Description
The present invention relates to a modified cereal flour and cereal flour-based processed foods prepared by using the modified cereal flour. More specifically, the present invention relates to a modified cereal flour prepared by the action of transglutaminase during the process of preparing cereal flour such as wheat flour from cereal grains such as wheat as a raw material, and various cereal flour-based processed foods including bread, noodles such as macaroni and spaghetti, Chinese noodles (including the outer shell of ravioli, wang-tang and the like), Japanese noodles such as udon and soba, tempura, oil-fried breads such as doughnuts, cakes, snacks, fresh pastries and Japanese-style pastries.
Beispiele sindExtensive research has been carried out in the state of the art regarding the modification of wheat flour for use in bread, pastries and cakes. Examples are
1. ein Verfahren, welches Weizenmehl in einer Atmosphäre von Kohlendioxidgas und Ethanol bei 40ºC oder höher hält (Japanische Patentveröffentlichung (Kokoku) Nr. 6-36725);1. a method of keeping wheat flour in an atmosphere of carbon dioxide gas and ethanol at 40ºC or higher (Japanese Patent Publication (Kokoku) No. 6-36725);
2. ein Verfahren, welches das Zugeben von 40 bis 500 Gew.-% Wasser zu Weizen als Ausgangsmaterial, das Trocknen des erhaltenen Weizens bei einer Temperatur, bei der keine Denaturierung auftritt, um Weizenmehl für Torten herzustellen (Japanische Patentveröffentlichung (Kokoku) Nr. 5-4055); und 3. ein Verfahren, welches das Zugeben eines Oxidationsmittels und Wasser in Weizenmehl umfaßt, um Gluten mit hervorragender Verarbeitbarkeit für verarbeitete Lebensmittel zu gewinnen (Japanische Patentveröffentlichung (Kokoku) Nr. 6-34682), und dergleichen.2. a process which comprises adding 40 to 500 wt% of water to wheat as a raw material, drying the resulting wheat at a temperature at which no denaturation occurs to produce wheat flour for cakes (Japanese Patent Publication (Kokoku) No. 5-4055); and 3. a method which comprises adding an oxidizing agent and water into wheat flour to obtain gluten having excellent processability for processed foods (Japanese Patent Publication (Kokoku) No. 6-34682), and the like.
In addition, methods have been reported in which wheat flour is modified using transglutaminase (hereinafter sometimes abbreviated as "TG"), which is an enzyme that catalyzes the acyl transfer of the γ-carboxyamide group in a glutamine residue in peptides. For example, a method comprising adding a predetermined amount of TG into commercially available wheat flour for cakes to produce wheat flour having excellent taste and texture for cakes (Japanese Patent Laid-Open (Kokai) No. 2-286031) and a method comprising adding TG into commercially available wheat flour to produce dough having good elasticity for bread making (USP No. 5,279,839) have been reported.
Each of the above methods is excellent in some respects. However, as described below, no method is known that overcomes the problems specific to wheat flour.
Genauer gesagt gibt es die folgenden Probleme:More specifically, there are the following problems:
1. Beeinträchtigung, wie durch Oxidation des Weizenmehls, welche nach Lagerung über einen längeren Zeitraum auftritt;1. Deterioration, such as oxidation of the wheat flour, which occurs after storage over a long period of time;
2. Weizenmehl verursacht Allergien;2. Wheat flour causes allergies;
The preferred taste and texture cannot be transferred to the final products (processed foods made from flour), such as bread, pastries, cakes, pasta and the like.
Therefore, it is an object of the present invention to provide a new cereal flour, for example wheat flour, which (1) does not suffer from deterioration such as oxidation after storage for a long period of time and (2) which shows little allergenic effect and (3) which can impart the preferred taste and texture to end products such as bread, pastries, cakes, pasta and the like.
