Description
- Die vorliegende Erfindung bezieht sich auf berührungslosen Messungen an Proben bzw. Probenmaterialien, wie z.B. biologischen und/oder chemischen Proben, Systeme für solche Messungen sowie Probenträger, die für solche Systeme geeignet sind.
- Es wird davon ausgegangen, dass µTAS (Micro Total Analysis Systems = Mikrogesamtanalysensysteme) oder Lab-on-Chips in Bereichen, wie z.B. globaler Gesundheits- und Medizinforschung, eine wichtige Rolle spielen werden. Sie reduzieren Kosten und Zeit für Tests und Analysen. Die Beschränkung, dass dieselben Einwegartikel sind, die Materialauswahl, der Entwurf und die Herstellung sind alles Aspekte, die berücksichtigt werden müssen, um die Chipkosten niedrig zu halten und somit ein Bauelement mit Marktfähigkeit zu produzieren. Zahlreiche biologische und chemische Untersuchungen sind temperaturabhängig, wie z.B. PCR (Polymerase Chain Reaction = Polymerase-Kettenreaktion) und MCA (Melting Curve Analysis = Schmelzkurvenanalyse). Wenn eine solche Untersuchung auf Mikrogröße herunterskaliert wird, werden eine präzise Temperatursteuerung und eine thermische Homogenität integrierter miniaturisierter Erwärmungs- und Erfassungselemente in mikrofluidischen Merkmalen zu kritischen Faktoren für ein funktionsfähiges Bauelement.
- Mikroerwärmungsvorrichtungen sind allgegenwärtig in verschiedenen MEMS- und mikrofluidischen Bauelementen. Letztere stellen verschiedene Funktionen in physikalischen oder chemischen Sensoren (S-C Roth, Y-M Choi und S-Y Kim Sensors Actuators A, 2006, 128, 1-6, D. Briand, S. Colin und A. Gangadharaiah, Sensors Actuators A, 2006, 132, 317-24), chemischen Reaktoren (T. Becker, S. Muhlberger und W. Benecke, J. Microelectromech. Syst. 200, 9, 478-84, A. Splinter, J. Sturmann und O. Bartels, Sensors Actuators B, 2002, 83, 169-74) oder Pumpen ( Z. Yin und A. Prosperetti, J. Micromech. Microeng. 2005, 1683-91) usw. bereit. Ein typisches Layout einer Mikroerwärmungsvorrichtung ist eine dünne metallische oder dotierte Si-Schicht, die in einer Serpentinenform auf einem dielektrischen Substrat strukturiert ist. Eine Temperatur-Messung kann durch Integration einer zweiten Leiterbahn oder durch eine Vierpunktmessung ermöglicht werden. Unterschiedliche Anwendungen haben unterschiedliche Beschränkungen, aber allgemein ist eine thermische Homogenität über die erwärmte Oberfläche hinweg wichtig für genaue Messungen und Steuerungen. Das aber ist aufgrund physikalischer Tatsachen nicht ohne weiteres zu erreichen. Materialauswahl und Erwärmungsvorrichtungsentwurf sind Faktoren, die modifiziert werden können, um die Wärmeverteilung in dem erwärmten Bereich zu beeinträchtigen (W.-J. Hwang, K-S. Shin, und J-H Roh. Sensors, 2011, 11, 2580-2591, D. Caputo, G. de Cesare und M. Nardini, IEEE SENSORS JOURNAL, 2012, 12(5), 1209-13, http://www.allflexheaters.com/Profiled Heaters.cfm ).
- Für biologische Anwendungen ermöglicht eine optische Transparenz des Substrats eine Online-Überwachung und -Beobachtung der Probe. Transparente Erwärmungselemente für Lab-on-Chip-Anwendungen wurden hergestellt durch Strukturieren von ITO (Indium-Tin-Oxide = Indiumzinnoxid) auf Glas ( K. Sun, A. Yamaguchi und Y. Ishida, Sensors und Actuators B, 2002, 84, 283-289, J.-Y. Cheng, M.-H. Yen und C.-T. Kuo, Biomicrofluidics, 2008, 2, 024105-(1-12), J.-L. Lin, M.-H. Wu und C.-Y. Kuo, Biomed Microdevices, 2010, 12, 389-398, S. Kumar Jha, R. Chand und D. Han, Lab Chip, 2012, doi:10.1039/C2LC40727B). Aufgrund seines geringen Vorkommens in der Erdkruste ist ITO jedoch relativ teuer und folglich ungeeignet für kostengünstige Diagnosen.
