Description
PTH-Varianten kodierende DNA-Sequenzen, PTH-Varianten,
Expressionsvektor, bakterieller Wirt, Verwendung und therapeutische Zusammensetzung
Parathormon (PTH) reguliert den Calciumspiegel des Blutplasmas, indem es u. a. auf- und abbauende Wirkungen auf das Skelett entfaltet, aber auch die Calciumrückresorption durch die Niere verstärkt. Es gibt Anhaltspunkte dafür, daß der knochenaufbauende Effekt des PTH zur klinischen Behandlung von Osteoporose ausgenutzt werden kann; vgl. J. Bone Minera l Res., 1 (1987) 377-382.
Es gibt zudem Hinweise, daß die multiplen Funktionen des PTH von verschiedenen Regionen des Hormonmoleküls ausgeübt werden. So wurde beispielsweise gefunden, daß die N-terminalen 34 Aminosäuren die Bindung des Hormons an seinen Nierenzellen-Rezeptor mit nachfolgender Stimulierung der Adenylatoyclase vermitteln (vgl. Adv. Prot. Chem., 32 (1982) 323-395), daß jedoch ein mitogener Effekt von der mitt-regionalen Sequenz 28-48 ausgeht (vgl. DE-A-37 25 319.0), der nicht über die Adenylatcyclase, sondern möglicherweise über Proteinkinase C vermittelt wird. Es erscheint somit reizvoll, durch gezielte Variation der Aminosäuresequenz des PTH oder von PTH-Fragmenten einzelne Hormoneffekte zu modulieren und so eventuell die katabole Wirkung des Hormons zu reduzieren oder seine anabole Wirkung zu verstärken.
Zum Stand der Technik sei auch verwiesen auf US-PSen
4 086 196 und 4 423 037 sowie John et al. in Adv. Prot.
Chem., 32 (1982) 323.
Dazu wird erfindungsgemäß eine DNA-Sequenz vorgesehen, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 1 bis 27 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind, wobei
- ggf. N-terminal (Position +1) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. C-terminal (Position +84) um eine oder mehrere
Aminosäuren verkürzt ist, wobei die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
Gemäß einer weiteren Ausführungsform wird eine DNA-Sequenz vorgesehen, die eine Variante von PTH(1-84) kodiert, bei der

- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
Gemäß einer weiteren Ausführungsform wird eine DNA-Sequenz vorgesehen, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 32 bis 34 eine oder zwei
Aminosäuren gegen eine andere Aminosäure ausgetauscht sind, wobei an der Position 34 Tyrosin ausgenommen ist und ferner
- ggf. N-terminal (Position +1 ) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
Gemäß einer weiteren Ausführungsform wird eine DNA-Sequenz vorgesehen, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 30 bis 34 eine oder zwei
Aminosäuren gegen eine andere Aminosäure ausgetauscht sind, wobei
- ggf. N-terminal (Position +1) um eins bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
Gemäß einer weiteren Ausführungsform wird eine DNA-Sequenz vorgesehen, die eine Variante von PTH(1-84) kodiert, bei der
(a) der Bereich der Positionen 28 bis 34 und insbesondere 30 bis 34 unverändert ist oder
(b) im Bereich der Positionen 28 bis 34 und insbesondere 30 bis 34 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind und
zusätzlich zu (a) oder (b) außerhalb des Bereichs der
Positionen 28 bis 34 bzw. 30 bis 34 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind und
- ggf. N-terminal (Position +1) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminsäuren verkürzt ist, wobei
die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
Vorzugsweise kodiert die DNA-Sequenz eine PTH-Variante, deren N-Terminus um Prolin verlängert ist. Bevorzugt sind außerdem hPTH-, bPTH-, pPTH-, rPTH- oder cPTH-Varianten.
Für die Numerierung der DNA-Sequenzen gilt, daß 1 das erste Nukleotid des ersten PTH-Kodons bezeichnet, so daß die der folgenden Tabelle 1 zu entnehmenden Zahlen jeweils die Position des ersten gezeigten Nukleotids in Bezug zum ersten Nukleotid des ersten PTH-Kodons bezeichnen.