The inventors of the present invention have conducted studies to solve the above-mentioned problems. They have found that the problems can be overcome by a transglutaminase treatment in which the action of transglutaminase is exerted during the process for producing grain flour such as wheat flour which can be used as a material for processed products made from grain flour. Based on these findings, the present invention has been completed.
More specifically, the present invention relates to a modified cereal flour obtained by transglutaminase treatment in (1) the process for dressing the cereal and/or (2) the milling process for producing the cereal flour from the cereal grains as a raw material, and further relates to processed cereal flour-based foods using the modified cereal flour as a raw material.
As described above, examples in which transglutaminase is used to modify wheat flour are already known. However, in these examples, transglutaminase is used to treat the raw materials, such as wheat flour milled from wheat, for processed foods based on wheat flour, these examples are completely different from the present invention because transglutaminase exerts its effect in the present invention during the milling process. Needless to say, the use of transglutaminase during milling is surprisingly more effective.
According to the present invention, the above-mentioned various problems can be overcome, for example, by exerting the action of transglutaminase during the process of producing wheat flour. In addition, the gelling capacity, viscosity and water-retaining capacity of wheat protein contained in the wheat flour can be improved, thereby providing a modified wheat flour of high quality as a raw material for bread, tempura, cakes, baked breads such as doughnuts and batter.
The present invention is described in detail below.
Der hierin verwendete Ausdruck "Getreidemehl" bedeutet jegliches Getreidemehl aus Weizen, Gerste, Mais, Japanisches Soba-Ko, Roggen, Hafer, Chinesische Hirse und Sojabohnen oder ein Gemisch von Getreidemehl aus zwei oder mehr dieser Getreidemehle in Kombination, Diese Getreidemehle werden im allgemeinen zur Herstellung von Broten, Pasta, wie Makkaroni und Spaghetti, Chinesischen Nudeln (einschließlich der Umhüllung von Gyoza, Wang-Tang und dergleichen), Japanischen Nudeln, wie Udon und Soba, Tempura, ölgebackenen Broten, wie Doughnuts, Kuchen, Snacks, frischem Gebäck und Gebäck nach Japanischer Art eingesetzt. The term "cereal flour" as used herein means any cereal flour made from wheat, barley, corn, Japanese soba-ko, rye, oats, Chinese millet and soybeans or a mixture of cereal flours made from two or more of these cereal flours in combination. These cereal flours are generally used in the manufacture of breads, pastas such as macaroni and spaghetti, Chinese noodles (including the wrapping of gyoza, wang-tang and the like), Japanese noodles such as udon and soba, tempura, oil-fried breads such as doughnuts, cakes, snacks, fresh pastries and Japanese-style pastries.
The transglutaminase used for producing the modified grain flour according to the present invention is an enzyme that catalyzes the acyl transfer reaction of the γ-carboxyamide group in the glutamine residue in peptides. When the transglutaminase acts on the ε-amino group of the lysine residue in a protein as an acyl receptor, ε-(γ-Glu)Lys bonds are formed intramolecularly and intermolecularly, thereby promoting the cross-linking reaction of the protein in the wheat flour as a raw material for producing the processed food, so that a wheat flour having properties such as higher gel formation capacity, higher viscosity and higher water retention capacity can be obtained.
For example, the detailed properties of the enzyme (transglutaminase) derived from microorganisms are described in Japanese Patent Laid-Open (Kokai) No. 64-27471.
Transglutaminase is classified into calcium-independent type and calcium-dependent type, and each of these types is satisfactorily used for the production of the modified grain flour. Examples of the former include those from microorganisms (see, for example, Japanese Patent Laid-Open (Kokai) No. 64-27471). Examples of the latter include those from guinea pig liver (see, for example, Japanese Patent Publication (Kokoku) No. 1-50382), from animal blood (also called "factor XIII") and from fish (see, for example, Journal of Japan Fisheries Academy, Vol. 56, pp. 125-132 (1990)) and Proceedings of Nippon Fishery Federation, 1994, p. 219). In addition, there may be mentioned those produced by the method of gene engineering (see Japanese Patent Publication (Kokai) Nos. 1-300889 and 5-199883), and any of these transglutaminases can be produced satisfactorily without any limitation with respect to regardless of the origin and the manufacturing process.
