- Es ist ebenfalls berichtet worden (Madras, B. K., J. B. Kamien, M. Fahey, D. Canfield, et al. (1990), Pharmacol Biochem. Behav. 35: 949-9531, dass die Einführung einer Alkyl- oder Allylgruppe die Bindungspotenz nicht beseitigte. Eine N-Iodallylgruppe am Tropan lieferte potente und selektive Liganden für den DAT, und Altropan wird zur Zeit als ein SPELT-Bildgebungsagenz entwickelt (Elmaleh, D. R., B. K. Madras, T. M. Eine 99mTechnetium-markierte Verbindung, Technepin, die potent und selektiv an den DAT bindet und ausgezeichnete in vivo-SPECT-Bilder liefert, ist berichtet worden. (Madras, B. K., A. G. Es wurde über 2-Carbomethoxy-3-3(bis(4-fluorophenyl)methoxy)tropane berichtet worden (Meltzer, P. C., A. Y. Liang und B. K. Madras (1994), J. Med. Chem. 37: 2001-2010). Das S-Enantiomer, (S)-(+)-2β-Carbomethoxy-3α-(bis(4-fluorophenyl)methoxy)tropan (Dufluoropin) war wesentlich potenter (IC50: 10,9 nM) und selektiver (DAT v. SERT: 324) als irgendein anderes der weiteren sieben Isomere, einschließlich der R-Enantiomere.
- Ferner beschreiben P. C. Meltzer et al. J. Med. Chem..39, 371-379 (1996) die Synthese und Inhibierung von 2-Carbomethoxy-3-(diarylmethoxy)-1αH, 5αH-tropananaloga bei der Bindung an den Dopamintransporter (DAT). Innerhalb der Phenyltropanreihe wird nicht nur auf die Affinität dieser Verbindungen zum DAT, sondern auch auf ihre DAT vs. Serotonin-Transporterselektivität Bezug genommen.
- Es werden Wirkstofftherapien für den Kokainmissbrauch benötigt. Es besteht auch ein Bedürfnis nach Schutzmitteln für neurodegenerative Erkrankungen wie die Parkinson'sche Erkrankung und die Alzheimer'sche Krankheit wie auch nach therapeutischen Mitteln für die Dopamin-verwandte Fehlfunktion, wie der Attention Deficit Disorder. Um solche Therapien zu etablieren wird nach Verbindungen gesucht, die die Monoaminwiederaufnahme in das System des Säugers inhibieren.
- Die Inhibierung des 5-Hydroxytryptamin-Wiederaufnahme besitzt eine Wirkung gegenüber Erkrankungen, die mit 5HT-Rezeptoren verknüpft sind. Verbindungen, die eine solche Inhibierung liefern, können zum Beispiel als therapeutische Antidepressiva nützlich sein.
- Kokainerkennungsstellen sind auf Monoamintransportern lokalisiert, wie zum Beispiel dem Dopamintransporter (DAT) und dem Serotonintransporter (SERT). Diese Transporter sind ihrerseits auf Monoamin-Nervenenden lokalisiert. Verbindungen, die an diese Stellen binden, können nützlich sein als (i) Sonden für neurodegenerative Erkrankungen (z.B. Parkinson'sche Krankheit), (ii) therapeutische Wirkstoffe für neurodegenerative Erkrankungen (z.B. Parkinson'sche und Alzheim'sche Krankheit), (iii) therapeutische Wirkstoffe zur Dopaminfehlfunktion (z.B. Attention Deficit Disorder), (iv) Behandlung der psychiatrischen Fehlfunktion (z.B. Depression) und (v) Behandlung der klinischen Fehlfunktion (z.B. Migräne).
- ZUSAMMENFASSUNG DER ERFINDUNG
- Die Verbindungen dieser Erfindung sind neue Tropananaloga, die an Monoamintransporter binden. Somit stellt die vorliegende Erfindung Tropananaloga mit einem der folgenden Formeln bereit: Worin:

mit der Ausnahme, dass wenn X = N, R1 nicht COR6 ist. - Die Substituenten an der 2- und 3-Position des Rings kann α- oder (3- sein. Obgleich R1 in der 2-Position gezeigt ist, sollte klar sein, dass die Substitution an der 3-Position auch eingeschlossen ist, und die Position ist von der Nummerierung des Tropanrings abhängig. Die Verbindungen der vorliegenden Erfindung können racemisch, reine R-Enantiomere oder reine S-Enantiomere sein. Somit bezwecken die hier veranschaulichten Strukturformeln eine Darstellung von sowohl dem Enantiomer als auch dem Diastereomer der veranschaulichten Verbindung.
- Die Verbindungen der vorliegenden Erfindung können radiomarkiert sein, zum Beispiel zum Testen von Kokainrezeptoren. Bestimmte bevorzugte Verbindungen der vorliegenden Erfindung besitzen eine hohe Selektivität für den DAT gegenüber dem SERT. Die vorliegende Erfindung stellt auch pharmazeutische therapeutische Zusammensetzungen zur Verfügung, die die Verbindungen, in einem pharmazeutisch akzeptablen Träger formuliert, umfassen.
- Ferner stellt die Erfindung ein Verfahren zum Inhibieren der 5-Hydroxytryptamin-Wiederaufnahme eines Monoamintransporters bereit, in dem der Monoamentransporter mit einer (5-HT-inhibierenden) Menge einer Verbindung der vorliegenden Erfindung, die die Wiederaufnahme eines 5-Hydroxy-Tryptamins hemmt, kontaktiert wird. Die Inhibierung der 5-Hydroxy-Trpytamin-Wiederaufnahme eines Monoamentransporters in einem Säuger wird gemäß der vorliegenden Erfindung bereitgestellt durch Verabreichung eines Säugers mit einer 5-HT-inhibierenden Menge einer Verbindung der vorliegenden Erfindung in einem pharmazeutisch akzeptablen Träger. Bevorzugte Monoamentransporter für die Praxis der vorliegenden Erfindung schließen den Dopamintransporter, den Serotonintransporter und den Norepinephrintransporter ein.
- In einer bevorzugten Ausführungsform liefert die Erfindung auch ein Verfahren zum Inhibieren der Dopaminwiederaufnahme eines Dopamintransporters durch Kontaktieren des Dopamintransporters mit einer Menge an einer Verbindung der vorliegenden Erfindung, die die Dopaminwiederaufnahme inhibiert. Die Inhibierung der Dopaminwiederaufnahme eines Dopamintransporters in einem Säuger wird gemäß der vorliegenden Erfindung bereitgestellt durch Verabreichung einer Dopamin inhibierenden Menge einer Verbindung der vorliegenden Erfindung in einem pharmazeutisch akzeptablen Träger zu einem Säuger.
- Der hier verwendete Ausdruck "Niedrigalkyl" bezeichnet aliphatische gesättigte, verzweigte oder geradkettige, monovalente Kohlenwasserstoffsubstituenten, die 1 bis etwa 8 Kohlenstoffatome enthalten, wie Methyl, Ethyl, Isopropyl, n-Propyl, n-Butyl, (CH2)nCH3, C(CH3)3; usw., weiter bevorzugt 1 bis 4 Kohlenstoffatome. Der Ausdruck "Niedrigalkoxy" bezeichnet Niedrigalkoxy-Substituenten, die von 1 bis etwa 8 Kohlenstoffatome enthalten, wie etwa Methoxy, Ethoxy, Isopropoxy, usw., weiter bevorzugt 1 bis 4 Kohlenstoffatome.
