Definitions
- the present invention relates to a process for photocleavage of nucleic acids using dications 2,7-diazapyrenium DAP 2+ or their dimeric bis-DAP 4+ species.
- 2,7-diazapyrenium dications coupled to oligonucleotides can constitute photoexcision reagents specific for nucleic acids.
- metal complexes which have been used, there may be mentioned in particular the compounds carrying EDTA-Fe (II) groups and the 1,10-phenanthroline-copper complexes.
- HERTZBERG et al. [J. Am. Chem. Soc. 1982 104 p. 313-315] recommends the use of (methidiumpropyl-EDTA) -Fe (II) as a cofactor in cutting DNA for a number of anti-tumor antibiotics.
- Methyl viologene which is 1,1'-dimethyl-2 + 4,4'-bipyridinium chloride, commonly known as MV, can damage DNA under these conditions.
- the present invention relates to a process for the photocleavage of nucleic acids which consists in bringing a nucleic acid into contact with a solution of a 2,7-diazapyrene derivative as defined below and in subjecting the reaction mixture obtained to irradiation with visible light.
- the hydrocarbon chains which constitute the radical Z can contain 2 or more carbon atoms and be interrupted by one or more heteroatoms chosen from oxygen, sulfur or nitrogen atoms.
- divalent radicals Z are the radicals of formulas III to V below: or
- the aliphatic hydrocarbon radicals preferably have 1 to 10 carbon atoms. Linear or branched alkyl groups having 1 to 6 carbon atoms are particularly preferred.
- diazopyrenic macrocycles which correspond to the general formula (III): in which p is an integer between 1 and 15, preferably equal to 8.
- diazapyrenic derivatives of formula (I) and (II) can all be obtained from 2,7-diaz a pyrene or DAP by one of the methods below.
- the diazapyrenic derivatives of formula (I) will be designated by the expression "DAP 2+ derivatives” and those of formula (II) by the expression "bis-DAP 4+ derivatives”.
- X 1 and X 2 identical or different, have the same meaning as X.
- This process essentially comprises two stages of N-substitution of DAP, respectively by the radicals R and R ', the compound obtained can then be optionally transformed, by ion exchange, into the corresponding compound in which X 1 and X 2 are identical . It will be noted that when R and R ′ are identical, the two substitution steps are carried out simultaneously. These steps are carried out in an appropriate solvent, for example dimethylformamide (DMF), chloroform or acetonitrile.
- DMF dimethylformamide
- chloroform chloroform
- acetonitrile acetonitrile
- Ts can be replaced by another anion.
- This process is suitable for obtaining DAP 2+ derivatives in which the radical R is an aliphatic hydrocarbon radical substituted by a hydroxy group, in particular a hydroxyalkyl group and R 'is an alkyl group, for example methyl.
- This process consists in first manufacturing a 2-dinitrophenyl-2,7-diazapyrenium salt, for example tosylate, as a synthesis intermediary by reacting DAP with a 2,4-dinitrophenyl salt, such as tosylate.
- the second step is an N-substitution step with a halide of formula R'X as in method A.
- a suitable solvent for example a chloroform / methanol mixture at reflux
- This process is suitable for obtaining DAP 2+ derivatives in which R 'is an alkyl group, for example methyl and R is an aliphatic hydrocarbon residue substituted with an amino group.
- R ' is an alkyl group, for example methyl and R is an aliphatic hydrocarbon residue substituted with an amino group.
- This process is similar to process B above with the only difference that a diamine is used as an amine, one of the amine functions of which is protected, which is then deprotected before the second substitution step.
- 2-dinitrophenyl-2,7-diazapyrenium tosylate is a new compound useful as a synthetic intermediate; it constitutes another object of the invention.
- This process is suitable for obtaining the DAP 2+ derivative in which R and R 'are identical and represent a radical R 1 Y - as defined above;
- This process consists in reacting together DAP partially dissolved in anhydrous acetone and propansultone at the reflux temperature.
- This process therefore consists in reacting DAP with aziridine in the presence of collidine and collidinium chloride.
- the process for obtaining these bis-DAP 4+ derivatives essentially consists in reacting two molecules of DAP with a molecule of a dihalide of formula X 1 - Z -X 1 in which X 1 , X 2 and Z are such that defined above and then proceed to N-substitution of the remaining nitrogen in the DAP molecules.