- Transparente Elektroden sind auch bei einem großen Bereich von optoelektronischen Bauelementen, wie z.B. organische LEDs, Fotovoltaikzellen und Flüssigkristallanzeigen, wesentliche Elemente, die zur Spannungserzeugung für die optoelektronische bzw. elektrooptische Wandlung dienen. In diesen Bereichen wurden Metallgitter als halbtransparente Elektrodenmaterialien verwendet, um auf der Suche nach einem günstigeren Elektrodenmaterial das üblicherweise verwendete ITO zu ersetzen ( M.-G. Kang und L. J. Guo, Advanced Materials, 2007, 19(10), 1391-1396, M.-G. Kang, M-S. Kim und J. Kim, Advanced Materials, Adv. Mater. 2008, 20, 4408-4413) oder um den Widerstand zu verringern und elektrische Homogenität des Materials zu verbessern (S. Choi, W. J. Potscavage, Jr. und B. Kippelen, JOURNAL OF APPLIED PHYSICS, 2009, 106, 054507). Die Firma PolyIC hat eine Patentanmeldung für transparente leitfähige Oberflächen inne, die aus Metallnetzen hergestellt sind, für Anwendungen in der Fotovoltaik, bei Anzeigen und LED-Bereichen (
WO 2010/108692 A2 ). Ferner wurden Metallnetze seit Jahrzehnten (J. Zhang, P. A. R. Ade und P. Mauskopf, Appl. Opt. 2011, 50(21), 3750-7; P. A. R. Ade, G. Pisano und C. Tucker, Proc. ofSPIE Vol. 6275 62750U-1) in FIR und Submillimeterastronomieinstrumenten verwendet. - Wie im Vorhergehenden erwähnt, gibt es bereits Probenträger, bei denen eine Heizbahn mäanderförmig auf einem Substrat gebildet ist. Das Problem bei diesen Probenträgern besteht allerdings darin, dass eine Unterbrechung der Leiterbahn für die Erwärmungsvorrichtung unweigerlich zur Folge hätte, dass letztere ihre Funktion verliert. Dies erhöht den Herstellungsaufwand, denn entweder müssen bei der Herstellung Abmessungen, Materialauftrag usw. mit mehr Sicherheitsabständen zu ansonsten möglichen Grenzwerten eingestellt werden, oder die Produktion muss aufwendig überwacht werden, was wiederum dem Ziel nach möglichst geringen Herstellungskosten entgegensteht. Ein weiterer Nachteil einer mäanderförmigen Führung der Heizbahn über das Substrat des Probenträgers hinweg besteht in der inhärent auftretenden Flächendichteerhöhung an denjenigen Rändern des Substrates, an denen die Mäanderform ihre Schleifenumkehrpunkte aufweist.
- Die
US 2011/0056926 A1 schlägt zur Durchführung von PCR-Untersuchungen eine mikrofluidische Vorrichtung vor, die Heizelemente in Form von Dünnfilmwiderständen aufweist, die sich bahnenförmig jeweils entlang eines mikrofluidischen Kanals der mikrofluidischen Vorrichtung erstrecken, um einzeln ansteuerbar zu sein. - Die XP 11441122 offenbart zur Durchführung von PCR-Analysen als Heizvorrichtung einen Mikroskopglasprobenträger mit einem Widerstandsdünnfilm, der in Form einer Serpentine gebildet ist, die sich zwischen zwei Elektroden erstreckt.
- Die
WO 2009/019658 A2 offenbart eine integrierte Mikrofluidikvorrichtung mit mehreren PCR-Kammern, die einzeln befüllbar und über einen Serpentinenleiter beheizbar sind. - Die
US 2009/0211336 beschäftigt sich mit sogenannten "preconcentrators", die dazu da sind, einen interessierenden Analyten reversibel einzufangen, um denselben daraufhin in einer sogenannten "Desorptionsphase" wieder an einen Detektor abzugeben. Deshalb ist das Heizelement derUS 2009/0211336 als offenes Gitter ausgeführt, um es einem fluidischen Medium mit dem interessierenden Analyten zu ermöglichen, durch das Heizelement zu strömen, um in der Einfangphase von dem Heizelement eingefangen zu werden. - Die
EP2314552 A1 offenbart ein Heizelement mit einer transparenten Substanz, einer leitfähigen Heizleitung, Sammelschienen und einem Leistungsabschnitt, wobei 30% oder mehr der gesamten Fläche der transparenten Substanz eine leitfähige Heizleitungsstruktur aufweist, bei der, wenn die gerade Linie, die die leitfähige Heizleitung schneidet, gezogen wird, ein Verhältnis einer Standardabweichung bezüglich eines Mittelwerts von Abständen zwischen benachbarten Schnittpunkten der geraden Linie und der leitfähigen Heizleitung 2% oder mehr beträgt. - Die
EP1972375 A1 offenbart ein mikrofluidisches Bauelement, das ein zweidimensionales Array von Komponenten aufweist zum Verarbeiten eines Fluids und/oder zum Erfassen von Eigenschaften des Fluids. Jede Komponente ist mit zumindest einem Steueranschluss gekoppelt, was es einer aktiven Matrix ermöglicht, den Zustand jeder Komponente individuell zu ändern. Die Komponenten umfassen zumindest ein Heizelement. Die aktive Matrix umfasst ein zweidimensionales Array von elektronischen Komponenten, die in Dünnfilmtechnologie auf einem flexiblen Substrat realisiert sind, dies sorgt für eine hohe Anpassungsfähigkeit des Bauelements. - Die Aufgabe der vorliegenden Erfindung besteht deshalb darin, ein System und ein Verfahren zur Durchführung einer berührungslosen Messung einer Probe sowie einen Probenträger mit verbesserten Charakteristika zu schaffen.
