und etwas Cedrendiol fällt aus.
7) Die wässrige Phase (ca. 2300 g) wird abgetrennt. 8) Ethylacetat (700 g), Soda (40 g) und Wasser (360 g) werden zugeben. Dadurch wird das ausgefallene Cedrendiol wieder gelöst.
9) Es wird auf ca. 55 °C temperiert und dabei gut durchgemischt.
10) Die wässrige Phase wird abgetrennt.
11) Natriumchlorid (30 g) und Wasser (270 g) werden zugeben. 12) Es wird erneut auf 55 °C temperiert und dabei gut durchgemischt.
13) Die wässrige Phase wird abgetrennt.
Das Produkt fällt anschließend in Ethylacetat an; das Cedrendiol fällt beim Abkühlen teilweise aus. Wenn man das Lösungsmittel entfernt, erhält man ca. 102 g Cedrendiol der Formel (I) (Reinheit laut GC ca. 93 %, ca. 0.40 mmol, ca. 87 % Ausbeute). Andere, laut GC enthaltene Stoffe sind: Cedren (< 1%), Cedralon (<1 %), Cedrenol (ca. 1 %) und das Isomer der Formel (II) (< 1 %).
Claims
1. Verfahren zur Herstellung von c/s-Cedrendiol umfassend die Schritte: i) Bereitstellen von alpha-Cedren in einem Lösungsmittel umfassend oder bestehend aus Wasser und mindestens einem weiteren protischen Lösungsmittel, bevorzugt ausgewählt aus der Gruppe bestehend aus sekundären und tertiären Alkoholen, besonders bevorzugt ausgewählt aus der Gruppe bestehend aus fe/f-Butanol und Isopropanol, ii) Zugeben von Kaliumpermanganat und einer Base zu der Lösung aus Schritt i), und, optional iii) Entfernen des weiteren protischen Lösungsmittels aus der Reaktionsmischung aus Schritt ii).
2. Verfahren nach Anspruch 1 , wobei das Verfahren zusätzlich die Schritte umfasst: iv) Extrahieren des entstandenen c/s-Cedrendiols aus der Reaktionsmischung aus Schritt iii), und v) Entfernen des Extraktionsmittels aus Schritt iv) und Erhalten von c/s-Cedrendiol.
3. Verfahren nach Anspruch 1 oder 2, wobei in Schritt i) das alpha-Cedren eine Reinheit von mindestens 70 %, bevorzugt mindestens 80 %, besonders bevorzugt mindestens 90 % aufweist und/oder wobei das alpha-Cedren vor Schritt i) destilliert wird.

Verfahren nach einem der vorhergehenden Ansprüche, wobei in Schritt i) das alpha-Cedren in einem Gewichtsverhältnis von 1 : 5 bis 1 : 10 alpha-Cedren zu dem weiteren protischen Lösungsmittel bereitgestellt wird, bevorzugt in einem Gewichtsverhältnis von 1 : 6 bis 1 : 9 alpha-Cedren zu dem weiteren protischen Lösungsmittel, besonders bevorzugt in einem Gewichtsverhältnis von 1 : 7 bis 1 : 8 alpha-Cedren zu dem weiteren protischen Lösungsmittel und/oder wobei in Schritt i) das alpha-Cedren in einem Gewichtsverhältnis von 1 : 1 bis 1 : 6 alpha-Cedren zu Wasser bereitgestellt wird, bevorzugt in einem Gewichtsverhältnis von 1 : 2 bis
1 : 5 alpha-Cedren zu Wasser, besonders bevorzugt in einem Gewichtsverhältnis von 1 : 3 bis 1 : 4 alpha-Cedren zu Wasser.
5. Verfahren nach einem der vorhergehenden Ansprüche, wobei das Zugeben in Schritt ii) über einen Zeitraum von 30 Minuten bis 10 Stunden, bevorzugt 3 bis 7 Stunden, besonders bevorzugt 4 bis 6 Stunden stattfindet.
