20 g salzsaures p-Aminobenzylcyanid werden mit 12 g Cyanamid und 8 ecm Wasser im Wasserbad erhitzt. Es bildet sich allmählich eine klare Schmelze, die mit Wasser verdünnt und mit konzentrierter Natronlauge alkalisch gemacht wird. Nach Abkühlen in Eis wird das p-Guanidinoberizylcyanid abgesaugt und mit eiskaltem Wasser öfters gewaschen. Man erhält so die reine Base, die bei 165 bis i66° unter ■ Zersetzung schmilzt. Sie läßt sich aus wenig Wasser Umkristallisieren. Das salpetersaure Salz schmilzt bei 1890 unter Zersetzung. Wird 1 Mol dieser Base mit ι Mol 2-Methyl-4-aminochinolyl-(6)-azid (vgl. Beispiel 9 der Patentanmeldung F 3817 IVb/i2p) in einer alkoholischen. p-Amino-phenylguanidin läßt sich als Carbonat der Zusammensetzung (C7H10N4)2 · H2CO3 leicht gewinnen, und zwar entweder durch Reduktion des p-Nitrophenylguanidins oder durch Verseifung des p-Acetamino-phenylguänidins. Die Herstellung der genannten Nitroverbindung erfolgt zweckmäßig analog der der p-Guanidinobenzoesäure (s. Beispiel 4). Sie schmilzt bei 198 bis 1990 unter Zersetzung (Literaturangabe: i88°). Die Reduktion geschieht am besten katalytisch mit Palladium und Wasserstoff in wäßriger Lösung des essigsauren Salzes. Nach der Reduktion scheidet sich auf Zusatz von überschüssiger gesättigter Pottasche das Carbonat des p-Amino-phenylgüanidins aus, das mit eisgekühltem Wasser gut gewaschen wird. Es zeigt aus wenig Wasser umkristallisiert den Schmelzpunkt 180 bis 181° unter Zersetzung. Überschüssige konzentrierte Natronlauge fällt eine dickölige Base, die beim Verdünnen mit Wasser sofort in Lösung geht. Zur Herstellung aus p-Aminoacetanilid werden 2 Teile des salzsauren Salzes mit einem Gewichtsteil Wasser und einem Gewichtsteil Cyanamid im Wasserbad bis zur Bildung einer klaren, dünnen Schmelze erwärmt, die auf Zusatz von Sodalösung das Carbonat des p-Acetamino-phenylguanidins ausscheidet. Nach Umkristallisieren aus Wasser erhält man ein farbloses Pulver vom Schmelzpunkt 2200 unter heftiger Zersetzung. Beim Erwärmen mit verdünnter Salzsäure im Wasserbad wird die Acetylgruppe abgespalten, und man gewinnt das p-Amino-phenylguanidin wie oben angegeben als Carbonat. 5 Gewichtsteile des Carbonats werden mit 3,5 Gewichtsteilen salzsaurem N1 - [2 - Methyl- 4 - aminochinolyl- (6)] - diamino - cyanurchlorid (vgl. die Patentschrift 606 497, Beispiel 10) in 100 Gewichtsteilen Wasser 3 Stunden unter Rückfluß gekocht. Die heiß filtrierte Lösung scheidet auf Zusatz von verdünnter Salpetersäure beim Stehen ein Nitrat aus, aus dessen wäßriger Lösung Natronlauge eine Base fällt, die sich nach dem Umkristallisieren aus viel Wasser als identisch mit der im Beispiel 8 der Patentanmeldung F38i7lVb/i2p beschriebenen erweist. p-Amino-phenylguanidine can easily be obtained as a carbonate with the composition (C 7 H 10 N 4 ) 2 · H 2 CO 3 , either by reducing the p-nitrophenylguanidine or by saponifying the p-acetaminophenylguanidine. 18.1 g p-Amino-phenylguanidin-carbonat (s. Beispiel 2), gelöst in einer Mischung von 200 ecm Wasser und 22 ecm konzentrierter Schwefelsäure, werden mit einer Lösung von 7 g Natriumnitrit in 30 ecm Wasser in üblicher Weise diazotiert. Zu dieser Diazolösung gibt man dann unter weiterem Rühren und Kühlen langsam eine Lösung von 6,6 g Natriumazid in 20 ecm Wasser. Nach Beendigung der Umsetzung wird die Lösung mit überschüssiger 2 η-Salpetersäure versetzt, worauf sich das fast farblose Nitrat des p-Guanidinophenylazids ausscheidet. Es läßt sich aus Wasser Umkristallisieren und schmilzt bei 200° unter heftiger Zersetzung. Mit Natronlauge wird die Base gebildet, die, aus Wasser umkristallisiert, bei 147 bis 1480 unter Zersetzung schmilzt. Sie ist ein bräunlichgraues Kristallpulver, das sich am Licht gelbbraun färbt. Zu einer Lösung von 2,3 g Natrium in etwa 150 ecm Alkohol gibt man 9,85 g 2-Methyl-4-aminochinolyl-(6)-acetonitril (s. Patentschrift 708 116, Beispiel 1) und danach 11,95 g p-Guanidino-phenylazid-nitrat und kocht das Gemisch dann 1 Stunde unter Rückfluß. Das bereits in der Wärme sich ausscheidende Konden-' sationsprodukt der oben angegebenen Zusammensetzung wird nach Abkühlung abgesaugt, mit Alkohol bis zur Farblosigkeit des Filtrats und danach mehrmals mit warmem Wasser gewaschen. Aus Alkohol umkristallisiert, ist die Verbindung ein sehr feines, farbloses Pulver, das sich bei 245 ° unter langsamem Aufblähen zersetzt. Es löst sich leicht in verdünnter Essigsäure und bildet ein leichtlösliches salzsaures Salz. 16.2 g p-Aminobenzoesäure werden mit 8 ecm Wasser und 9,7 ecm konzentrierter Salzsäure angeteigt. Der nach Zugabe von 11,5 g Cyanamid sich bildende dünne Brei wird darauf vorsichtig im Wasserbad erwärmt. Es bildet sich ganz allmählich unter gelindem Aufsieden eine klare Schmelze. Diese wird unter Erwärmen im Wasserbad mit viel verdünnter Salzsäure versetzt. Wird danach die Lösung abgekühlt, so entsteht ein dicker Brei des Hydrochlorids der p-Guanidinobenzoesäure, der mit verdünnter Salzsäure gewaschen wird. Die Verbindung ist in heißem Wasser leicht, in kaltem schwer löslich. Durch Zusatz von Sodalösung entsteht eine Fällung, die in verdünnter Essigsäure nicht, in Natronlauge leicht löslich ist. Sie läßt sich aus Wasser Umkristallisieren und schmilzt danach bei 2800 unter heftigem Aufschäumen. Wird das salzsaure Salz mit überschüssigem Thionylchlorid etwa 1Z2 Stunde unter Rückfluß gekocht und danach das überschüssige Thionylchlorid im Vakuum entfernt, so hinterbleibt das salzsaure p-Guanidino-benzoylchlorid in Form eines farblosen Pulvers. Dieses wird in eine Lösung von ι Mol 4, 6-Diamino-chinaldin in Eisessig unter Rühren eingetragen. Unter schwacher Erwärmung bildet sich ein dichter Brei, der in Wasser gelöst wird, worauf aus der Lösung mit Natronlauge die Base gefällt und oft mit heißem Wasser gewaschen wird.