- Diese Aufgabe wird durch die Gegenstände der unabhängigen Patentansprüche gelöst.
- Gemäß einem ersten Aspekt der vorliegenden Erfindung wird bei einer Durchführung einer berührungslosen Messung an einer Probe ein Probenträger verwendet, bei dem die Heizbahnen auf dem Substrat eine Parallelschaltung zwischen dem ersten und dem zweiten Heizabschluss bilden, so dass, wenn durch Anlegen einer Spannung zwischen dem ersten und dem zweiten Heizanschluss eine auf dem Probenträger aufgebrachte Probe erwärmt wird, die Erwärmungseigenschaften des Probenträgers auch dann nicht beeinträchtigt werden, wenn einzelne Unterbrechungen der Heizbahnen aufgrund von beispielsweise Prozessschwankungen bei der Herstellung der Heizbahnen vorhanden sind.
- Bilden die Heizbahnen sogar eine zweidimensionale Gitterstruktur, so wirken sich vereinzelte Unterbrechungen und Querschnittverjüngungen der Heizbahnen weniger negativ auf eine Homogenität der Probenerwärmung über die durch die Heizbahnen abgedeckte Erwärmungsregion des Substrats aus, da deren Auftreten in einzelnen Stegen des zweidimensionalen Gitters durch die Gitterstruktur bzw. benachbarte Stege überbrückt werden. Es ist also möglich, die Herstellungstoleranzen zu Gunsten geringerer Herstellungskosten zu reduzieren. Ferner ist es möglich, die Heizbahnen optisch intransparent auszuführen, und dennoch eine Semitransparenz des Probenträgers bei eventuell vorhandener Transparenz des Substrats beizubehalten, da die Gitterzwischenräume beziehungsweise Maschen des zweidimensionalen Gitters die Transparenz des Substrats in Dickerichtung des Substrats bewahren.
- Gemäß einem Ausführungsbeispiel werden die Heizbahnen derart angeordnet, dass die Flächendichte der Heizbahnen von einer Flächenmitte des Substrats nach außen hin zunimmt. Auf diese Weise ist es möglich, die ansonsten häufig
auftretenden Inhomogenitäten des Erwärmungsprofils über das Substrat hinweg, wonach meistens die Erwärmung am Rand des Substrats hin abnimmt, zu vermeiden. - Weitere bevorzugte Ausführungsbeispiele der vorliegenden Erfindung sind Gegenstand der abhängigen Patentansprüche. Fig. 1 zeigt ein System 10 zur Durchführung einer berührungslosen Messung an einer beispielsweise biologischen und/oder chemischen Probe 12. Bei der Probe kann es sich beispielsweise um eine Flüssigkeit handeln, wie zum Beispiel eine Lösung, eine Suspension oder dergleichen. Beispielsweise enthält die Probe 12 einen bestimmten Analyten. Bei der Probe 12 könnte es sich allerdings auch um einen Feststoff handeln, wie zum Beispiel ein Sediment. Bei dem vorerwähnten Analyten kann es sich um Atome, Moleküle oder andere Stoffe jeglicher Art handeln, wie zum Beispiel biologische Zellen, DNA, Gen-Antigen-Verbindungen usw. Im folgenden wird ohne Einschränkung und lediglich zu Veranschaulichung davon ausgegangen, dass es sich bei der Probe 12 um eine biologische/chemische Probe handelt. - Das System umfasst einen Probenträger 14 mit einem Substrat 15, einem ersten und einem zweiten Heizanschluss 16 und 18 sowie Heizbahnen 20. Die Heizbahnen 20 sind auf dem Substrat 15 angeordnet, wie zum Beispiel einer Vorderseite 22 desselben. Auf eben jener Vorderseite 22 können auch die Heizanschlüsse 16 und 18 angeordnet sein, wobei gemäß einer Alternative letztere aber auf einer der Vorderseite 22 abgewandten Rückseite 24 des Substrats 15 angeordnet sind oder auf einer Seitenfläche des Substrats 15.