- Der hier verwendete Ausdruck "Niedrigalkenyl" bezeichnet alipathische und gesättigte, verzweigte oder geradkettige Vinyl-Kohlenwasserstoffsubstituenten, die von 2 bis etwa 8 Kohlenstoffatome enthalten, wie etwa Allyl, usw., weiter bevorzugt 2 bis 4 Kohlenstoffatome. Der Ausdruck "Niedrigalkynyl" bezeichnet Niedrigalkynyl-Substituenten, die von 2 bis etwa 8 Kohlenstoffatome, weiter bevorzugt 2 bis 4 Kohlenstoffatome enthalten, wie etwa zum Beispiel Propyn, Butyn, usw.
- Falls hier verwendet schließen den Ausdrücke substituiertes Niedrigalkyl, substituiertes Niedrigalkoxy, substituiertes Niedrigalkenyl und substituiertes Niedrigalkynyl entsprechende Alkyle-, Alkoxy-, Alkenyl- oder Alynyl-Gruppen ein, die mit Halogen-, Hydroxy-, Carboxylsäure oder Carboxamid-Gruppen usw. substituiert sind, wie etwa zum Beispiel -CH2OH, -CH2CH2COOH, -CH2CONH2, -OCH2CH2OH, -OCH2COOH, -OCH2CH2CONH2, usw. Wie hier verwendet bedeuten die Ausdrücke Niedrigalkyl, Niedrigalkoxy, Niedrigalkenyl und Niedrigalkynyl solche wie oben beschriebener Gruppen dann wenn sie praktisch substituiert sind.
- Wenn X ein Kohlenstoffatom als Ringmitglied enthält, erfolgt die Bezugnahme auf X hier gelegentlich als einer Kohlenstoffgruppe. Wie dieser Ausdruck hier verwendet wird, bedeutet er somit, wenn X eine Kohlenstoffgruppe ist, daß ein Kohlenstoffatom ein Ringmitglied an der X-Position ist (d.h. der 8-Position).

- Gemäß der vorliegenden Erfindung werden neue Tropanverbindungen bereitgestellt, die an Monoamintransporter, vorzugsweise dem DAT binden. Bestimmte bevorzugte Verbindungen besitzen auch eine hohe Selektivität gegenüber DAT im Vergleich zu SERT. Gemäß der vorliegenden Erfindung werden Topanaloga mit Substituenten in 6- oder 7-Position der Tropanstruktur bereitgestellt. Bevorzugte Verbindungen dieser Ausführungsform der Erfindung schließen solche mit der Formel: ein, worin X, Ar, R2 und m dieselbe Bedeutung wie oben definiert haben. Besonders bevorzugte Verbindungen weisen auf: X, das ein Stickstoffatom als einem Ringmitglied einschließt, R2 ist Hydroxy oder Methoxy, und Ar ist Phenyl, substituiertes Phenyl wie Mono- oder Di-Halogen-substituiertes Phenyl, oder ein Diarylmethoxy einschließlich einer Halogensubstitution solcher Gruppen.

1 ) gezeigt hergestellt werden. Die Synthese dieser 6- und 7-oxygenierten Tropane gemäß dem Schema 1 beruht auf einer Mannich-Reaktion. (Robinson, R. (1917), J. Chem. Soc. 111: 762.) Dimethoxydihydrofuran1 wird für 12h (Stunden) in 3N HCl gerührt und dann durch Zugabe von wässrigem NaOH neutralisiert. Methylaminhydrochlorid in Wasser sowie Aceton-Dicarboxylsäureanhydrid2 in MeOH werden zugegeben. Eine Mischung von 6- und 7-Hydroxy-3β-Ketoester3 und4 wird erhalten. Die Exo(β)-Stereochemie der Hydroxylgruppe beim C6 (3) oder C7 (4) wird durch NMR-Analyse bestätigt. - Eine chromatographische Trennung liefert auch die 6- und 7-Methoxyverbindungen
12 und13 . Die 6- und 7-Hydroxy-β-Ketoester3 und4 werden jeweils mit Dimethoxymethan und pTSA methoxymethyliert, um5 zu liefern. Obgleich beide 6- und 7-substituierten Verbindungen durch die nachfolgenden Reaktionsschritte gezogen werden, veranschaulicht Schema 1 das Reaktionsschema für die 7-substituierte Verbindung. Die anschließende Umwandlung (Carroll, F. I., P. Kotian, A. Dehghani, J. L. Gray, et al. (1995), J. Med. Chem. 38: 379-388; Keverline, K. I., P. Abraham, A. H. Lewin and F. I. Carroll (1995), Tetrahedron Lett. 36: 3099-3102) in das Enoltriflat6 wird durch Bistrimethyl-silylamid und Phenyltriflimid erreicht. Die Alkene7 werden dann erhalten (85%) durch Suzuki-Kopplung der Triflate mit 3,4-Dichlorophenylborsäure. Eine Reduktion von7 mit SmI2 bei -78°C in Gegenwart von McOH und anschließende Chromatographie ergibt die gesättigten Tropane,8 (61%) und9 (20%). Die Verbindung8 existiert in einer Dreh-Wannen-Konformation, während Verbindung9 eine Dreh-Sessel-Konformation annimmt. Die Methoxymethyl(MOM)-Gruppe von jeweils8 und9 wird schließlich in hoher Ausbeute (85%) mit Trimethylsilylbromid in CH2Cl2 bei 0°C entfernt, um die entsprechenden Hydroxytropane10 und11 zu ergeben. (Chen, Z., S. Izenwasser, J. L. Katz, N. Zhu, C. L. Klein und M. L. Trudell (1996), J. Med. Chem. 39: 4744-4749; #214.) Das Alken 7 kann auf die gleiche Weise zum Entfernen der MOM-Gruppe in dieser Stufe behandelt werden und liefert ein 6- oder 7-substituiertes, ungesättigtes Propananalogon. - Die Ester können durch Acylierung mit geeigneten Säurechloriden oder -anhydriden erhalten werden. Die 7- (und 6-) Methoxy-3β-Ketoester
12 (und13 ) werden in ihre Enoltriflate umgewandelt, und analoge Umwandlungen liefern dann die Methoxytropane14 und15 . - Die biologischen Daten für die jeweiligen 2-Carbomethoxy-3-(3,4-dichlorophenylpropane der vorliegenden Erfindung sind in Tabelle 1 gezeigt.
- Die 7-OH-Verbindungen können extrem potent und selektiv eine Bindung an den DAT ausüben. Somit manifestiert die Verbindung
10 einen IC50 < 2 nM und eine hohe Selektivität (DAT gegenüber SERT > 500), und die 3β-Verbindung11 ist gleich potent beim DAT (IC50 = 1 nM), jedoch weniger selektiv (DAT gegenüber SERT = 20). Während die Stammverbindung 2-Carbomethoxy-3,4-dichlorophenyltropan potent ist (IC50 = 1,09 nM), jedoch keine Selektivität aufweist (DAT gegenüber SERT = 2), behalten die 7β-Hydroxyverbindungen überraschenderweise ihre Potenz und sind ebenfalls viel stärker selektiv (DAT gegenüber SERT = 20 für 3β (Sessel) und = 1450 für 3α (Wanne)). Die Einführung der 7β-Hydroxylgruppe hat die Selektivität dieser Verbindungen zu DAT stark und unerwartet verbessert. - Diese Verbindungen sind racemisch. Die reinen Enantiomere, Verbindung
10 und Verbindung11 , werden enantiorein synthetisiert. Die Auflösung wird erreicht durch Bildung der diastereomeren Tartratsalze der Verbindung5 , oder durch Bildung und Trennung der diastereomeren Enolcamphanate. Somit wird das Tartratsalz der (MOM- oder AcO-) geschützten Verbindung5 durch Standardmethoden, die dem Fachmann gut bekannt sind, rekristallisiert, um jedes der diastereomeren Salze bereitzustellen. Die Behandlung mit einer Base liefert die enantioreine (1R)-Verbindung5 und (1S)-Verbindung5 . Alternativ liefert auch die Bildung der Diastereomerenenolcamphanatester der Verbindung5 , die Rekristallisierung und die Hydrolyse mit LiOH ebenfalls die enantioreine (1R)-Verbindung5 und (1S)-Verbindung5 . Diese enantioreinen Ketone werden durch die ganze Abfolge getragen, um enantioreine Zielverbindungen zu liefern. - Somit können 3β- (Sessel-) und 3α- (Wannen-) Hydroxypropane oder verschiedentliche analoge Alkoxytropane hergestellt werden.
