isohydrischen pH (= 7,4) im Bereich von etwa pH 4 - 9 akzeptiert werden (Physikalische Pharmazie, Ed.: H. Stricker, Wissenschaftliche Verlagsgesellschaft mbH Stuttgart, 3. Aufl., 1987). In Tabelle 1 sind pharmazeutisch gebräuchliche Puffersysteme für die parenterale Anwendung aufgelistet (Pharmazeutische Technologie, Eds.: H. Sucker, P. Fuchs und P. Eine bedeutende Gruppe von parenteral angewendeten Arznei¬ mitteln stellen die iodhaltigen Röntgenkontrastmittel dar, die zumeist als Infusionslösungen eingesetzt werden. Hier finden als Puffersubstanzen neben den in Tab. 1 aufgeführten Trometamol [Tris (hydroxymethyl)aminomethan, 2-Amino-2- (hydroxymethyl)-1,3-propandiol] (enthalten z.B. in Ultravist®, Omnipaque oder Solutrast } und Natrium- dihydrogenphosphat (z.B. in Conray ) beispielsweise auch Natriumhydrogencarbonat (z.B. in Isovist ) Verwendung.
Aufgrund seiner guten Pufferwirkung im Bereich von pH 7,2 - 9, 0 und der damit einhergehenden Möglichkeit der Einstellung eines isohydrischen pH-Wertes hat der Trometamol-Puffer (= Tris) hier eine Sonderstellung erlangt. Im Vergleich zum Phosphatpuffer, der den pH-Bereich von 5 - 8 abdeckt, sind für den Trometamol-Puffer darüber hinaus keine Probleme hinsichtlich einer möglichen Fällung mit Ionen aus dem Glasprimärbehältnis beschrieben.An important group of medicaments used parenterally are the iodine-containing X-ray contrast media, which are mostly used as infusion solutions. Infolge des Abfalls des pH-Wertes von Trometamol- oder Meglumin-gepufferten Lösungen (pH 7,0 - 7,6) auf pH-Werte zwischen 3,5 und 5,5 während der Sterilisation konnte so eine deutliche Reduktion der Iodidbildung während der Herstellung erreicht werden. Eine 20 mM Trometamol-Lösung wird mit 0,1 N-HCl auf pH 7,5 eingestellt und der pH in Abhängigkeit von der Temperatur mit einem pH-Meter 761 der Fa. Knick mit und ohne Temperaturkorrektur bzw. -kompensation bestimmt. Die Ergebnisse sind in Figur 1 dargestellt.

Die jeweilige Temperatur der Pufferlösungen wurde mit einem externen Pt-100 Fühler gemessen und die Temperatur am pH-Meter entsprechend korrigiert. Die so erhaltenen Daten sind in den Figuren 2 und 3 dargestellt.

Analog zu Beispiel 1 und 2 hergestellte, gepufferte lopromidlösungen (300 mg Iod/ml) wurden zunächst in 10 ml Injektionsflaschen autoklaviert (20 min, 121°C) und anschließend über drei Monate bei 40°C in einem Klimaschrank gelagert. Hierbei wurden parallel Lösungen eingelagert, die auf einen Ausgangs-pH Wert (vor Sterilisation) von 7,5 bzw. 6,5 eingestellt worden waren. Die so erhaltenen Proben wurden hinsichtlich ihres Gehalts an lodid, Amin (freies aromatisches Amin) sowie Natriumcalciumedetat untersucht.Buffered lopromide solutions (300 mg iodine / ml) prepared analogously to Examples 1 and 2 were first autoclaved in 10 ml injection bottles (20 min, 121 ° C.) and then stored in a climatic cabinet at 40 ° C. for three months. Solutions were stored in parallel which had been adjusted to an initial pH value (before sterilization) of 7.5 or 6.5. The samples thus obtained were examined for their iodide, amine (free aromatic amine) and sodium calcium edetate content.
Es zeigte sich, daß die Stabilität der Röntgenkontrast- mittellösung durch Verwendung von reinem Trometamol Puffer im pH-Bereich < 6, 5 bereits verbessert war. Bei Verwendung erfindungsgemäßer Puffer konnte jedoch eine weitere Stabilisierung erreicht werden. Dies gilt insbesonders für die Kombination Trometamol mit Bernsteinsäure.

1.