- Each step is advantageously carried out in acetonitrile at the reflux temperature.
- the process for obtaining macrocycles of formula III also called "bis-DAP 4+ macrocycles” consists in reacting 1,4,5,8-naphthalene tetracarboxylic anhydride of formula VI with a diamine of formula NH 2 - (CH 2 ) p -NH 2 in dimethylformamide and in the presence of acetic acid.
- the macrocycle thus formed is then reduced in the presence of lithium aluminum hydride and aluminum chloride to form the compound of formula VII, which is then transformed into the compound of formula III.
- the first step of this process is a modified version of the process described by JW Verhoeven in the Collection Journal of the Royal Netherlands Chemical Society, 95/4, April 1976 p. 89.
- the diazapyrenic derivatives according to the invention are fluorescent compounds.
- the diazapyrene (DAP) used as starting material in the above syntheses can be obtained from the 1,4,5,8-naphthalene tetracarboxylic anhydride of formula VI commercially available according to the reaction scheme below:
- 1,4,6,8-naphthalenetetracarboxylic anhydride of formula VI. is first transformed into the corresponding bis (N-methylimide) of formula X by reaction with methylamine at room temperature.
- the imide X. is then treated with a LiAlH 4 / AlCl 3 mixture in tetrahydrofuran at reflux to form the compound of formula XI, which is then reacted for approximately 4 hours in the presence of selenium at a temperature between 265 and 300 ° C to obtain the DAP.
- the diazapyrenic derivatives of formulas (I) or (II) comprising one or two dications2,7-diazapyrenium have a planar structure. These compounds, thanks to this flat structure, interact with the nucleotides of DNA or RNA in the same way as intercalation drugs, such as actinomycin, bleomycin.
- intercalation drugs such as actinomycin, bleomycin.
- the derivatives according to the invention can cut DNA or RNA into several strands.
- the derivatives of formula (II) or bis-DAP 4+ derivatives are more reactive than the derivatives of formula (I) or DAP 2+ derivatives because of the presence in their molecule of two cations 2, 7-diazapyrenium. They also have greater selectivity and stability during photocleavage.
- the derivatives of formula (I) in which one of the substituents R or R 'is an aliphatic hydrocarbon radical substituted by a hydroxy or amino group can be coupled by covalent bond to an oligonucleotide or an oligodeoxynucleotide comprising a chain natural nucleotides, possibly modified or synthesized.
- This coupling can be achieved by methods known to those skilled in the art.
- the synthesis methods described in patent FR 83 01 223 cited in the present application may be used for reference. These methods include inter alia coupling the hydroxyl of the DAP 2+ derivative with a 3′-phosphodiester or 5′-phosphodiester oligonucleotide, with a 3′-oligonucleotide, 5′- b i s (phosphodiester), or with an oligodeoxynucleotide 2 '-phosphodiester, the oligonucleotides or oligodeoxynucleotides being properly protected.
- the oligonucleotide or oligodeoxynucleotide chain of the coupling product can selectively bind to any complementary sequence. Thanks to the presence of at least one 2,7-diazapyrenium dication in this coupling product, it is easy to selectively cut DNA or RNA at particular sites, the position of which is determined by the oligonucleotide that binds
- the oligonucleotide -DAP 2+ coupling product can subsequently be coupled with another reagent, which can be any revealing agent, for example a marker, such as in particular an acridine dye or an intercalating agent, and provide a bifunctional coupling product, one of the functions being photocleavage, the other function being provided by the second reagent.

- the reaction was carried out in the presence or absence of air (oxygen) or EDTA.
- scDNA The supercoiled circular double-stranded DNA, hereinafter called scDNA, was prepared according to known procedures using E. coli bacteria infected with the plasmid pBR 322 and subsequently purified by electroelution so as to be practically free of cut scsc DNA.
- the plasmid pBR322 gives, by incubation with the restriction enzyme ECOR1, a linear plasmid or double-stranded linear DNA which was used as a marker in the electrophoresis analysis carried out after the above photocleavage assay.

- proflavin is an acridine type dye and it was found that proflavin is a reagent for photocleavage of DNA comparable to Bis-MDAP 4+ and that it gives the concentration of 2 10 -4 M an extensive cleavage of DNA into linear DNA. At lower concentrations ranging from 2 10 M to 2 10 -7 M, the efficiency decreases.