- Die Potenz der in Tabelle 1 gezeigten 7-Hydroxy- und 7-Methoxy-Analoga von 2-Carbomethoxy-3-(3,4-di-chlorophenyltropan, 3β (Sessel) und 3α (Wanne), wurden durch den Dopamintransportertest und den Serotonintransportertest wie nachfolgend beschrieben bestimmt.
- Die Herstellung der 3-Diarylmethoxytropane ist in Schema 2 veranschaulicht unter Verwendung eines 7-substituierten 3β-Ketotropans zum Bilden der entsprechenden 7β-OH-Verbindung (siehe
2 ). Die 7α-OH- (sowie 6α-OH- und 6β-OH-) Verbindungen werden durch eine identische Abfolge getragen. Andere 3-Aryloxytropane können durch analoge Reaktionsschemas hergestellt werden. Die Synthese der 7β-OH-Verbindung21 beruht auf den vorangehenden Synthesewegen. Somit wird der MOM-geschützte (1S)-Ketoester5 mit LiBH(Bu1)3 in Tetrahydrofuran (THF) reduziert, um die 2β-COOCH3-3α-OH-Verbindung19 zu liefern. Alternativ kann die Reduktion mit NaBH4 ausgeführt, um eine Mischung der 2α-, 2β- sowie 3α-, 3β-substituierten Verbindungen zu liefern. Die 2α-COOCH3-3α-OH-Verbindung kann durch Behandlung mit NaHCO3 invertiert werden, um die bevorzugte 2β-COOCH3-3α-OH19 zu liefern. Die Reaktion der Verbindung19 mit Diarylmethylchlorid (van der Zee 1980) ergibt die MOM-geschützte Verbindung20 , die mit Trimethylsilylbromid (TMSBr) in CH2Cl2 entschützt wird, um die gewünschte Zielverbindungen21 zu ergeben. Der Arylring kann mit einem oder mehreren Halogenatomen, vorzugsweise Chlorid oder Iodid, Hydroxygruppen, Nitrogruppen, Aminogruppen einschließlich mono- und di-alkyl-substituierten Gruppen mit 1-8 Kohlenstoffatomen, Cyanogruppen, Niedrigalkylgruppen mit 1-8 Kohlenstoffatomen, Niedrigalkoxy gruppen mit von 1-8 Kohlenstoffatomen, Niedrigalkenylgruppen mit von 2-8 Kohlenstoffatomen, Niedrigalkynylgruppen mit von 2-8 Kohlenstoffatomen, sowie Kombinationen solcher Substituenten substituiert sein. Bevorzugte Arylgruppen haben Substituenten einschließlich Br, Cl, I, F, OH, OCH3, CF3, NO2, NH2, CN, NHCOCH3, N (CH3)2, (CH2)nCH3, worin n= 0-6, COCH3, C (CH3)3, 4-F, 4-Cl, 4-I, 2-F, 2-Cl, 2-I, 3-F, 3-Cl, 3-I, 3,4-diCl, 3,4-diOH, 3,4-diOAc, 3,4-diOCH3, 3-OH-4-Cl, 3-OH-4-F, 3-Cl-4-OH, 3-F-4-OH, Allyl, Isopropyl und Isobutyl. - Potente "6,7-Brücken"-Hydroxyverbindungen der Tropananaloga der vorliegenden Erfindung, z.B. der 8-Azafamilie, können demethyliert und mit einer Vielzahl von Alkyl- und Alkylaryl-Gruppen realkyliert werden. Somit wird zum Beispiel
21 mit ACE-Cl behandelt, um die NOR-Verbindung zu liefern, oder eine Demethylierung kann vor der Entschützung durch Behandlung von20 mit ACE-Cl in Gegenwart von 2,6-Lutidin ausgeführt werden. Die Reaktion mit geeigneten Alkylchloriden in Gegenwart von K2CO3 oder alternativ mit KF/Celite liefert dann N-substituierte Verbindungen wie N-(CH2)n-Ar (n=1-3; Ar=Phenyl oder Halophenyl). - Die MOM-Gruppe wird mit TMSBr entfernt.
- Zur Beschreibung der Bezugsverbindungen 8-Oxa- und 8-Carba-Tropane sowie die Herstellung davon wird bezug genommen auf die veröffentlichte Anmeldung WO 9902526 A1.
- Ein Vergleich der Bindungsstärke der 8-Oxabicyclen mit den 8-Azabicyclen zeigt, dass, obgleich die Potentesten in beiden Klassen nahezu äquipotent sind (IC50 =1-3nM), die weniger potenten Verbindungen (R=H, F) typischerweise schwächer in der 8-Oxa als in der 8-Aza-Serie sind.
- Das Enoltriflat wird mit den passenden Poly-aromatischen Borsäuren umgesetzt (Thompson, W.J. und J. Gaudino (1984), J. Org. Chem. 49: 5237-5243), um die 2,3-ene zu liefern. Eine SmI2-Reduktion ergibt die 3α und 3β-Konformere: worin X ein Sauerstoffatom oder ein Kohlenstoffatom ist, Ar vorzugsweise 4-substituiertes Phenyl, Naphthyl, Anthracenyl oder Phenanthrenyl ist, und R vorzugsweise Niedrigalkyl, Niedrigalkoxy oder -Amino wie oben definiert ist. Die Verbindungen werden als Racemate und Enantiomerenpaare hergestellt.
- Enantiomere und Diastereomere können durch Silicagel-Chromatographie unter Verwendung von 2% Ammoniak in Ethylacetat als Eluent, oder nach Bedarf mit anderen Lösungsmittelsystemen getrennt werden. Verbindungen aus der obigen Reihe werden hydrolysiert (LiOH) und mit Oxalylchlorid und nachfolgend mit Aminen wie Morpholin oder Piperidin zum Liefern von Amiden behandelt.
- 2,3- (und 3,4-) ungesättigte 8-Aza-Tropane sind weitere bevorzugte Ausführungsformen der vorliegenden Erfindung. Beispiele von bevorzugten solchen Verbindungen besitzen die folgende Formel: worin X vorzugsweise Sauerstoff, N-Alkyl oder eine Kohlenstoffstruktureinheit ist, R vorzugsweise Morpholinyl, Piperidinyl oder Methoxy ist, Ar vorzugsweise Phenyl oder Naphthyl ist, die jeweils substituiert sein können mit Halogen, Alkenyl mit 2-8 Kohlenstoffatomen oder Alkynyl mit 2-8 Kohlenstoffatomen, wie zum Beispiel 4-Cl, 4-F, 4-Br, 4-I, 3,4-Cl2, Ethenyl, Propenyl, Butenyl, Propynyl, Butynyl, etc.