- Wie es im Folgenden noch näher beschrieben werden wird, bilden die Heizbahnen 20 auf dem Substrat eine Parallelschaltung zwischen den Heizanschlüssen 16 und 18, so dass die biologische/chemische Probe nach Aufbringen derselben auf den Probenträger 14 durch Anlegen einer Spannung zwischen den Heizanschlüssen 16 und 18 erwärmbar ist.
- Das System 10 umfasst ferner eine Messanordnung 26 zur Durchführung einer berührungslosen Messung an der biologischen/chemischen Probe 12. In
Fig. 1 ist sie als eine optische Messanordnung mit einer Lichtquelle 28 und einer Optik 30 zur Durchführung einer transmissiven optischen Messung angedeutet, nämlich zur optischen Beobachtung durch einen Benutzer des Systems, aber hierzu existieren eine Vielzahl von Alternativen, wie zum Beispiel andere optische Messungen, wie zum Beispiel reflektive Messungen, interferometrische Messungen, oder nicht-optische Messungen. - Es wird darauf hingewiesen, dass, obwohl in
Fig. 1 die Probe 12 so dargestellt wurde, als ob sie auf der Vorderseite 22 beziehungsweise auf den Heizbahnen 20 aufgebracht sei, die Aufbringung zur Durchführung der berührungslosen Messung auch auf der Rückseite 16 erfolgen könnte. In jedem Fall erfolgt die Aufbringung so, dass in einer Projektion entlang einer Dickerichtung des Substrats 15 die Probe 12 und der durch die Heizbahnen 20 lateral abgedeckte Erwärmungsbereich 32 überlappen. -
Fig. 2 zeigt exemplarisch eine Draufsicht auf die Vorderseite 22 des Substrats 15, um eine mögliche Ausführungsform der Heizbahnen 20 und deren Führung auf der Vorderseite 22 zu zeigen. Zu sehen sind inFigur 2 die Heizanschlüsse 16 und 18 und die dazwischen befindlichen Heizbahnen 20. Gemäß dem Ausführungsbeispiel vonFig. 2 bilden die Heizbahnen 20 eine zweidimensionale Gitterstruktur aus Leiterbahngitterstegabschnitten 34, die sich in Gitterknotenpunkten treffen, und Gitterzwischenräumen beziehungsweise Maschen 36, die von nicht weiter verkleinerbaren Schleifen aus Stegen 34 umschlossen sind, und in welchen das Gitter aus Heizbahnen 20 das Substrat 15 freilegen lässt. In dem Fall vonFig. 2 sind die Gitterzwischenräume 36 exemplarisch quadratisch gebildet, sie könnten allerdings gemäß alternativen Ausführungsformen auch andere Formen aufweisen, wie zum Beispiel rechteckige, kreisförmige, sechseckige oder ähnliche Formen besitzen. Die Stege 34 können je nachdem, welche Maschenform vorliegt, zwischen den jeweiligen Gitterknoten 40, die sie verbinden, einen entlang ihrer Länge konstanten Querschnitt besitzen oder einen variablen, vor allem einen in einer Richtung quer zu ihrer Länge variierenden Querschnitt bei beispielsweise aber konstanter Dicke. Die Anzahl der sich in den Gitterknoten 40 treffenden Gitterstege 34 kann sich unter den Knoten 36 unterscheiden, je nachdem beispielsweise, ob sich der Knoten am Rand des Gitters befindet oder nicht. Die Formen der Maschen 36 können zueinander deckungsgleich sein, müssen dies aber nicht. Sie müssen auch keine ähnliche Form aufweisen, wie dies bei einem nachfolgend beschriebenen Ausführungsbeispiel noch der Fall ist. - Dadurch, dass die Heizbahnen 20 gemäß
Fig. 2 eine zweidimensionale Gitterstruktur bilden, können eventuell auftretende Unterbrechungen der Heizbahnen 20, wie sie exemplarisch inFig. 2 bei 38 in einem Steg des Gitters dargestellt sind, oder Querschnittverjüngungen der Heizbahnen 20 in Stegen des Gitters überbrückt werden, so dass keine negativen Auswirkungen auf die Erwärmungshomogenität über dem Erwärmungsbereich 32 hinweg auftreten beziehungsweise selbige gelindert werden. - Während in
Fig. 2 ein Probenträger gezeigt ist, bei dem die Heizbahnen 20 eine zweidimensionale Gitterstruktur bilden, die wiederum ein regelmäßiges Gitter bildet, bei dem eine Flächendichte der Heizbahnen 20 über die Fläche 22 des Substrats hinweg in dem Bereich 32 konstant ist, muss diese lateral konstante Flächendichte nicht unbedingt sein.Fig. 3 zeigt ein Ausführungsbeispiel, bei dem die zweidimensionale Gitterstruktur ein unregelmäßiges Gitter bildet, bei dem eine Flächendichte der Heizbahnen 20 von einer lateralen Mitte des unregelmäßigen Gitters nach außen hin zunimmt, d.h. die Maschen 36 vom Inneren des Gitters nach außen hin kleiner werden. Auch hier gilt, dass die Maschen nicht unbedingt zueinander ähnliche Formen aufweisen müssen. Vielmehr können sich die Formen der Maschen auch voneinander unterscheiden. - In den nachfolgend kurz beschriebenen Figuren sind verschiedene weitere Varianten exemplarisch dargestellt.