- the derivatives of the invention being flat molecules, they interact with nucleotides in DNA or RNA in the same way as intercalation drugs, such as actinomycin, bleomycin, etc. It has also been noted that the fluorescence of the derivatives disappears when there is attachment to the DNA. - bands 2 and 3 are representative of the DNAsc and ssDNA subjected to irradiation in the absence of diazapyrenic derivative; these DNAs were obtained respectively by the action of the enzyme ECoR 1 and of the nucleus ase S 1 in a known manner.
- diazapyrenic derivatives used in the process according to the invention keep their property of photocleavage of DNA even when they are incorporated in micelles or in liposomes.
- Diazapyrenic derivatives containing a long chain hydrocarbon radical are particularly suitable for this type of use, since the hydrocarbon radical allows better fixation on the liposome or the micelle.

Abstract
Description
It also relates to new diazapyrenic derivatives; these are salts which, in solution, release DAP 2+ indications or bis-DAP 4+ tetracations.
Finally, 2,7-diazapyrenium dications coupled to oligonucleotides can constitute photoexcision reagents specific for nucleic acids.
Much research has been undertaken in recent years to find artificial reagents capable of cleaving nucleic acids and more particularly deoxyribonucleic acid or ribonucleic acid, commonly known as DNA and RNA.
On a également utilisé des colorants du type acridine pour la photodestruction de l'ADN.Most of the studies have been done with metal complexes that cleave DNA by an oxygen-dependent redox process or by photo-activation. Acridine dyes have also been used for DNA photodestruction.
Among the metal complexes which have been used, there may be mentioned in particular the compounds carrying EDTA-Fe (II) groups and the 1,10-phenanthroline-copper complexes.
For example, BOUTORIN et al. in FEBS LETTERS vol. 172, No. 1 June 1984 describes reagents carrying EDTA-Fe (II) groups which are suitable for the direct cleavage of single-stranded nucleic acids.
230, 1985 p. 679-681 uses EDTA-Fe (II) to cut DNA.
HERTZBERG et al. [J. Am. Chem. Chem. Soc. 1982 104 p. 313-315] recommends the use of (methidiumpropyl-EDTA) -Fe (II) as a cofactor in cutting DNA for a number of anti-tumor antibiotics.
Chem. Soc. Vol. 103 No. 12, 1981 et par R. FAGGIANI et al. dans J. Am. Chem. Soc. Vol. 102 p.5419-5421. 1980.The use of the 1,10-phenanthroline-copper complex for the cleavage of DNA is described in particular by REICH et al. in J. Am. Chem. Soc. Flight. 103 No. 12, 1981 and by R. FAGGIANI et al. in J. Am. Chem. Soc. Flight. 102 p.5419-5421. 1980.
On the other hand, it is known that dyes of the acridine type damage DNA. For example FREIFELDER et al. LBiophy- sical Journal Vol. 1, 1961 / have shown that DNA irradiated with visible light in the presence of acridine orange depolymerizes and cuts.
It should also be noted that the 4,4'-bipyridinium ions, and in particular methyl viologene, have been used in various redox and photo-redox processes [AMQUYAL et al. Israel Journal of Chemistry 22, 1982 p. 117-123]. The latter can only be carried out in the presence of a photosensitizer or under ultraviolet irradiation.
Methyl viologene, which is 1,1'-dimethyl-2 + 4,4'-bipyridinium chloride, commonly known as MV, can damage DNA under these conditions.
It has now been found that it is possible to use diazapyrenic derivatives and more particularly the 2,7-diazapyrenium cations for the photocleavage of nucleic acids in visible light without the need to use photosensitizers.
Thus the present invention relates to a process for the photocleavage of nucleic acids which consists in bringing a nucleic acid into contact with a solution of a 2,7-diazapyrene derivative as defined below and in subjecting the reaction mixture obtained to irradiation with visible light.