- Die Synthese der 2,3-ungesättigten 8-Aza- und 8-Oxa-Tropane ist im Schema 6 (siehe
6 ) veranschaulicht. Die 3-(substituiertes Aryl)-8-Oxabicyclo(3.2.1)octane können aus dem Ketoester45 erhalten werden Somit wird 2,5-Dimethoxytetrahydrofuran43 mit 1,3-bis(Trimethylsiloxy)-1-methoxybuta-1,3-dien44 in CH2Cl2 in Gegenwart von TiCl4 umgesetzt, um das Keton45 zu ergeben. Das Keton45 wird dann durch Reaktion mit N-Phenyltrifluoromethansulfonimid und Natrium-bis(trimethyl-silyl)amid in THF in die Verbindung46 umgewandelt. Das Enoltriflat46 wird mit Arylborsäure in Gegenwart von Tris(dibenzyliden-aceton)dipalladium(0) gekoppelt, um die Aryloctene47 zu bekommen. - Die Verbindungen
47e und f werden aus der enantioreinen Verbindung (1R)-45 und der Verbindung (1S)-45 synthetisiert. Alternativ wird eine Diastereomerenmischung von Enolcamphanaten durch Reaktion der Verbindungen (1R/S)-45 mit (S)-(-)-Camphanylchlorid in THF hergestellt. Die Rekristallisierung aus CH2Cl2/Hexan ergibt dann das reine Diastereomer (1R)-Enolcamphanat, wie durch NMR bestätigt. Die restliche Mischung der Enolcamphanatdiastereomere wird mit LiOH zum Herstellen der Verbindungen (1R/S)-45 behandelt, die dann mit (R)-(+)- Camphanylchlorid umgesetzt wird. Die Rekristallisierung dieses Enolcamphanats ergibt dann das reine (1S)-Enolcamphanat-Diastereomer. Eine quantitative Hydrolyse der enantiomer reinen, einzelnen Camphanatester mit LiOH liefert die Ketone (1S)-45 und (1R)-45 (chirale HPLC-OC-Säule: (1R)-45 und (1S)-45 >96% ee für jedes der Enantiomere). Die gereinigten Enantiomere werden dann der zuvor beschriebenen Abfolge von Schritten unterzogen, um die enantiomer reinen 8-Oxatropenanaloga27e und f zu erhalten (Schema 3,3 ). Die absolute Konfiguration wurde durch Röntgenstrukturanalyse bestätigt. Eine Isomerisierung des 2,3-Ens zum Bilden der 3,4-Ene wird mit einer Base erreicht. - Die biologischen Daten für repräsentative 2,3-Ene der vorliegenden Erfindung mit einem 3-(3,4-Dichloro)phenyl-Substituenten sind in Tabelle 2 gezeigt.
- Das (1R)-Enantiomer
47e bindet potent an den DAT (IC50 = 4,6 nM) und sehr schwach an den SERT (IC50 = 2120 nM) und ist etwa 460-fach selektiver. Überraschend und unerwartet behält das (15)-Enantiomer47f die Potenz DAT:IC50 = 58,2 nM) und im wesentlichen die Selektivität DRT/SERT = 800) bei. Die 8-Aminanaloga dieser Verbindungen können über die analoge Enoltriflatchemie hergestellt werden. Somit wird das 2-Carbomethoxytropan-2-on ähnlich in sein Enoltriflat umgewandelt und mit 3,4-Dichlorophenylborsäure zum Liefern der Verbindung48 gekoppelt. Die Verbindung48 gehört zu den potentesten und selektivsten (790-fach) Verbindungen (DAT IC50 = 1, 1 nM); SERT IC50 = 867 nM). Diese Verbindungen bieten die Möglichkeit, zwischen der Bindung an den SERT gegenüber dem DAT zu differenzieren, sowie einen Vorteil zu ziehen aus den unterschiedlichen biologischen Profilen (biologische t1/2, Toxizität, Metabolismus), die diese Verbindungen bieten. - Gemäß einer anderen bevorzugten Ausführungsform der vorliegenden Erfindung wurden die 2,3- (und 3,4-) Didehydrotropane und 8-Oxatropane an den 6- und 7-Positionen hydroxyliert oder alkoxyliert, um Verbindungen bereitzustellen, die zur intramolekularen Wasserstoffbindung an den 8-Oxa- und 8-Aza-Positionen in der Lage sind. Die bei den 2,3-En- und den 7-OH-Verbindungen beobachtete Selektivität liefert einen Synergismus und bietet extrem potente und selektive Verbindungen. Die 7β-Hydroxytropane
10 und11 sind potent und selektiv. Das (IR)2,3-En-Enantiomer47e bindet potent und selektiv an den DAT, und das (1R)8-Aza-2,3-En48 ist unter den potentesten und selektivsten DAT-Inhibitoren. Verbindungen, die beide Funktionalitäten zeigen, sind besonders bevorzugt. Die Umwandlung eines Enol-Triflats über Suzuki-Kupplung mit passenden Arylborsäuren liefert die bevorzugten Verbindungen (siehe auch Schema 1,1 ). Solche Verbindungen, d.h. Bicyclo(3.2.1)octan, schließen Verbindungen ein, die die folgende Formel aufweisen: worin X NCH3 ist, R vorzugsweise Morpholinyl, Piperidinyl oder Methoxy ist, R2 vorzugsweise Hydroxy oder Methoxy in der 6- oder 7-Position ist, Ar vorzugsweise Phenyl oder Naphthyl ist, das jeweils mit Halogen, Alkenyl mit 2-8 Kohlenstoffatomen oder Alkynyl mit 2-8 Kohlenstoffatomen wie zum Beispiel 4-Cl, 4-F, 4-Br, 4-I, 3,4-Cl2, Ethenyl, Propenyl, Butenyl, Propynyl, Butynyl, etc. substituiert sein kann. - Diese Verbindungen können entweder als freie Basen oder als pharmakologisch aktive Salze davon wie das Hydrochlorid, das Tartrat, das Sulfat, das Naphthalen-1,5-disulfonat oder dergleichen hergestellt werden.
- Die vorliegende Erfindung stellt ebenfalls pharmazeutische Zusammensetzungen bereit, vorzugsweise mit den Verbindungen der vorliegenden Erfindung in einem pharmazeutisch akzeptablen Träger. Pharmazeutisch akzeptable Träger sind dem Fachmann gut bekannt. In einer bevorzugten Ausführungsform ist die pharmazeutische Zusammensetzung eine flüssige Zusammensetzung in einem pyrogen-freien, sterilisierten Behälter oder einem Vial. Der Container kann eine Einheitsdosis oder eine Mehrfachdosis sein.
- Die Verbindungen und pharmazeutischen Zusammensetzungen der vorliegenden Erfindung können verwendet werden zum Inhibieren der %-Hydroxytryptaminwiederaufnahme eines Monoamintransporters, insbesondere der Wiederaufnahme durch den Dopamintransporter, den Serotonintransporter oder den Norepinephrintransporter. Eine wirksame Dosis der Verbindung wird einem Patienten auf der Grundlage der in vitro bestimmten IC50-Werte verabreicht. Der Verabreichungsweg kann unterschiedlich sein, wird jedoch hauptsächlich aus intravenösen, nasalen und oralen Verabreichungswegen ausgewählt. Die wirksame Dosis kann je nach der Verabreichungsart variieren, wie im Stand der Technik gut bekannt.
- Eine Fehlfunktion der Dopaminneuronen spielte in mehreren neuropsychiatrischen Erkrankungen eine Rolle. Eine Bildgebung von Dopaminneuronen bietet wichtige klinische Information, die zur Diagnose und zu therapeutischen Behandlungen relevant ist. Dopaminneuronen erzeugen Dopamin, setzen den Neurotransmitter frei, und beseitigen das freigesetzte Dopamin mit einem Dopamintransporterprotein. Verbindungen, die an den Dopamintransporter binden, sind wirksame Messmittel von Dopaminneuronen und können zu Bildgebungsmitteln zur PET und zur SPELT-Bildgebung umgewandelt werden. Bei der Identifizierung einer geeigneten Verbindung für den Dopamintransporter ist ein erster wichtiger Schritt, die Affinität und Selektivität eines Kandidaten zum Dopamintransporter zu messen. Die Affinität wird durch ein Ausführen von Radiorezeptortests gemessen. Ein radiomarkierter Marker für den Transporter, z.B. (3H)WIN35,428, wird mit einem ummarkierten Kandidaten und einer Quelle des Transporters, gewöhnlicherweise Hirnstriatum, inkubiert. Die Wirkung unterschiedlicher Konzentrationen des Kandidaten zum Inhibieren der (3H)WIN35,428-Bindung wird quantifiziert. Die Konzentration der Verbindung, die 50% der (3H)WIN35,428-Bindung an den Transporter inhibiert (IC50-Wert), wird als Maß seiner Affinität gegenüber dem Transporter verwendet. Ein geeigneter Bereich von Konzentrationen des Kandidaten ist typischerweise 1-10 nM.