Fig. 4a zeigt beispielsweise, dass die die Maschen 36 umgebenden Stege 32 des aus den Heizbahnen 20 gebildeten Gitters nicht notwendigerweise geradlinig zwischen den Gitterknotenpunkten 40 verlaufen müssen, sondern auch lateral gekrümmt sein können.Fig. 4b zeigt, dass die Heizanschlüsse 16 und 18 durch Elektroden gebildet sein könnten, die in gegenüberliegenden Randbereichen 42 und 44 des beispielsweise rechteckigen beziehungsweise quaderförmigen Substrats 15 auf der rechteckigen Vorderseite 22 angeordnet sind. In anderen Worten ausgedrückt, befindet sich in dem Fall vonFig. 4b der Erwärmungsbereich 32 lateral zwischen den Heizanschlüssen 16 und 18. Er kann, wie es inFig. 4b veranschaulicht ist, länglich sein, um sich entlang seiner Länge zwischen den Heizanschlüssen 16 und 18 zu erstrecken, so dass sich letztere an den beiden Enden des Bereiches 32 befinden.Fig. 4c zeigt hierzu eine Alternative, wonach der Erwärmungsbereich 32 zwar länglich ist, aber entlang seiner Länge gebogen, um sich nichtgeradlinig wie eine Bahn über die Vorderseite 22 zu erstrecken, wobei sich über die Bahnbreite und die Bahnlänge des Bereichs 32 hinweg das durch die Heizbahnen 20 gebildete Gitter von einem Heizanschluss 16 zu dem anderen Heizanschluss 18 erstreckt. InFig. 4c sind auf diese Weise exemplarisch der Hauptanschluss 16 und der Hauptanschluss 18 in der Nähe der gleichen kürzeren Kante der hier exemplarisch rechteckigen Vorderseite 22 lokalisiert. -
Fig. 4d zeigt schließlich lediglich noch ein alternatives Beispiel eines Probenträgers zuFig. 1 , wonach eine Schutzschicht 46 die Vorderseite 22 des Substrats 15 sowie die darauf angeordneten Heizbahnen 20 und, wie hier exemplarisch der Fall, die Elektroden 16 und 18 bedeckt.Fig. 4e zeigt noch, dass die Vorderseite 22 des Substrats 15 nicht unbedingt für die Probe komplett freiliegend sein muss. Vielmehr ist es möglich, dass die Vorderseite 22 durch beispielsweise ein Deckelelement 90 bedeckt ist, derart, dass zwischen Vorderseite 22 des Substrats 15 und Deckelement 90 ein Kanal oder eine Kammer 92 gebildet wird, der entlang der Vorderseite 22 entlangführt bzw. die an die Vorderseite angrenzt, und zwar so, dass der Erwärmungsbereich 32 an den Kanal bzw. die Kammer 92 angrenzt. Auf diese Weise kann die beispielsweise flüssige Probe durch den Kanal 92 an den Erwärmungsbereich 32 vorbeigeführt werden bzw. durch Einfüllen in die Kammer an den Erwärmungsbereich 32 gebracht werden. Das Deckelelement 90 kann beispielsweise ein Substrat aus ebenfalls transparentem Material sein. Es kann an die Vorderseite 22 geklebt oder anderweitig befestigt sein. Das Beispiel vonFig. 4e ist natürlich mit dem Beispiel von 4d kombinierbar, d.h. es kann auch die Schutzschicht 22 zur Bedeckung der Heizbahnen 20 vorgesehen sein, wobei die Schutzschicht beispielsweise ein inertes Material sein kann. Das Material der Schutzschicht 22 kann beispielsweise ein aushärtbares Material, wie zum Beispiel Polymer, sein. - Bei dem Substrat 15 von
Fig. 1 handelt es sich um ein für das Licht des optischen Messsystems vorzugsweise transparentes Substrat aus einem transparenten Material, wie zum Beispiel Glas oder dergleichen. In dem Fall anderer Messanordnungen, die nicht-optisch arbeiten, könnte die Transparenz natürlich auch fehlen. Das Material der Heizbahnen 20 muss nicht notwendigerweise nach Gesichtspunkten der Transparenz ausgewählt werden. Es muss also nicht unbedingt aus ITO bestehen. Das Material für die Heizbahnen 20 kann Metall oder ein geeignetes Halbleitermaterial sein. Die Aufbringung kann mikrolithographisch erfolgen oder aber auf andere Weise. Das Substrat 15 kann starr oder flexibel sein. Wie es inFig. 2 noch exemplarisch dargestellt ist, kann zur Durchführung einer Vierpunktmessung der Probenträger zusätzlich auch Elektroden 50 und 52 aufweisen, die mit den Elektroden 16 beziehungsweise 18 einstückig gebildet sind. Eine inFig. 2 gestrichelt versinnbildlichte Ausleseschaltung 54 könnte über diese beiden Elektroden 50 und 52 einen Sensorwert erfassen, der ein Maß für einen Widerstand der Heizbahnen 20 sein könnte. Die Ausleseschaltung könnte Teil des Systems vonFig. 1 sein. -