- - l'hydrogène ;
- R and R ', identical or different, represent:
- - hydrogen;
- - an aliphatic hydrocarbon radical, saturated or unsaturated, linear or branched, optionally interrupted by one or more heteroatoms, such as for example oxygen, sulfur or nitrogen, said hydrocarbon radical possibly being optionally substituted by hydroxy groups , amino, thio, by an aryl group, by a heterocyclic group; or
- R and R 'are identical and represent a group of formula R 1 Y in which R 1 is the group - (CH 2 ) 3 - and Y is the sulfonato ion SO 3 - ; or a group of formula
- By "aliphatic hydrocarbon radical optionally interrupted by one or more heteroatoms" is also meant in this description, aliphatic hydrocarbon groups containing the corresponding onium cations, in particular aliphatic hydrocarbon groups containing one or more quaternized nitrogen atoms and in particular the group of formula
Chem. 1980 N°19 (1968)7The compounds of formula I above in which R and R ′ are identical and represent hydrogen or a methyl group, are known compounds and described by Hunig et al. [Angew. Chem. 1980 N ° 19 (1968) 7
- - R, R 'and X are as defined above and ②st is a divalent radical which represents a straight or branched, saturated or unsaturated aliphatic hydrocarbon chain, optionally interrupted by one or more heteroatoms and / or possibly interrupted by one or more groups divalent aromatic or heterocyclic.
The hydrocarbon chains which constitute the radical Ⓩ can contain 2 or more carbon atoms and be interrupted by one or more heteroatoms chosen from oxygen, sulfur or nitrogen atoms.
In formulas (I) and (II) above, the aliphatic hydrocarbon radicals preferably have 1 to 10 carbon atoms. Linear or branched alkyl groups having 1 to 6 carbon atoms are particularly preferred.
The diazapyrenic derivatives of formula (I) and (II) can all be obtained from 2,7-diaz a pyrene or DAP by one of the methods below. For convenience, in the remainder of this description, the diazapyrenic derivatives of formula (I) will be designated by the expression "DAP 2+ derivatives" and those of formula (II) by the expression "bis-DAP 4+ derivatives".
X1 et X2, identiques ou différents, ont la même signification que X.X 1 and X 2 , identical or different, have the same meaning as X.
This process essentially comprises two stages of N-substitution of DAP, respectively by the radicals R and R ', the compound obtained can then be optionally transformed, by ion exchange, into the corresponding compound in which X 1 and X 2 are identical . It will be noted that when R and R ′ are identical, the two substitution steps are carried out simultaneously. These steps are carried out in an appropriate solvent, for example dimethylformamide (DMF), chloroform or acetonitrile. DMF is suitable when it is desired to obtain the two substitutions simultaneously (R = R '= CH 3 for example), the reaction being carried out at room temperature, while chloroform should be used for the first substitution step and l 'acetonitrile for the second (R ≠ R'), in which case it is advantageous to operate at room temperature for the first substitution step and at reflux for the second.
Note that Ts can be replaced by another anion.
A.This process is suitable for obtaining DAP 2+ derivatives in which the radical R is an aliphatic hydrocarbon radical substituted by a hydroxy group, in particular a hydroxyalkyl group and R 'is an alkyl group, for example methyl. This process consists in first manufacturing a 2-dinitrophenyl-2,7-diazapyrenium salt, for example tosylate, as a synthesis intermediary by reacting DAP with a 2,4-dinitrophenyl salt, such as tosylate. , in a suitable solvent, for example a chloroform / methanol mixture at reflux, then reacting this intermediate with an amine substituted by a radical R; the second step is an N-substitution step with a halide of formula R'X as in method A.
Ce procédé convient pour l'obtention du dérivé DAP2+ dans lequel R et R' sont identiques et représentent un radical R1Y- tel que défini précédemment ;
This process consists in reacting together DAP partially dissolved in anhydrous acetone and propansultone at the reflux temperature.
This process therefore consists in reacting DAP with aziridine in the presence of collidine and collidinium chloride.
The process for obtaining these bis-DAP 4+ derivatives essentially consists in reacting two molecules of DAP with a molecule of a dihalide of formula X 1 - Ⓩ -X 1 in which X 1 , X 2 and Ⓩ are such that defined above and then proceed to N-substitution of the remaining nitrogen in the DAP molecules. Each step is advantageously carried out in acetonitrile at the reflux temperature.
Verhoeven dans Recueil Journal of the Royal Nether- lands Chemical Society, 95/4, avril 1976 p. 89.The process for obtaining macrocycles of formula III also called "bis-DAP 4+ macrocycles" consists in reacting 1,4,5,8-naphthalene tetracarboxylic anhydride of formula VI with a diamine of formula NH 2 - (CH 2 ) p -NH 2 in dimethylformamide and in the presence of acetic acid. The macrocycle thus formed is then reduced in the presence of lithium aluminum hydride and aluminum chloride to form the compound of formula VII, which is then transformed into the compound of formula III. The first step of this process is a modified version of the process described by JW Verhoeven in the Collection Journal of the Royal Netherlands Chemical Society, 95/4, April 1976 p. 89.