- Es ist auch wichtig, die Selektivität des Kandidaten zum Dopamin- im Vergleich zum Serotonintransporter zu messen. Der Serotonintransporter ist auch im Striatum detektierbar, d.h. dem Hirnbereich mit der höchsten Dichte an Dopaminneuronen, sowie in Hirnregionen, die das Striatum umgeben. Es ist nötig, zu bestimmen, ob die Kandidatenverbindung beim Dopamin- potenter ist als beim Serotonintransporter. Bei größerer Selektivität (>10-fach) erlaubt die Sonde genaue Messwerte des Dopamintransporters in diesem interessierten Bereich, oder liefert eine wirksame Behandlungsmodalität für den Dopamintransporter. Deshalb wird eine Messung der Sondenaffinität des Serotonintransports durch Tests parallel zu den Dopamintransportertests durchgeführt. (3H)Citalopram wird zum Radiomarkieren von Bindungsstellen auf dem Serotonintransporter verwendet, und Kompetitionsstudien werden mit der Kandidatenverbindung bei unterschiedlichen Konzentrationen ausgeführt, um einen IC50-Wert zu erzeugen.
- Diese Erfindung wird durch die folgenden Beispiele weiter veranschaulicht. Es ist nicht beabsichtigt, dass diese Beispiele den Umfang der beanspruchten Erfindung in irgendeiner Weise einzuschränken. Die Beispiele liefern geeignete Methoden zur Herstellung von Verbindungen der vorliegenden Erfindung. Der Fachmann kann jedoch Verbindungen der vorliegenden Erfindung durch irgendwelche anderen Mittel herstellen. Wie dem Fachmann gut bekannt können für die veranschaulichten Verbindungen durch geeignete Modifikation der Reaktanden andere Substituenten bereitgestellt werden.
- Alle Beispielszielverbindungen werden voll analysiert (SP, TLC, CHN, GC und/oder HPLC) und charakterisiert (1H-NMR, 13C-NMR, MS, IR), bevor eine biologische Untersuchung durchgeführt wird. Die Affinität aller Verbindungen gegenüber DAT, SERT und NET wird gemessen. NMR-Spektren werden auf einem Bruker 100-, einem Varian XL 400- oder einem Bruker 300-NMR-Spektrometer aufgenommen. Tetramethylsilan ("TMS") wird als interner Standard verwendet. Schmelzpunkte sind unkorrigiert und werden auf einem Gallenkamp-Schmelzpunktgerät gemessen. Dünnschichtchromatographie (TLC) wird auf Baker Si 250F-Platten durchgeführt. Die Sichtbarmachung erfolgt mit Joddampf, UV-Bestrahlung oder durch Behandlung mit Phosphormolybdänsäure (PMA). Präparative TLC wird auf Analtech Uniplates Silica Gel GF 2000 Micron durchgeführt. Eine Flash-Chromatographie wird auf Baker Silica Gel 40 mM durchgeführt. Elementaranalysen werden durch Atlantic Microlab, Atlanta, GA ausgeführt und liegen innerhalb 0,4% der für jedes Element kalkulierten Werte. Zur Szintiallionsspektrometrie wird ein Beckman 1801 Szintillationszähler verwendet. 0,1%-Rinderserumalbumin ("RSA") und (-)-Kokain wird von Sigma Chemicals gekauft. Alle Reaktionen werden unter inerter (N2-) Atmosphäre ausgeführt.
3H-WIN34,428 (3H-CFT, 2β-Carbomethoxy-3β-(4-fluorophenyl) -N-3H-methyltropan, 79,4-87,0 Ci/mMol) und 3H-Citalopram (86,8 Ci/mMol) wird von DuPont-New England Nuclear (Boston, MA) gekauft. (R)-(-)-Kokain-Hydrochlorid für die pharmakologischen Studien wurde vom National Institute on Drug Abuse (NIDA) geschenkt. Fluoxetin wurde von E. Lilly & Co. geschenkt. HPLC-Analysen wurden auf einem Waters 510-System mit einer Detektion bei 254 nm auf einer Chiralcel OC-Säule (Durchflussrate: 1 mL/min) durchgeführt. - Beispiel 27 6(7)-Hydroxy-2-methoxycarbonyl-8-azabicyclo-(3.2.1)octan-3-on; und 6(7)-Methoxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-3-on;
- Zu einer Lösung von 60 ml Essigsäure und 43 ml Essigsäureanhydrid wurden bei 0°C langsam 40 g (0,27 Mol) Acetondicarboxylsäure zugegeben. Die Mischung wurde gerührt, und die Temperatur wurde auf über 10°C ansteigen gelassen. Die Säure wurde langsam aufgelöst, und es bildete sich ein blassgelbes Präzipitat. Nach 3 h wurde das Produkt filtriert, mit 30 ml eiskalter Essigsäure und 100 ml Benzol gewaschen. Das erhaltene weiße Pulver wurde unter Hochvakuum getrocknet, um 30 g Aceton-Dicarboxylsäureanhydrid (Ausbeute 86%) zu geben. Sp. 137-138°C.
- Zu einem Kolben, der 50 g (0,39 Mol) Acetondicarboxylsäureanhydrid enthielt, wurden 160 ml kaltes trockenes McOH zugegeben. Die Monomethylester-Lösung wurde für 1 h stehen gelassen und filtriert. Das Filtrat von Acetondicarboxylsäuremonomethylester wurde direkt für die folgende Kondensationsreaktion verwendet.
- Zu einem 3L-Kolben mit 53,6 g (0,41 Mol) 2,5-Dimethoxydihydrofuran wurden 1000 ml einer 3N HCl-Lösung zugegeben. Die Mischung wurde 12 h bei Raumtemperatur stehen gelassen und dann mit eiskalter NaOH-Lösung (gleiche Mole) bei 0°C neutralisiert. Zu dieser roten Lösung wurden 41,3 g (0,62 Mol) Methylaminhydrochlorid in 300 ml H2O, die vorgefertigte Methanollösung des Monomethlyesters (50 g (0,39 Mol) des Acetondicarboxylsäureanhydrids in 160 ml Methanol) und 50 g Natriumacetat in 200 mL H2O zugegeben. Die Mischung (pH 4,5) wurde für zwei Tage gerührt, und die Azidität verringerte sich auf pH 4,9. Die rote Lösung wurde mit Hexan (450 ml × 2) zum Entfernen von nichtpolaren Nebenprodukten extrahiert. Die wässrige Lösung wurde zuerst mit NaOH(1N) auf neutralen pH gebracht und dann mit Kaliumcarbonat alkalisiert. Natriumchlorid (etwa 200 g) wurde zugegeben. Die gesättigte Lösung wurde mit CH2Cl2 (250 ml × 8) und dann mit einem Mischlösungsmittel (t-Butyl: 1,2-Dichloroethan, 37:63, 250 ml × 8) extrahiert. Das CH2Cl2-Extrakt wurde über K2CO3 getrocknet, und Lösungsmittel wurde entfernt, um 19,6 g einer Rohmischung zu liefern, die durch Säulenchromatographie (SiO2, 10% Et3N, 30-90% EtOAc in Hexan und 10% Methanol in EtOAc) aufgetrennt, um 7,5 g 6(7)-Methoxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-2-on als einem Öl und 7 g von 6(7)- Hydroxy-2-methoxycarbonyl-8-azabicyclo-(3.2.1)octan-3-on als einem kristallinen Feststoff zu geben.