Fig. 5 zeigt exemplarisch den Ablauf eines Verfahrens zur Durchführung einer berührungslosen Messung in einer Probe. Zunächst wird in einem Schritt 56 eine der im Vorhergehenden beschriebenen Probenträger bereitgestellt. Daraufhin wird in einem Schritt 58 die Probe auf dem Probenträger aufgebracht. Wie gesagt, kann die Aufbringung auf der Vorderseite 22 oder auf der Rückseite 24 erfolgen, allerdings jeweils lateral mit dem Erwärmungsbereich 32 überlappen. In einem Schritt 60 wird daraufhin die Probe erwärmt, indem eine Spannung zwischen den Elektroden 16 und 18 angelegt und damit ein Strom durch die Heizbahnen 20 zum Fließen gebracht wird. In einem Schritt 62 wird dann in dem aufgewärmten Zustand der Probe die berührungslose Messung an derselben durchgeführt. - In anderen Worten ausgedrückt, zeigten obige Ausführungen Beispiele für einen Probenträger, der als Erwärmungselement bei biologischen bzw. chemischen Anwendungen dienen könnte. Als Material für die Heizbahnen 20 könnte Metall verwendet werden. In dem Fall der Ausführung in Form eines Netzes bildete dann die Metallnetzstruktur ein Array von regelmäßigen oder allmählich in der Größe variierenden Öffnungen 36 in der Form von beispielsweise Quadraten oder Rechtecken, wie es im Vorhergehenden gezeigt wurde, oder von Kreisen oder jeglicher Art von regelmäßigen Polygonen. Die Heizbahnen könnten insbesondere aus einer elektrisch leitenden Dünnfilmschicht gebildet sein, wie zum Beispiel durch Strukturierung derselben. Die Dünnfilmschicht könnte auf das beispielsweise transparente, starre oder flexible Substrat 15 auf unterschiedliche Weise aufgebracht sein. Auch die Strukturierung kann auf unterschiedliche Art erfolgen.
- Die Anwendungen obiger Ausführungsbeispiele umfassen Proben in der Biologie oder Chemie. Biologische Anwendungen, wie z.B. Proteomik, Genetik und Zellenproben sowie Bioreaktoren mit dem Bedarf an Erwärmung und optischer Transparenz können in Betracht gezogen werden. Andere Anwendungen können Hydrogels und andere Polymersysteme sein, die Temperatursteuerung benötigen.
- Im Vergleich zu herkömmlichen Einzeldrahterwärmungstechnologien besitzen obige Ausführungsbeispiele, die eine Netzstruktur aus Heizbahnen verwenden, folgende Vorteile:
- Eine Netzstruktur stellt eine höhere Herstellungsrobustheit dar im Vergleich zu einem/einer Mäander/Serpentine. Falls eine Unterbrechung eines Leiters in einem Mäander auftreten würde, würde die Erwärmungsvorrichtung beziehungsweise der Probenträger ihre/seine Funktion verlieren, während eine Netzerwärmungsvorrichtungsstruktur nach wie vor funktionieren würde, da die elektrische Leitung anstatt nur über einen über mehrere Leiter beziehungsweise Stege 32 verteilt ist. Außerdem ist die Netzerwärmungsvorrichtung toleranter in Bezug auf die Kompensation von Abweichungen von der Entwurfsgeometrie des Leiters. Beispielsweise führen Leiterbeschränkungen aufgrund von Ätzdefekten zu lokalen hohen Widerständen und heißen Punkten in einer Mäanderstruktur. Die Netzstruktur versucht stattdessen die Quelle der Nicht-Einheitlichkeit zu kompensieren, indem sie Strom von den Hochwiderstandsbereichen weg leitet.