Chem. 1973, 339.The second and third steps of this process are an adaptation of the process described by HUNIG et al. Ann. Chem. 1973, 339.
The diazapyrenic derivatives according to the invention are fluorescent compounds.
Chem. 180 (1968) No. 19, 1,4,6,8-naphthalenetetracarboxylic anhydride of formula VI. is first transformed into the corresponding bis (N-methylimide) of formula X by reaction with methylamine at room temperature. The imide X. is then treated with a LiAlH 4 / AlCl 3 mixture in tetrahydrofuran at reflux to form the compound of formula XI, which is then reacted for approximately 4 hours in the presence of selenium at a temperature between 265 and 300 ° C to obtain the DAP.
- 1) les composés de formulé (I) dans laquelle R et R' sont un groupe méthyle ou dérivé MDAP2+;
- 2) les composés de formule (I) dans laquelle l'un des substituants R ou R' est un alkyle inférieur, par exemple le méthyle et l'autre est un groupe alkyle substitué par un groupe hydroxy,thio ou un groupe amino, à savoir les groupes de formules -(CH2)n -OH, -(CH2)n -SH,-(CH2)n -NH2 dans lesquels n est de nréférence commris entre 1 et 10 ;
- 1) the compounds of formula (I) in which R and R 'are a methyl group or MDAP 2+ derivative;
- 2) the compounds of formula (I) in which one of the substituents R or R ′ is a lower alkyl, for example methyl and the other is an alkyl group substituted by a hydroxy group, thio or an am ino group, namely the groups of formulas - (CH 2 ) n -OH, - (CH 2 ) n -SH, - (CH 2 ) n -NH 2 in which n is of reference committed between 1 and 10;
- 3) the compounds of formula (II) in which Ⓩ corresponds to one of formulas III to V and R and R 'are a lower alkyl group, for example methyl.
The diazapyrenic derivatives of formulas (I) or (II) comprising one or two dications2,7-diazapyrenium have a planar structure. These compounds, thanks to this flat structure, interact with the nucleotides of DNA or RNA in the same way as intercalation drugs, such as actinomycin, bleomycin. In addition, under the action of irradiation with visible light, the derivatives according to the invention can cut DNA or RNA into several strands.
It should be specified that the derivatives of formula (II) or bis-DAP 4+ derivatives are more reactive than the derivatives of formula (I) or DAP 2+ derivatives because of the presence in their molecule of two
On the other hand, the derivatives of formula (I) in which one of the substituents R or R 'is an aliphatic hydrocarbon radical substituted by a hydroxy or amino group can be coupled by covalent bond to an oligonucleotide or an oligodeoxynucleotide comprising a chain natural nucleotides, possibly modified or synthesized.
This coupling can be achieved by methods known to those skilled in the art. For example, the synthesis methods described in patent FR 83 01 223 cited in the present application may be used for reference. So can get oligonucleotide coupling products (or oligodeoxynucleotide) -DAP 2+ or DAP 2+ -oligonucleotide (or oligodeoxynucleotide) -DAP 2+ which can constitute very selective photoexcision reagents.
Thanks to the presence of at least one 2,7-diazapyrenium dication in this coupling product, it is easy to selectively cut DNA or RNA at particular sites, the position of which is determined by the oligonucleotide that binds
It will be noted that the oligonucleotide -DAP 2+ coupling product can subsequently be coupled with another reagent, which can be any revealing agent, for example a marker, such as in particular an acridine dye or an intercalating agent, and provide a bifunctional coupling product, one of the functions being photocleavage, the other function being provided by the second reagent.
The process of the invention will now be described with reference to MDAP 2+ or its bis-MDAP 4+ dimeric species for simple convenience, without however limiting it to these derivatives only.
Tests carried out on DNA under the conditions indicated below have shown that the dication MDAP 2+ or its dimeric species bis-MDAP 4+ cut the DNA under irradiation with visible light.