- Die Mischlösungsmittel-Extrakte wurden getrocknet und in Vakuum abgezogen, um einen blassgelben Feststoff von 19,2 g zu ergeben: 6(7)-Hydroxy-2-methoxy-carbonyl-8-azabicyclo(3.2.1)octan-3-on. Die Hydroxytropanone wurden ohne weitere Reinigung verwendet.

Enol:Keto (1:1). - Beispiel 28 6(7)-Methoxymethoxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-3-one
- Zu einer Lösung von 6(7)-Hydroxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-3-on (18 g, 84,5 mMol) in 200 ml CH2Cl2 wurden 70 ml Dimethoxymethan zugegeben, gefolgt von (18 g, 93 mMol) p-Toluolsulfonsäure-monohydrat. Der Rundkolben war mit einem Soxhlet-Extraktor ausgestattet, der 3-4A-Molekularsiebe enthielt. Die Reaktionsmischung wurde unter Rückfluss mit Rühren erhitzt, bis das Ausgangsmaterial verschwand (TLC). Die Mischung wurde abgekühlt und mit gesättigter Natriumbicarbonatlösung behandelt und mit CH2Cl2 extrahiert. Die vereinigten organischen Phasen wurden über K2CO3 getrocknet, in Vakuum abgezogen und auf eine Säule (Silicagel, 10% Et3N, 30% EtOAc/Hexan) aufgebracht. 6-Methoxymethoxy-2-methoxy-carbonyl-8-azabicyclo(3.2.1)octan-3- on (2,2 g) wurde als ein gelbes Öl erhalten. Rf 0,3 (10% Et3N, 30% EtOAc in Hexan).
1H-NMR (CDCl2, 100MHz): σ 11,7 (s), 4,64 (2 s), 3,76(s), 3,74(s), 3,36(s), 3,35(s), 2,69(s), 2,62(s), 2,41 (s), 4,1-1,8 (m). Enol:Keto (1:2).
4,34 g 7-Methoxymethoxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-3-on wurde als gelbes Öl erhalten. Rf 0,38 (10% Et3N, 30% EtOAc in Hexan).
1H-NMR (CDCl3, 100MHz): σ 11,75(s), 4,67-4,58 (m), 3,81 (S), 3,79(s), 3,39 (s), 3,37 (s), 2,66 (s), 2,60 (s), 2,42 (s), 4,1-1,8 (m). - Eine Mischung der (3.2.1)-Octan-3-one (2,37 g) und des Ausgangsmaterials (4,8 g) wurden aus der Chromatographie erhalten. Ausbeute = 56% auf der Grundlage des wiedergewonnenen Ausgangsmaterials.
- Beispiel 29 2-Carbomethoxy-3-trifluoromethylsulfonyloxy-7-methoxymethoxy-8-azabicyclo(3.2.1)-2-octen
- Zu einer Lösung von 2-Carbomethoxy-7-methoxymethoxy-8-azabicyclo(3.2.1)octanon (4,25 g, 16,5 mMol) in THF (150 mL) wurden Natriumbistrimethylsilylamid (1,OM-Lösung in THF, 25 mL) tropfenweise unter Stickstoff bei -70°C zugegeben. Nach Rühren für 30 min wurden in einer Portion bei -70°C N-Phenyltrifluoromethansulfonimid (7,06 g, 19,8 mMol) zugegeben. Die Lösung wurde auf Raumtemperatur aufwärmen gelassen und über Nacht gerührt. Die flüchtigen Stoffe wurden mit einem Rotationsverdampfer entfernt. Der Rückstand wurde in CH2Cl2 (200 mL) aufgelöst, mit H2O (100 mL) und Salzlösung (100 mL) gewaschen. Die (MgSO4-) getrocknete CH2Cl2-Schicht wurde zur Trockne konzentriert. Der Rückstand wurde durch Flash-Chromatographie (10% Et3N, 20% EtOAc/Hexan) gereinigt, um 3,63 g (65%) 2-Carbomethoxy-3- trifluoromethylsulfonyloxy-7-methoxymethoxy-8-azabicyclo(3.2.1)-2-octen als einem blassgelben Öl zu geben.

- In einen Rundkolben, der 1,1 g 2-Carbomethoxy-7-methoxy-8-azabicyclo(3.2.1)-octan-2-on und 3 ml Et3N in 25 ml CH2Cl2 (wasserfrei) enthielt, wurden 1,22 ml (7,2 mMol) Triflicanhydrid tropfenweise bei 0°C zugegeben. Die Mischung wurde auf Raumtemperatur aufwärmen gelassen und wurde über Nacht gerührt. Das Lösungsmittel wurde entfernt, und NaHCO3 (gesättigt) wurde zugegeben. Die Mischung wurde mit CH2Cl2 extrahiert. Die organischen Phasen wurden vereinigt und getrocknet (K2CO3), in Vakuum entfernt, und das Produkt mittels Säulenchromatographie (SiO2, 10% Et3N, 30% EtOAc, 60% Hexan) aufgetrennt.

- In einen Rundkolben mit 2-Carbomethoxy-3-{[(trifluoromethyl)-sulfonyl]oxy}-7-methoxymethoxy-8-oxabicyclo(3.2.1)-2-octen (1 Äq), LiCl (2 Äq) und Tris (dibenzylidenaceton) dipalladium(0) (5% molekulare Äq) in Diethoxymethan (10 mL) und Na2CO3 (2M-Lösung, 2 Äq) wurden 3,4-Dichlorophenylborsäure (1,3 Äq) zugegeben. Die Mischung wurde unter Rückfluss erhitzt, bis das Ausgangsmaterial verschwand (TLC). Die Mischung wurde auf Raumtemperatur abgekühlt, durch Celite filtriert und mit Ether (100 mL) gewaschen. Die Mischung wurde mit NH4OH alkalisiert und mit Salzlösung gewaschen. Die (MgSO4-) getrocknete Etherschicht wurde zur Trockne konzentriert. Der Rückstand wurde durch Flash-Chromatographie (10% Et3N, 30% EtOAc, 60% Hexan) gereinigt, um Tropen als ein hellgelbes Öl zu ergeben.

- Zu einer THF-Lösung von 2-Carbomethoxy-3-aryl-6 (7)-methoxy-methoxy-8-azabicyclo(3.2.1)-2-octen (1 Äq) mit wasserfreiem Methanol (20 Äq) bei -78°C unter N2 wurden SmI2 (0,1 M-Lösung in THF, 10 Äq) zugegeben. Die Mischung wurde für 4 h bei -78°C gerührt und dann mit H2O (10 mL) abgeschreckt. Nach Aufwärmen auf Raumtemperatur wurde NaHCO3 (gesättigt) zugegeben, und die Mischung wurde durch Celite filtriert und mit Ether (400 mL) gewaschen. Die Etherschicht wurde mit Salzlösung gewaschen. Die (MgSO4-) getrocknete Etherschicht wurde zur Trockne konzentriert. Die Isomeren wurden mittels Gravitätssäule (2-4% Methanol/CH2Cl2) aufgetrennt, um die Wannen- und Sessel-Isomere zu ergeben.