- Eine Mäandererwärmungsvorrichtung hat einen kreisförmigeren heißen Punkt (hot spot), wenn dieselbe erwärmt ist, während ein Netz einen eher rechteckigen aufweist, was bedeutet, dass das Netz die Wärme effizienter ausbreitet relativ zu ihrer eigenen Erwärmungsvorrichtungsoberfläche als eine Mäandererwärmungsvorrichtung. Folglich kann unter Verwendung eines Netzes eine größere Oberfläche einheitlich erwärmt werden. Vergleiche beispielsweise
Fig. 6 und7 .Fig. 6a zeigt eine Mäandererwärmungsvorrichtung mit 15 µm Leitung/Heizbahn und 150 µm Platz auf einem PEN-Foliensubstrat undFig. 6b zeigt ihr entsprechendes Wärmeprofil in einer auf dem Substrat befindlichen thermochromischen Flüssigkristallschicht bei 62°C. Die Erwärmungsvorrichtungsoberfläche ist 1,5 x 3 mm2.Fig. 7a zeigt demgegenüber eine Netzerwärmungsvorrichtung beziehungsweise einen Probenträger mit 15 µm Leitung/Heibahn und 150 µm Platz auf einem PEN-Foliensubstrat undFig. 7b zeigt ihr entsprechendes Wärmeprofil, wie es sich in einer thermochromischen Flüssigkristallschicht bei 62°C ergibt. Die Erwärmungsvorrichtungsoberfläche ist 1,5 x 3 mm2. Wie es zu sehen ist, verteilt sich die Wärme in dem Fall derFig. 7b gleichmäßiger. - Die Wärmeverteilung auf der Gesamterwärmungsvorrichtungsoberfläche kann weiter verbessert werden unter Verwendung des Arrays von Geometrien mit allmählich variierender Größe wie in
Fig. 3 , wo Wärmeverluste an der Peripherie der Erwärmungsvorrichtung kompensiert werden kann durch Erhöhen der Anzahl von Erwärmungsleitungen. - Durch Reduzieren der Netzgröße (dünnere Leitungen beziehungsweise Stege 32 und kleinere Öffnungen beziehungsweise Maschen 32) kann thermische Homogenität auf der Mikroskala (definiert durch Abmessungen, die viel geringer sind als die Substratdicke) erreicht werden (vgl.
Fig. 7 und8 ). Auf diese Weise kann eine Erwärmungsvorrichtung auf einem Substrat mit geringer thermischer Leitfähigkeit, wie z.B. Glas oder Polymer, in nur einem Metallisierungsschritt hergestellt werden; d.h. keine zusätzliche Wärmeausbreitungsschicht, die die Herstellungskosten erhöht und die Transparenz begrenzt, ist nötig.Fig. 8a zeigt im Vergleich mitFig. 7a eine Netzerwärmungsvorrichtung mit 5 µm Leitung und 50 µm Platz auf einem PEN-Foliensubstrat undFig. 8b zeigt ihr entsprechendes Wärmeprofil, wie es sich in einer thermochromischen Flüssigkristallschicht bei 62°C ergibt. Die Erwärmungsvorrichtungsoberfläche ist 1,5 x 3 mm2. - Falls eine oder mehrere Metallschichten (z.B. aufgedampft oder gesputtert) zur Bildung der Bahnen 20 verwendet wird, ist/sind eine oder mehrere sehr dünne Schichten ausreichend, um die angemessene Widerstandsfähigkeit zu erreichen. Dies ist besonders vorteilhaft für Proben, wo die Erwärmungsvorrichtungstopografie eine weitere Verarbeitung stört, oder, wenn dieselbe in fluidische Strukturen integriert ist, Flusscharakteristika in Kanälen stören könnte, wie z.B. in dem Fall von
Fig. 4e . - Die Heizbahnen müssen nicht aus ITO gebildet sein, was teuer ist. Das fördert folglich eine kostengünstige Herstellung, die für Diagnoseprodukte wesentlich ist.
- Ein Netz hat eine geringere Widerstandsfähigkeit als ein Mäander. Daher muss für eine gegebene Erwärmungsleistung an das Netz weniger Spannung angelegt werden als an eine Mäandererwärmungsvorrichtung. Dies ist ein Vorteil für Systeme mit niedrigen Versorgungsspannungen, insbesondere batteriebetriebene tragbare Vorrichtungen.
- Weitere Alternativen zu obigen Ausführungsbeispielen wären natürlich denkbar.
