Description
Verfahren zur Darstellung neuer Derivate des 1, 8-Naphthyridins Gegenstand der Erfindung ist ein Verfahren zur Darstellung neuer Verbindungen der allgemeinen Formel worin R1 ein Alkoxy- oder Alk@7lthioradikal ist, wobei ein Wasserstoffatom der Alkylgruppe durch eine Alkoxy- oder eine Dialkylaminogruppe ersetzt sein kann, R2 einen Alkyl-, Aryl- oder Aralkylrest bedeutet und R3 die Amino-, eine Alkylamino-, Dialkylamino- oder Acylaminogruppe ist oder die gleiche Bedeutung wie R1 hat. Es wurde gefunden, daß diese Derivate des Kaphthyridins eine überraschend gute Wirksamkeit gegen Kleinorganismen, z. B. Bakterien, Protozoen, wie Amöben, besitzen und daher als Desinfektionsmittel sowohl für Gegenstände als auch für den lebenden Körper, insbesondere als Wund-und Darmdesinfizienzien, wertvoll sind. Sie können in Form von Pulvern, Lösungen oder zu Sprays, Emulsionen, Suspensionen, Pasten, Salben, Pudern usw. verarbeitet, zum Einreiben, Einnehmen, Injizieren, Versprühen, Imprägnieren usw. verwendet werden. DasVerfahren zurDarstellungderneuenN'aphthyridinderivate der oben erwähnten allgemeinen Formel ist dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen Formel worin 1 ein Halogenatom, R2 ein Alkyl-, Aryl- oder Aralkylradikal und 1 ein Halogenatom, die Amino-, eine Alkylamino-, Dialkylamino- oder Acylaminogruppe bedeuten, zweckmäßig in Gegenwart eines Kondensationsmittels mit einem aliphatischen Alkohol oder Thioalkohol umsetzt, in welchem ein Wasserstoffatom der Alkylgruppe durch eine Alkoxy- oder Dialkylaminogruppe ersetzt sein kann. Als Kondensationsmittel wird vorteilhaft ein Alkalialkoholat oder Alkalithioalkoholat des jeweils zum Umsatz benützten Alkohols bzw. Thioalkohols verwendet. Beispiel i i0,5 g Natrium werden in 2,51 abs. n-Butanol aufgelöst und der heißen Lösung Bog 2-Chlor-4-methyl-7-amino-i,8-naphthyridin zugefügt. Das Gemisch wird 3 Stunden am Rückflußkühler gekocht, dann erkalten gelassen. Die Reaktionslösung wird mit Wasser bis zur neutralen Reaktion gewaschen, getrocknet und im Vakuum eingedampft. Der aus 2 n-Butoxy-4-methyl-7-amino-i, 8-naphthyridin bestehende Rückstand wird aus Essigester umkristallisiert. Ausbeute 76 g, entsprechend 95 % d. Th. Zur Lösung fügt man unter Turbinieren und Kochen portionenweise 15o g 2-Chlor-4-methyl-7-aminoi, 8-naphthyridin und kocht 24 Stunden mit aufgesetztem Rückflußkühler. Das ausgeschiedene Kochsalzwird abgenutschtund mit Benzol gutausgewaschen. Die Filtrate werden mit Wasser ausgeschüttelt und getrocknet, das Lösungsmittel wird verdampft und der hinterbleibende Rückstand aus einer Mischung von Benzol und Petroläther umkristallisiert. Das so in einer Menge von 1519 = 71 % d. Th. erhaltene 2 - (2'- Diäthylaminoäthoxy) - 4- methyl- 7 - amino-i, 8-naphthyridin schmilzt bei 123 bis i24° C, ist sehr leicht löslich in verdünnten Säuren, mit Ausnahme von Petroläther gut löslich in den gebräuchlichen organischen Lösungsmitteln, schwer löslich in Wasser. Beispiel 4 23,6 g Natrium werden in siedendem Toluol unter starkem Turbinieren verteilt und dann 121 g 2-Diäthylaminoäthanol langsam zugegeben. Nach 3stündigem Kochen werden Zoo g 2, 7-Dichlor-4-methyl-i, 8-naphthyridin, in 50o ccm heißem Toluol gelöst, zugetropft, worauf das Gemisch 24 Stunden am Rückflußkühler weitergekocht wird. Das entstandene Kochsalz wird abfiltriert, das Filtrat mit Wasser gewaschen und getrocknet. Nach dem Verdampfen des Lösungsmittels wird der Rückstand im Hochvakuum rektifiziert. Dasso erhaltene 2, 7-Bis-(2'-diäthylaminoäthoxy)-4-methyli, 8-naphthyridin ist ein schwach gelbes 01 vom Sdp o,oo8 2o2 bis 2o4° C.
