Description
Process for the extraction of substances with an antibiotic effect, It is noted in patent 853,633 that the process of heat treating Substances with high or optimal heating rates, apart from the special ones the resulting effects in the killing of microorganisms, even a more far-reaching one Importance. It has been shown that after such a method treated substances also with the possibility of re-infection (open storage containers) keep fresh for a long time. This is in an example in the main patent with treated milk in more detail and already as a possible explanation means that through the 'special kind of germ killing see from this special Could form substances that have corresponding effects. Follow up on this Has made observations on the subject matter of the present invention guided.
If you namely in a bacterial population emthaltende solution, in dler the germs present, starting from z. B. room temperature, by heating at an optimal speed to the treatment temperature, at which you only change for a short time (e.g. for a certain germ: heating speed between 50 and roo ° / sec, treatment temperature = 68 °, Exposure time of the treatment temperature = 2 seconds), damaged, inoculated new germs of the same species, there is no growth of these germs. These relationships are illustrated in FIG. In this case, growth is determined in the known manner by measuring turbidity; The measured turbidity is plotted as the ordinate as a measure of growth, and the time as the abscissa. The curve r shows normal growth when a certain type of germ is inoculated into a sterile nutrient solution and incubated at an optimal growth temperature; Curve 2 shows the complete prevention of growth when the same germ is inoculated in the same amount into the filtrate from the previously treated incubation and incubation under the same conditions. It is thus evident that by the described treatment of a suspension of a certain germ according to the described method bacteriostatic or bactericidal effects can be produced which act against at least the same germ species as the treated one.
Die Gewinnung von Hemmstoffen mit Hilfe von auf diesen Erkenntnissen aufgebauten Verfahren bildet Gegenstand der Erfindung.2 shows the result of repeating what has been described Experiment (killing a bacterial population in a nutrient solution using an optimal heating rate and re-inoculation of the filtrate with the same Type of germ), but the results here are not based on turbidity measurement, but determined by counting germs according to the plate counting method. As controls I and II are in turn measured normal growth in the nutrient solution. While at Inoculation of a certain number of germs in a certain amount of sterile nutrient solution the number of germs increases in a known manner, remains when a certain number is introduced Germs in a substance treated according to the method keep this germ count for a long time Get time. The greater the ratio, the longer the duration of the inhibiting effect the initial bacterial count treated according to the invention (and thus the amount of generated inhibitor) to the subsequently inoculated to test the heroin effect Germ count is. So if you have an agglomeration of germs with an optimal heating-up speed treated and in the process kills the germs contained in the suspension, presumably in or on the germ, an inhibitor is formed which diffuses into the solution and at least one's own kind, d. H. thus the same type of germ as the treated one, inhibits. Whether the formation of the inhibitor or its death is primary is an open question stay. It is likely that not all germs are of such a specific nature Effect, but there are also more or less unspecific effects of the generated To observe the inhibitor. The extraction of inhibitors with the help of these The invention relates to a method based on knowledge.
In order to gain as much of the respective inhibitor as possible, one becomes work with the highest possible initial germ count in the treatment. For many cases becomes the inhibitor in the concentration or dilution in which it is after the treatment is sufficient. Then the method according to the invention is characterized by extraordinary Simplicity. Since the Aufschweminung does not have to take place in a nutrient solution, but The effect is also given in a so-called resting culture, so it can be from from the outset, the approach of the suspension takes place in such a way that the substance as such (possibly after filtration or after the germs have settled after the treatment and Diffusion of the inhibitors formed into the solution) can be used. Naturally The composition of the nutrient solution can also be used for certain areas of application from the outset be adapted so that they can be used as such with the inhibitors formed, z. B. is injectable. If the concentration obtained in this way is not sufficient, then will by isolating the inhibitor by known methods, the concentration can increase.
Is it not just about inhibiting a certain type of germ, but rather a certain germ flora is to be inhibited in growth, for which none is sufficient If there is an unspecific effect of an inhibitor, then one becomes after the procedure produce different specific acting inhibitors, which then act against the bacterial flora works. The production of such then more or less unspecific or combined inhibitor can be done in such a way that the necessary germ mixture is treated; or they can be concatenated with regard to their treatment dates Groups treated for themselves and 'the flora to be inhibited then the mixture of these Way to be exposed to separately manufactured inhibitors. The procedures in which a more extensive germinal flora must be treated and in which this entire flora is dealt with in one or more work steps, there may be restrictions subject than the growth and behavior in the solution before and during treatment usually do not need to be equal for all germs. One could in that Case that the individual germs, or at least some of them, are growing hinder the germs to grow separately and only shortly before the treatment mix the individual suspensions.
The areas of application do not need to be explained in more detail because they are readily apparent. The most essential areas are the fight against Infectious Diseases and Keeping Food Fresh. As in .. the main patent already stated, the method can also be applied in such a way that one can use direct Treatment damages the existing germs and at the same time thereby in the to be protected Organism or the fresh substance to be obtained produces inhibitors that are have an effect against further germs. If a germ flora is present under certain circumstances, however, it may also be necessary here to treat the (good) several times, the Conditions adapted to the individual germ species contained in it, to be treated.
The apparatus of the method according to the invention is carried out most expediently in the passage of the substance through a Aufheizvorricbtung, .die the set conditions are met, especially with regard to the heating rate and for the most diverse; Embodiments are conceivable. The heat can be generated in various ways Supply of energy from outside as well as through high-frequency heating. The latter Art will be necessary with poorly thermally conductive media, especially with direct ones Treatment of such an asset or a living body.
EIS is, of course, subject to the probable prerequisite that the inhibitor forms in the microorganism, it is also conceivable to separate the treated microorganisms from the solution as soon as possible after the treatment (by filtration, centrifugation or precipitation) before the inhibitors formed the microorganisms can diffuse into the solution, and then the: sludge separated from the solution in this way, in which most of the inhibitor is still present; continue to use.
As with the method of the patent 853,633 is also in general in the context of the process claimed here. a suitable recooling needle thought of the temperature treatment, which is usually necessary for this reason alone, to damage the inhibitors formed by too long exposure to the achieved Temperature range to be avoided.