- Die Reaktion von 2-Carbomethoxy-3-3,4-dichlorophenyl)-7-methoxymethoxy-8-azabicyclo(3.2.1)-2-octen (0,494 g, 1,28 mMol) in THF (10 mL) und SmI2 (0,1 M-Lösung in THF, 128 mL, 12,8 mMol) wie oben beschrieben ergab 196 mg (40%) 2β-Carbomethoxy-3α-(3,4-dichlorophenyl)-7-methoxymethoxy-8-azabicyclo(3.2.1)-octan als einem Öl und 114 mg (22%) von 2β-Carbomethoxy-3β-(3,4-dichlorophenyl)-7-methoxymethoxy-8-azabicyclo(3.2.1)octan als einem Öl.

- Die Reaktion von 2-Carbomethoxy-3-(fluorophenyl)-7-methoxy-8-azabicyclo(3.2.1)-2-octen (0,585 g, 1,92 mMol) in THF (10 mL) und SmI2 (0,1 M-Lösung in THF, 192 mL, 19,2 mMol) wie oben beschrieben ergab 230 mg (40%) von 2β-Carbomethoxy-3α-(fluorophenyl)-7-methoxy-8-azabicyclo(3.2.1)octan als einem Öl und 245 mg (42%) von 2β-Carbomethoxy-3β-(fluorophenyl)-7-methoxy-8-azabicyclo(3.2.1)-octan als einem Öl.

- Zu einer Lösung von MOM-geschütztem Alkohol in CH2Cl2 (wasserfrei), enthaltend 4 Å molekulare Siebe bei 0°C, wurden TMSBr (10 Äq) zugegeben. Die Lösung wurde für 1 h bei 0°C gerührt, dann auf Raumtemperatur aufgewärmt. Nach Rühren über Nacht wurde NaHCO3 (gesättigt) zugegeben und mit CH2Cl2 extrahiert. Der Extrakt wurde getrocknet (Na2CO3) und im Vakuum reduziert zum Anwenden auf eine Säule (Silicagel, 10% Et3N, 30-90% EtOAc in Hexan). Das gewünschte Produkt wurde als einem Feststoff erhalten und wurde in einem minimalen Volumen von EtOAc aufgelöst. Zu dieser Lösung wurde etherisiertes HCl (1M, 1,1 Äq) tropfenweise zum Geben des HCl-Salzes hinzugegeben.
- Beispiel 40 2β-Carbomethoxy-3α-(3,4-dichlorophenyl)-7-hydroxy-8-azabicyclo(3.2.1)octan
- 2β-Carbomethoxy-3α-(3,4-dichlorophenyl)-7-methoxymethoxy-8-azabicyclo(3.2.1)octan (49 mg, 0,13 mMol) und TMSBr (0,17 mL, 1,3 mMol) wurden wie oben beschrieben umgesetzt. Das Produkt wurde in 87% Ausbeute (41 mg) erhalten. Rf 0,18 (10% Et3N, 40% EtOAc, 50% Hexan).
- Beispiel 41 2β-Carbomethoxy-3β-(3,4-dichlorophenyl)-7-methoxy-8-azabicyclo(3.2.1)octan
- 2β-Carbomethoxy-3β-(3,4-dichlorophenyl)-7-methoxymethoxy-8-azabicyclo(3.2.1)octan (50 mg, 0,13 mMol) und TMSBr (0,17 mL, 1,3 mMol) wurden wie oben beschrieben umgesetzt. Das Produkt wurde in 88% Ausbeute (42, 1 mg) erhalten. Rf 0,18 (10% Et3N, 40% EtORc, 50% Hexan).

- Biologische Tests wurden unter Verwendung der folgenden Prozeduren ausgeführt.
- A. Gewebequellen und -präparationen.
- Hirngewebe von erwachsenen männlichen und weiblichen Cynomolgus-Affen (Macaca fasicularis) wird bei -85°C in der Primatenhirnbank im New England Regional Primate Research Center gelagert. Das Caudate-Putamen wird aus koronalen Schnitten sektiert und ergibt 1,4 ± 0,4 g Gewebe. Membranen werden wie zuvor beschrieben präpariert. Kurz gesagt wird das Caudate-Putamen in 10 Volumina (w/v) eiskaltem Tris-HCl-Puffer (50 mM, pH 7,4 bei 4°C) homogenisiert und bei 3800 × g für 20 min in Kälte zentrifugiert. Das resultierende Pellet wird in 40 Volumina Puffer suspendiert, und die gesamte Prozedur wird zweimal wiederholt. Die Membransuspension (25 mg ursprüngliches Gewebe-Nassgewicht/ml) wird auf 12 ml/ml für den 3H-WIN 35,428- oder den 3H-Citalopram-Test im Puffer kurz vor dem Test verdünnt und mit einem Brinkmann-Polytron-Homogenisierer (Einstellung #5) für 15 s dispergiert. Alle Experimente werden im Triplikat ausgeführt, und jedes Experiment wird jeweils 2-3 Präparationen aus einzelnen Hirnen wiederholt.
- B. Dopamintransportertest.
- Der Dopamintransporter wird mit 3H-WIN35,428 (70-85 Ci/mMol, DuPont-NEN) markiert. Die Affinität der neuen Verbindungen mit dem Dopamintransporter wird in Experimenten bestimmt, indem Gewebe mit einer festgelegten Konzentration von 3H-WIN35,428 und einem Bereich einer Konzentration der Verbindung wie zuvor beschrieben inkubiert wird (Madras, 1989, #21). Stammlösungen werden seriell im Testpuffer verdünnt und zum Testmedium hinzugegeben (0,2 mL). Die Teströhrchen nehmen die folgenden Bestandteile bei einer Endtestkonzentration in Tris-HCl-Puffer (50 mM, ph 7,4 bei 0-4°C; NaCl 100mM) auf: Wirkstoff (0,3 ml; 1 pM-300μM, je nach Affinität), 3H-WIN35,428 (0,2 mL; 0,3 oder 1 nM); Membranpräparation (0,2 mL; 1-4 mg ursprüngliches Gewebe-Nassgewicht/ml) je nach Test. Die 2 h-Inkubation (0-4°C) wird durch Zugabe von Membranen begonnen und durch schnelle Filtration über Whatman GF/B-Glasfaserfilter, die in 0,1%-igem Rinderserumalbumin (Sigma Chem. Co.) eingetaucht wurden, beendet. Die Filter werden zweimal mit 5ml Tris-HCl-Puffer (50 mM) gewaschen, über Nacht bei 0-4°C in Szintillationsfärbung (Beckman Ready-Value, 5ml) inkubiert, und die Radioaktivität (dpm) wird durch Flüssigszintillationsspektrometrie (Beckman 1801) gemessen. Die Gesamtbindung ist definiert als 3H-WIN35,428, der in Gegenwart unwirksamer Konzentrationen des Wirkstoffs gebunden ist. Nicht-spezifische Bindung ist definiert als 3H-WIN35-428, das in Gegenwart eines Überschusses (30 μM) von (-)-Kokain oder Mazindol (1 μM) gebunden ist. Spezifische Bindung ist der Unterschied zwischen den beiden Werten.
- C. Serotonintransportertest.