Gemäß einer weiteren Ausführungsform betrifft die Erfindung außerdem PTH-Varianten, die von einer der vorstehend beschriebenen DNA-Sequenzen kodiert werden, und insbesondere Varianten des PTH vom Menschen (h), vom Rind (b), vom
Schwein (p), von der Ratte (r) und vom Huhn (c).
Die erfindungsgemäßen PTH-Varianten lassen sich formal in folgende vier Gruppen einteilen.
Gruppe 1: konservative Aminosäureaustausche in den Positionen 27 und 29, die die Stimulierung sowohl der Adenylatcyclase als auch der DNA-Synthese erhalten ("funktionelle PTH-Äquivalente"); Beispiele: Q29N; K27R.
Gruppe 2: nicht-konservative Aminosäureaustausche (Variationen unter Ladungsumkehrung oder -löschung, Veränderung von Wasserstoffbrücken, Umkehrung der Hydrophobizität) in den Bereichen 29 bis 31 bzw. 33 bis 34 sowie Austausche in Position 32, die zu einer signifikanten Reduktion der Stimulierung der DNA-Synthese und zu einem weitgehenden Erhalt des Effekts auf die Adenylatcyclase führen; Beispiele: Q29A, H32L, H32K, H32R.
Gruppe 3: nicht-konservative Aminosäureaustausche (wie vorstehend definiert) im Bereich 1 bis 27, die zu einer signifikanten Reduktion der Adenylatcyclase-Stimulation führen und die DNA-Synthese zum Teil steigern oder unbeeinflußt lassen. Beispiele für DNA-Synthesesteigerung: E4R, E4R/R20E, K13P, K27L; Beispiel für unbeeinflußte DNA-Synthese: R20E.
Gruppe 4: nicht-konservative Aminosäureaustausche im Bereich 1 bis 27, die die Stimulierung der Adenylatcyclase erhalten, die der DNA-Synthese jedoch reduzieren; Beispiel: L11K.
Schließlich betrifft die Erfindung Hybridpeptide, die eine erfindungsgemäße PTH-Variante umfassen, Expressionsvektoren zur Expression der erfindungsgemäßen Proteine, bakterielle Wirte, wie E. coli, für die Expressionsvektoren, die Verwendung der DNA-Sequenzen zur Herstellung der erfindungsgemäßen Peptide sowie therapeutische Zusammensetzungen, die diese Peptide enthalten. Relevanter Stand der Technik sind DE-A-37 25 319.0 und DE-A-37 25 320.4.
Nachstehend wird die Erfindung durch ein Beispiel und zwei Tabellen näher erläutert.
Beispiel
Es wurde von dem Expressionsplasmid pEX-pPTH der DE-A-37 25 319.0 ausgegangen. Ein 123 Basenpaare langes BamHI/SstI-Fragment dieses Plasmids wurde in den Phagenvektor Ml3mpl8 umkloniert. Damit wurden mit Hilfe synthetischer Oligonukleotide nach der Gapped-Duplex-Methode (Methods Enzymol., 100 (1983) 468-500) die in Tabelle 1 aufgelisteten Mutanten hergestel lt.
Nach Rückklonieren der mutierten PTH-Teilgene in den Expressionsvektor wurden die entsprechenden Varianten Cro-ß-Galaktosidase-hPTH-Fusionsproteine exprimiert, gereinigt und gespalten; die auf diese Weise freigesetzten PTH-Varianten wurden gemäß der DE-A-37 25 319.0 und DE-A-37 25 320.4 gereinigt. Die Reinigung über Carboxymethylcellulose wurde batchweise durchgeführt. Eine nachfolgende Reinigung über Umkehrphasen-HPLC ergab homogenes Material.
Die so erhaltenen PTH-Varianten wurden auf ihre Fähigkeit getestet, in einem In-Vitro-Test nach Mohr und Hesch
(Biochem. J., 188 (1980) 649-656) die Adenylatcyclase in isolierten Membranen von Nebennierenrinden des Schweins zu stimulieren.
Ferner wurden sie auf ihre Fähigkeit untersucht, gemäß der DE-A-37 25 319.0 und DE-A-37 25 320.4 die DNA-Synthese embryonaler Hühner-Chondrocyten zu steigern.
Die erhaltenen Ergebnisse sind in Tabelle 2 zusammengefaßt.