6. Verfahren nach einem der vorhergehenden Ansprüche, wobei die Base eine anorganische Base ist, bevorzugt ausgewählt aus der Gruppe der Alkali- und Erdalkalihydroxide und Alkali- und Erdalkalicarbonate, besonders bevorzugt ausgewählt aus NaOH, KOH, K2CO3 und CaC03.
7. Verfahren nach einem der vorhergehenden Ansprüche, wobei in Schritt ii) das
Kaliumpermanganat in einer Menge von 0.8 bis 5 Äquivalenten bezogen auf das in Schritt i) bereitgestellte alpha-Cedren zugegeben wird, bevorzugt in einer Menge von 1 bis 3 Äquivalenten, besonders bevorzugt in einer Menge von 1.2 bis 1.5 Äquivalenten und/oder wobei die Base in einer Menge von 0.5 bis 3 Äquivalenten bezogen auf das in Schritt i) bereitgestellte alpha-Cedren zugegeben wird, bevorzugt in einer Menge von 1 bis 1.5 Äquivalenten, besonders bevorzugt in einer Menge von 1 .2 bis 1 .3 Äquivalenten.

Verfahren nach einem der vorhergehenden Ansprüche, wobei in Schritt ii) eine Mischung enthaltend oder bestehend aus Kaliumpermanganat, Base und Wasser zu der Lösung aus Schritt i) gegeben wird, bevorzugt wobei die Mischung während dem Zugeben ständig gerührt und/oder wobei ein Gas durch die Mischung durchgeleitet wird.
9. Verfahren nach Anspruch 8, wobei das Wasser in der Mischung, die in Schritt ii) zu der Lösung aus Schritt i) gegeben wird, in einem Gewichtsverhältnis von 1 : 5 bis 1 : 20 Kaliumpermanganat zu Wasser vorliegt, bevorzugt 1 : 10 bis 1 : 15
Kaliumpermanganat zu Wasser, besonders bevorzugt 1 : 12 bis 1 : 14 Kaliumpermanganat zu Wasser.
10. Verfahren nach einem der vorhergehenden Ansprüche, wobei in Schritt i) das alpha-Cedren in dem Lösungsmittel bei einer Temperatur im Bereich von 5 bis 40 °C bereitgestellt wird, bevorzugt 15 bis 30 °C, besonders bevorzugt 18 bis 22 °C und/oder in Schritt ii) Kaliumpermanganat und die Base mit einer Temperatur von 10 bis 40 °C zugegeben werden, bevorzugt 15 bis 30 °C, besonders bevorzugt 20 bis 24 °C.
11. Verfahren nach einem der vorhergehenden Ansprüche, wobei das Entfernen des weiteren protischen Lösungsmittels in Schritt iii) mittels Destillation erfolgt.
12. Verfahren nach einem der Ansprüche 2 bis 11 , wobei als Extraktionsmittel in Schritt iv) ein aprotisches Extraktionsmittel verwendet wird, bevorzugt ausgewählt aus der Gruppe bestehend aus Ethern und Estern, besonders bevorzugt ausgewählt aus der Gruppe bestehend aus Ethylacetat und Methyl-fe/f-butylether.
13. Verfahren nach einem der Ansprüche 2 bis 12, wobei in Schritt iv) das Extraktionsmittel in einem Gewichtsverhältnis von 1 : 2 bis 1 : 20 des in Schritt i) bereitgestellten alpha-Cedrens zu Extraktionsmittel verwendet wird, bevorzugt in einem Gewichtsverhältnis von 1 : 5 bis 1 : 12 des in Schritt i) bereitgestellten alpha-
Cedrens zu Extraktionsmittel, besonders bevorzugt in einem Gewichtsverhältnis von 1 : 8 bis 1 : 9.5 des in Schritt i) bereitgestellten alpha-Cedrens zu Extraktionsmittel.