The tests were carried out by illumination for one hour at 3+ 1 ° C with visible light (250 W projector-slit fitted with an adequate filter to allow only visible light λ> 395 nm) to pass through. aqueous solution containing circular, double-stranded, supercoiled DNA (2.6 10 -8 M), MDAp2 + or bis-MDAP 4+ chloride (2.6 10 -4 M) in tris buffer at pH 7.8 .
La réaction a été effectuée en présence ou en l'absence d'air (oxygène) ou d'EDTA.The reaction was carried out in the presence or absence of air (oxygen) or EDTA.
The supercoiled circular double-stranded DNA, hereinafter called scDNA, was prepared according to known procedures using E. coli bacteria infected with the plasmid pBR 322 and subsequently purified by electroelution so as to be practically free of cut scsc DNA.
The plasmid pBR322 gives, by incubation with the restriction enzyme ECOR1, a linear plasmid or double-stranded linear DNA which was used as a marker in the electrophoresis analysis carried out after the above photocleavage assay.
By way of comparison, the same test was carried out by replacing the derivative of the invention with methyl-viologene MV 2+ . Was also subjected to the same treatment (illumination with visible light, under the conditions above) a solution of DNAsc alone, or a solution of DNAsc containing oxygen or a solution of DNAsc containing oxygen and EDTA, as controls.
- I lfADNsc, II l'ADNsc coupé et III l'ADN linéaire ;
- I DNAsc, II cut DNA and III linear DNA;
les abbréviations utilisées désignent respectivement :
- M = marqueur
- C = contrôle
- V = lumière visible
- 0 = oxygène
- A = EDTA.
- M = marker
- C = control
- V = visible light
- 0 = oxygen
- A = EDTA.
With lower concentrations of MDAP 2+, less cleavage occurs, transformation into the cut scDNA form being almost complete at the concentration of 2 10 M and only weak at 2 10 -6 M and below.
2) On notera qu'il n'y a pas eu de clivage significatif de l'ADNsc en l'absence de lumière.2) It will be noted that there was no significant cleavage of the scDNA in the absence of light.
3) L'élimination de l'oxygène par dégazage par congélation- décongélation diminue un peu l'efficacité du photoclivage.3) The elimination of oxygen by degassing by freezing-thawing slightly reduces the efficiency of photocleavage.
4) L'addition d'EDTA n'affecte pas la réaction en présence d'oxygène, mais semble la diminuer lorsque celle-ci est réalisée en l'absence d'oxygène.4) The addition of EDTA does not affect the reaction in the presence of oxygen, but seems to decrease it when it is carried out in the absence of oxygen.
It will be noted that the migration of the DNAsc is somewhat delayed by the photocleavage reagent used, which suggests that it is intercalated in the DNAsc.
-
Under the same conditions, bis-MDAP 4+ seems to be more effective than MDAP 2+, giving small DNA fragments by multiple cuts of the two strands (
bands 14 to 18). The cleavage reaction is so effective that the exclusion of light becomes difficult. It is nevertheless possible to observe that the migration of DNAsc is significantly delayed, which indicates a strong and probably multiple interaction of Bis-MDAP 4+ with the plasmid pBR322.
- 3) Etant donné l'efficacité de la réaction,il n'est pas possible de noter un effet quelconque de l'oxygène ou de l'EDTA.3) Given the efficiency of the reaction, it is not possible to note any effect of oxygen or EDTA.
The effect of bis-MDAP 4+ decreases at lower concentrations, the cleavage being still complete at 10 -5 M but only weak at a lower concentration.
These tests show that, under the same conditions, there is no, or practically no, cleavage with MV 2+. On the other hand, there is no reaction in the absence of photocleavage reagent.
Since other tests have shown that MDAP 2+ photo-oxidizes different substrates under irradiation with visible light, it can be considered, without wishing to be limited to any theory, that the cleavage reaction takes place by intermediate of a local photo-oxidation at the site of interaction by the excited state of the MDAP species. The electron donor may be ribose, since it has been found that ribose itself photo-reduces MDAP 2+ .
The delaying effect of EDTA in the absence of oxygen would indicate that the oxidation of the external donor competes with the oxidation of the internal donor.
Biophys. Acta 1973,314 , 372].The effect produced by the elimination of oxygen, which is small but not negligible, indicates that the cleavage reaction can also involve the reaction system as with the reduced photosensitive metal complexes. The DAP cation produced initially by photoreduction could then react with oxygen by generating superoxides, as happens with MV + itself (Biochim. Biophys. Acta 1973,314, 372].