Die Numerierung an den DNA-Sequenzen gibt die Position des ersten gezeigten Nukleotids in Bezug zu dem ersten Nukleotid des ersten PTH- Kodons (= 1) an. Die unterstrichenen Basen sind gegenüber dem rekombinanten Wildtyp verändert. Die Positionsnummern der ausgetauschten
Aminosäuren beziehen sich auf die erste Aminosäure des humanen Parathormons (Ser(1)).

Alle PTH-Varianten sind, wie der Wildtyp, N-terminal um einen Prolinrest verlängert. Die Aktivitäten beziehen sich ausnahmslos auf dieses rekombinante hPTH und geben die Fähigkeit der betreffenden Variante zur Stimulation entweder der Cyclase oder der DNA-Synthese über das Grundniveau an.
Claims
1. DNA-Sequenz, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 1 bis 27 eine oder zwei Aminosäure gegen eine andere Aminosäure ausgetauscht sind, wobei
- ggf. N-terminal (Position +1) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei
- die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
2. DNA-Sequenz, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 28 bis 31 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind, wobei
- ggf. N-terminal (Position +1) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei
- die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH , pPTH, rPTH, cPTH oder hPTH auftritt.
3. DNA-Sequenz, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 32 bis 34 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind, wobei an der Position 34 Tyrosin ausgenommen ist und ferner
- ggf. N-terminal (Position +1) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei
- die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
4. DNA-Sequenz, die eine Variante von PTH(1-84) kodiert, bei der
- im Bereich der Positionen 30 bis 34 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind, wobei
- ggf. N-terminal (Position +1) um eins bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist, wobei
- die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
5. DNA-Sequenz, die eine Variante von PTH(1-84) kodiert, bei der
(a) der Bereich der Positionen 28 bis 34 und insbesondere 30 bis 34 unverändert ist oder
(b) im Bereich der Positionen 28 bis 34 und insbesondere 30 bis 34 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind und
zusätzlich zu (a) oder (b) außerhalb des Bereichs der Positionen 28 bis 34 bzw. 30 bis 34 eine oder zwei Aminosäuren gegen eine andere Aminosäure ausgetauscht sind und - ggf. N-terminal (Position +1) um eine bis zwei Aminosäuren verlängert ist und ggf. zusätzlich C-terminal (Position +84) um eine oder mehrere Aminosäuren verkürzt ist oder
- ggf. N-terminal (Position +1) und/oder C-terminal (Position +84) um eine oder mehrere Aminsäuren verkürzt ist, wobei
- die Gesamtheit der von der DNA-Sequenz kodierten Aminosäuren nicht in einer Reihenfolge angeordnet ist, die in bPTH, pPTH, rPTH, cPTH oder hPTH auftritt.
6. DNA-Sequenz nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, daß sie eine PTH-Variante kodiert, die N-terminal um Prolin verlängert ist.
7. DNA-Sequenz nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, daß sie eine hPTH-Variante, bPTH-Variante, pPTH-Variante, rPTH-Variante oder cPTH-Variante kodiert.
8. Peptid, dadurch gekennzeichnet, daß es durch eine DNA-Sequenz gemäß einem der vorhergehenden Ansprüche kodiert wird.
9. Peptid, dadurch gekennzeichnet, daß es als Hybridpeptid die Aminosäuresequenz eines Peptids gemäß Anspruch 8 umfaßt.
10. Expressionsvektoren mit den DNA-Sequenzen gemäß einem der vorhergehenden Ansprüche zur Expression eines Peptids gemäß Anspruch 8 oder 9 in einem bakteriellen Wirt wie Escherichia coli.
11. Bakterieller Wirt, insbesondere Escherichia coli, mit einem Expressionsvektor gemäß Anspruch 10.
12. Verwendung einer DNA-Sequenz gemäß einem der vorhergehenden Ansprüche, gegebenenfalls in Form eines Expressionsvektors gemäß Anspruch 10 oder eines bakteriellen Wirts wie Escherichia coli gemäß Anspruch 11 zur Herstellung eines Peptids gemäß Anspruch 8 oder 9.
13. Therapeutische Zusammensetzung, bestehend aus oder mit einem Gehalt an einem Peptid gemäß Anspruch 9 oder einem Peptid gemäß Anspruch 9, wobei Tyrosin an der Position 34 nicht ausgenommen ist, ggf. neben übl ichen Trägern und Hilfsmitteln.