Claims (9)
- Probenträger (14) für eine Probe zur Durchführung einer berührungslosen Messung an der Probe mit folgenden Merkmalen:einem Substrat (15) in Form eines transparenten PEN-Foliensubstrats;einem ersten und einem zweiten Heizanschluss (16, 18) auf einer Vorderseits des Substrats; undHeizbahnen (20), die auf der der Vorderseits des Substrats angeordnet sind und eine Parallelschaltung zwischen dem ersten und dem zweiten Heizanschluss (16, 18) bilden, indem die Heizbahnen (20) eine zweidimensionale Gitterstruktur bilden, die sich zwischen dem ersten und dem zweiten Anschluss erstreckt,wobei der erste und der zweite Heizanschluss (16, 18) durch eine erste und eine zweite Elektrode gebildet werden,wobei die erste und die zweite Elektrode in gegenüberliegenden Randbereichen des Substrats (15) angeordnet sind,wobei die erste Elektrode einstückig mit einer dritten Elektrode verbunden ist, die seitlich neben der erste Elektrode auf dem Substrat (15) angeordnet ist, und die zweite Elektrode einstückig mit einer vierten Elektrode verbunden ist, die seitlich neben der zweiten Elektrode auf dem Substrat (15) angeordnet ist, so dass eine elektrische Messung zwischen der dritten und vierten Elektrode möglich ist.
- Probenträger (14) nach Anspruch 1, wobei die Heizbahnen (20) derart angeordnet sind, dass eine Flächendichte der Heizbahnen (20) von einer Flächenmitte des Substrats nach außen hin zunimmt.
- Probenträger (14) nach Anspruch 1 oder 2,
wobei die zweidimensionale Gitterstruktur eine Anordnung von Gitterzwischenräumen (36), die eine gleiche Form aus einer quadratischen, rechteckigen oder kreisförmigen Form aufweisen. - Probenträger (14) nach einem der Ansprüche 1 bis 3,
wobei die Heizbahnen (20) aus einer strukturierten elektrisch leitfähigen Dünnfilmschicht auf dem Substrat (15) gebildet sind. - Probenträger (14) nach einem der Ansprüche 1 bis 4,
wobei das Substrat (15) des Probenträgers (14) biegsam ist. - Probenträger (14) nach Anspruch 1,
wobei die zweidimensionale Gitterstruktur ein regelmäßiges Gitter bildet, bei dem eine Flächendichte der Heizbahnen (20) lateral konstant ist. - System zur Durchführung einer berührungslosen Messung an einer Probe, mit folgenden Merkmalen:einem Probenträger (14) für eine Probe zur Durchführung einer berührungslosen Messung an der Probe gemäß Anspruch 1;einer Messanordnung (26) zur Durchführung einer berührungslosen Messung an der Probe,wobei das System ferner eine Steuerung und eine Ausleseschaltung aufweist,wobei die Steuerung ausgebildet ist, um über die erste und die zweite Elektrode (A, B) eine Spannung für einen Heizstrom durch die Heizbahnen (20) anzulegen,wobei die Ausleseschaltung ausgebildet ist, um über die dritte und die vierte Elektrode einen Sensorwert zu erfassen, der ein Maß für einen Widerstand der Heizbahnen (20) ist.
- Das System nach Anspruch 7,
wobei die Messanordnung (26) ausgebildet ist, eine Messung an der Probe mittels Licht durchzuführen, und das Substrat (15) des Probenträgers (14) für das Licht transparent ist. - Verfahren zur Durchführung einer berührungslosen Messung an einer Probe, mit folgenden Schritten:Bereitstellen eines Probenträgers (14) miteinem Substrat (15) in Form eines transparenten PEN-Foliensubstrats; einem ersten und einem zweiten Heizanschluss (16, 18) auf einer Vorderseite des Substrats; undHeizbahnen (20), die auf der der Vorderseite des Substrats angeordnet sind und eine Parallelschaltung zwischen dem ersten und dem zweiten Heizanschluss (16, 18) bilden, indem die Heizbahnen (20) eine zweidimensionale Gitterstruktur bilden, die sich zwischen dem ersten und dem zweiten Anschluss erstreckt,wobei der erste und der zweite Heizanschluss (16, 18) durch eine erste und eine zweite Elektrode gebildet werden,wobei die erste und die zweite Elektrode in gegenüberliegenden Randbereichen des Substrats (15) angeordnet sind,wobei die erste Elektrode einstückig mit einer dritten Elektrode verbunden ist, die seitlich neben der erste Elektrode auf dem Substrat (15) angeordnet ist, und die zweite Elektrode einstückig mit einer vierten Elektrode verbunden ist, die seitlich neben der zweiten Elektrode auf dem Substrat (15) angeordnet ist;Aufbringen der Probe auf den Probenträger (14);Erwärmen der Probe durch Anlegen einer Spannung zwischen dem ersten und dem zweiten Heizanschluss (16, 18); undDurchführung der berührungslosen Messung an der Probe und einer elektrischen Messung zwischen der dritten und vierten Elektrode.
