Abstract

Process for encapsulating solid or liq. lipophilic agents (I) in a hydrating medium in phospholipid liposomes comprises mixing 1/2 - 3/4 of the amount of the hydration medium with the phospholipid (mixt.) with stirring to disperse the phospholipid; raising the temp. to above the phase change temp. of the soln. where the (I) is added to the dispersion, and the remaining hydration medium is added; and continuing to stir the mixt. at this temp. until no more free (I) can be detected in the hydration medium. The process is carried out in an inert atmos. The hydration medium is free of organic solvent and detergent. After the first step is complete, the liposomes are adjusted to the required size. Pref. the phospholipid is phosphatidytcholine, ethanolamine etc. and the active agent is an antibiotic, hormone, tranquiliser etc.