Other tests were carried out under the same conditions as above with proflavin, which is an acridine type dye and it was found that proflavin is a reagent for photocleavage of DNA comparable to Bis-
It is believed that the photocleavage process with proflavin involves photo-oxidation of DNA at the intercalation site with generation of a reduced dye. Dyes of the acridine type are indeed well known for undergoing photoreduction with an oxidation of the substrate.
The derivatives of the invention, being flat molecules, they interact with nucleotides in DNA or RNA in the same way as intercalation drugs, such as actinomycin, bleomycin, etc. It has also been noted that the fluorescence of the derivatives disappears when there is attachment to the DNA. It is therefore possible to follow the evolution of the neaction of photocleavage by fluorescence.
Other tests have been carried out with single-stranded DNA or ssDNA and are reported below.
The solutions of MDAP 2+ and bis-MDAP4 + at concentrations up to 4.10 M were prepared with twice distilled water and adjusted to pH 7.6 with hydrochloric acid.
Circular scDNA and circular ssDNA were obtained respectively from plasmids pBR 322 and M13mp19 and purified by standard electroelution methods.
7,6tampon Tris pH
- - the diazapyrenic derivative MDAP 2+ or bis-MDAP 4+
- - DNAs or ADHsc
- - Tris pH 7.6 buffer
en des quantités appropriées pour obtenir des concentrations en dérivés diazapyréniques de 5.10-6M et 10 M. Chaque tube a été maintenu sous une pression constante d'oxygène en utilisant des moyens connus et soumis à une irradiation à la lumière visible (projecteur 250 W; λ>395 nm) pendant 1/2 heure, les tubes étant maintenus à 3°C dans un bain d'eau glacée.in quantities suitable for obtaining concentrations of diazapyrenic derivatives of 5.10 -6 M and 10 M. Each tube was maintained under constant oxygen pressure using known means and subjected to irradiation with visible light (250 W projector ; λ> 395 nm) for 1/2 hour, the tubes being maintained at 3 ° C in an ice water bath.
This photograph shows that the circular DNA and ssNA are cleaved in the presence of the diazapyrenic derivatives of the invention.
It will be noted that the diazapyrenic derivatives used in the process according to the invention keep their property of photocleavage of DNA even when they are incorporated in micelles or in liposomes. We can therefore use micelles and liposomes as vehicles for diazapyrenic derivatives to transport them to the DNA to be cut.
Diazapyrenic derivatives containing a long chain hydrocarbon radical are particularly suitable for this type of use, since the hydrocarbon radical allows better fixation on the liposome or the micelle.
The invention will now be described in more detail by the following examples.

The yellow precipitate which formed was filtered off, washed with chloroform and dried in vacuo.
This product was suspended (0.3 g) in 50 ml of acetonitrile and 0.5 ml of methyl iodide, and heated to reflux for 4 hours with stirring. A red-brown solid insoluble in acetonitrile was obtained. This solid was filtered, washed with chloroform and dried in vacuo.
An ion exchange was then carried out by suspending the disel thus obtained (iodide / bromide) with silver chloride in water and stirring at room temperature for 24 hours. The solid formed was separated by filtration and washed with hot water. The raw product (yellow solid, very soluble in water) was recrystallized several times from methanol or a methanol / ether mixture.
According to another procedure, the methylation step can be carried out by mixing the product of the above formula with methyl bromide at 40-50 ° C in closed bottles for two to three days. A yellow product soluble in water is then obtained.

The amine obtained above was dissolved in 80 ml of hot CH 3 COOH (about 50 ° C) and the solution was filtered, then heated to reflux. 0.93 g (0.0053 mole) of NBS was added thereto. A solid reddish brown instantly precipitated.
The mixture was heated at reflux for 30 minutes and the solvent was evaporated in vacuo. The obtained crude product (brown oil) was mixed with a large volume "of water (about 2 liters), heated for 30 minutes at about 80 ° C and filtered. The filtrate (pale orange solution) was passed through an Amberlite DG-50H column (in acid form) and the column was washed well with water.
The desired product was removed from the column with aqueous hydrochloric acid (10%, then 20% and finally 38%) and evaporated in vacuo. It was then dissolved in a small volume of water, precipitated by addition of acetone, filtered and dried under vacuum.