- Der Serotonintransporter wird durch 3H-Citalopram (spezifische Aktivität- 82 Ci/mMol, DuPont-NEN) markiert. Der Serotonintransporter wird in Caudate-Putamen-Membranen unter Bedingungen, die mit denen für den Dopamintransporter ähnlich sind, getestet. Der Serotonintransporter wird bei relativ hoher Dichte im Caudate-Putamen (20 pMol/g) exprimiert, und die Affinität von 3H-Citalopram beträgt ungefähr 2 nM. Wirkstoffaffinitäten werden bestimmt, indem Gewebe mit einer festgelegten Konzentration von 3H-Citalopram und einem Konzentrationsbereich der Testverbindungen inkubiert wird. Die Teströhrchen nehmen die folgenden Bestandteile bei Endtestkonzentrationen in Tris-HCl-Puffer (50 mM, pH 7,4 bei 0-4°C; NaCl 100 mM) auf: Wirkstoff (0,2 ml von unterschiedlichen Konzentrationen); 3H-Citalopram (0,2 ml; 1 nM); Membranpräparation (0,2 ml; 4 mg ursprüngliches Gewebe/Nassgewicht ml). Die 2 h-Inkubation (0-4°C) wird durch Zugabe von Membranen begonnen und durch schnelle Filtration über Whatman GF/B-Glasfaserfilter, die in 0,1% Polyethylenimin eingetaucht wurden, beendet. Die Filter wurden zweimal mit 5 ml Tris-HCl-Puffer (50 mM) gewaschen, und die verbleibenden Schritte werden wie oben beschrieben durchgeführt. Gesamtbindung ist definiert als 3H-Citalopram, das in Gegenwart unwirksamer Konzentrationen von unmarkiertem Citalopram (1pM) oder den Testverbindungen gebunden ist. Nicht-spezifische Bindung ist definiert als 3H-Citalopram, das in Gegenwart eines Überschusses (10μM) von Fluoxetin gebunden ist. Die spezifische Bindung ist der Unterschied zwischen den beiden Werten.
- D. Norepinephrintransportertest.
- Die Wahl des Thalamus beruht auf einer vorangehenden Autoradiografiestudie, die besagt, dass diese Hirnregion eine hohe Dichte an 3H-Nisoxetin aufweist. Die Testbedingungen für Thalamus-Membranen (Madras 1996) sind ähnlich zu denen für den Serotonintransporter. Die Affinität von 3H-Nisoxetin (spezifische Aktivität: 74 Ci/mMol, DuPont-NEN) für den Norepinephrintransporter wird in Experimenten bestimmt, indem Gewebe mit einer festgelegten Konzentration von 3H-Nisoxetin und einem Konzentrationsbereich von unmarkiertem Nisoxetin inkubiert wird. Die Teströhrchen nehmen die folgenden Bestandteile bei einer Endtestkonzentration auf: Nisoxetin oder Wirkstoff (0,2 ml; 1pM-300μM), 3H-Nisoxetin (0,2ml; 0,6nM); Membranpräparation (0,2ml; 4mg ursprüngliches Gewebe-Nassgewicht/ml). Der Puffer im Testmedium ist Tris-HCl: 50mM, ph 7,4 bei 0-4°C; NaCl 300mM. Die 16 h-Inkubation bei 0-4°C wird durch Zugabe von Membranen begonnen und durch schnelle Filtration über Whatman GF/B-Glasfaserfilter, die in 0,1% Polyethylenimin eingetaucht wurden, beendet. Die verbleibenden Schritte sind oben beschrieben. Gesamtbindung ist definiert als 3H-Nisoxetin, das in Gegenwart unwirksamer Konzentrationen des Wirkstoffs gebunden ist. Nichtspezifische Bindung ist definiert als 3H-Nisoxetin, das in Gegenwart eines Überschusses (10μM) von Desipramin gebunden ist. Spezifische Bindung ist der Unterschied zwischen den Beiden Werten.
- E. Datenanalyse.
- Daten werden durch EBDA- und LIGAND-Computersoftware (Elsevier-Biosoft, UK) analysiert. Endschätzungen von IC50- und nH-Werten werden durch das EBDA-Programm errechnet. Basislinienwerte für die einzelnen Wirkstoffe werden durch Computeranalyse unter Verwendung der Basislinienwirkstoffe als Vorgabe geschaffen. Das LIGAND-Programm liefert Endparameterabschätzungen der neuen Verbindungen durch iterative, nichtlineare Kurvenanpassung und Beurteilung von Ein- oder Zwei-Komponenten-Bindungsmodellen.
- Die vorliegende Erfindung ist im Detail beschrieben worden, einschließlich der bevorzugten Ausführungsformen davon. Es dürfte jedoch klar sein, dass der Fachmann bei Betrachtung der vorliegenden Offenbarung Modifikationen und/oder Verbesserungen dieser Erfindung vornehmen kann und sich weiterhin innerhalb des Rahmens der Erfindung, wie in den nachfolgenden Ansprüchen festgelegt, befindet.

- Verwendung einer Verbindung mit der Strukturformel I, II oder III zur Herstellung eines Pharmazeutikums zur Hemmung der Dopamin-Wiederaufnahme eines Dopamintransporters in einem Säugetier: worin: R1 = COOCH3, Niedrigalkyl, Niedrigalkenyl, Niedrigalkynyl, CONHR4, oder COR6; R2 = ein 6α-, 6β-, 7α- oder 7β-Substituent, der aus OR3, F, Cl, Br und NHR3 ausgewählt werden kann; X = NR3 oder NSO2R3; R3 = H, CH3, CH3CH2, CH3(CH2)n, (CH2)nC6H4Y, CHCH2, Niedrigalkyl, Niedrigalkenyl oder Niedrigalkynyl; Y = H, Br, Cl, I, F, OH, OCH3, CF3, NO2, NH2, CN, NHCOCH3, N(CH3)2, (CH2)nCH3, COCH3 oder C(CH3)3; R4 = CH3, CH2CH3 oder CH3SO2; Ar = Phenyl-R5, Naphthyl- R5, Anthracenyl-R5, Phenanthrenyl-R5 oder Diphenylmethoxy-R5, R5 = Br, Cl, I, F, OH, OCH3, CF3, NO2, NH2, CN, NHCOCH3, N(CH3)2, (CH2)nCH3, COCH3, C(CH3)3, mit n = 0-6, 4-F, 4-Cl, 4-I, 2-F, 2-Cl, 2-I, 3-F, 3-Cl, 3-I, 3,4-diCl, 3,4-diOH, 3,4-diOAc, 3,4-diOCH3, 3-OH-4-Cl, 3-OH-4-F, 3-Cl-4-OH, 3-F-4-OH, Niedrigalkyl, Niedrigalkoxy, Niedrigalkenyl, Niedrigalkynyl, CO(Niedrigalkyl) oder CO(Niedrigalkoxy); R6 = Morpholinyl oder Piperidinyl; m = 0 oder 1; n = 0, 1, 2, 3, 4 oder 5; und mit der Ausnahme, daß wenn X = N, R1 nicht COR6 ist.
- Verwendung von Anspruch 25, wobei die Verbindung ausgewählt wird aus der Gruppe, die besteht aus: ah. 6-Hydroxy-2-methoxycarbonyl-8-azabicyclo-(3.2.1)octan-2-on; ai. 7-Hydroxy-2-methoxycarbonyl-8-azabicyclo-(3.2.1) octan-2-on; aj. 6-Methoxy-2-methoxycarbonyl-8-azabicyclo-(3.2.1)octan-2-on; ak. 7-Methoxy-2-methoxycarbonyl-8-azabicyclo-(3.2.1)octan-2-on; al. 6-Methoxymethoxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-2-on; am. 7-Methoxymethoxy-2-methoxycarbonyl-8-azabicyclo(3.2.1)octan-2-on; an. 2-Carbomethoxy-3-trifluoromethylsulfonyloxy-7-methoxymethoxy-8-azabicyclo (3.2.1)-2-octen; ao. 2-Carbomethoxy-3-(trifluoromethyl)sulfonyloxy-6-methoxymethoxy-8-azabicyclo(3.2.1)-2-octen; ap. 2-Carbomethoxy-3-trifluoromethylsulfonyloxy-7-methoxy-8-azabicyclo(3.2.1)-2-octen; aq. 2-Carbomethoxy-3-(3,4-dichlorophenyl(-7-methoxymethoxy-8-azabicyclo(3.2.1)-2-octen; ar. 2-Carbomethoxy-3-(4-fluorophenyl)-7-methoxymethoxy-8-azabicyclo(3.2.1)-2-octen; as.