For example, in the case of producing wheat flour, the amount of transglutaminase to be added (added) in the process for producing the modified grain flour is 0.01 to 100 units (U), preferably 0.1 to 50 units (U) per 1 g of the protein in the raw material wheat. If the amount to be added is less than the above range, the gelling capacity is reduced. In the case where the obtained modified wheat flour is used for making bread, the obtained bread also becomes too soft and is therefore not preferable. And if the amount is more than the above range, the obtained bread becomes too hard so that the preferable taste and feel of the bread are impaired. Even if the types of the raw material wheat are different, the range of the amount of transglutaminase to be added is the same.
For the purposes of the present invention, commercially available enzyme formulations containing transglutaminase as a main component (for example, "Activa" (specific activity of 1 U/ml), manufactured by Ajinomoto Co., Inc.) can be used.
In the present invention, the unit activity of the transglutaminase is determined and defined as follows. More specifically, after the transglutaminase reacts with benzyloxycarbonyl-L-glutaminylglycine and hydroxylamine as substrates to form an iron complex with the obtained hydroxamic acid in the presence of chloroacetic acid, the absorbance at 525 nm is measured. In this way, a calibration curve can be prepared based on the amount of hydroxamic acid, and the amount of enzyme which forms 1 µmol of hydroxamate per minute is defined as 1 unit (U) of the transglutaminase activity.
A detailed description of the test is disclosed in Japanese Patent Laid-Open (Kokai) No. 64-27471.
When the transglutaminase treatment is carried out in the presence of a foreign protein and/or a partial protein hydrolysate, the effect of the present invention can be enhanced so that the obtained wheat flour has additionally increased elasticity and viscosity in a kneaded mixture with water.
The term "foreign protein" means protein added from the outside intentionally for the transglutaminase treatment, excluding the protein naturally contained in the cereals to be milled. Such proteins include milk protein such as casein, sodium caseinate, calcium caseinate, whole milk powder and skim milk powder, wheat protein, gelatin, collagen, corn protein such as zein, rice protein, soybean protein such as separated soybean protein, extracted soybean protein and soybean whey protein, egg protein such as egg white albumin, milk whey protein and the like.
In addition, the partial protein hydrolysate includes partially hydrolyzed products of a protein to be used as a foreign protein, such as partially hydrolyzed products of wheat protein, milk protein and the like.
The partial protein hydrolysates which can be obtained by partially hydrolyzing the proteins, such as wheat protein and milk protein, by enzymes, acids and bases according to standard procedures can be used.
There is no particular limitation as long as the partial protein hydrolysate satisfies the objects of the present invention.
Commercially available peptides such as lysine peptide and glutamine peptide can also be used as partial protein hydrolysate. hydrolysate having the same effects as described above, which are different from the partial protein hydrolysate from natural protein. Therefore, the partial protein hydrolysate used in the present invention includes peptides such as lysine peptide composed of a single amino acid.
The average molecular weight of the partial protein hydrolysate used in the present invention is generally about 600 to 40,000, preferably about 3,000 to 20,000, and more preferably about 6,000 to 16,000.
The amount of the foreign protein and/or the partial protein hydrolysate to be added (used) is 0.001 to 2.0 g, preferably 0.01 to 1.0 g per 1 g of the wheat protein when the starting material cereal grain is wheat. The amount to be added does not vary depending on whether the foreign protein or partial protein hydrolysate is used singly or in combination of both the foreign protein and the partial protein hydrolysate (the total amount of these two is set to the above-mentioned range). If the amount to be added is less than 0.001 g, the noodles made by using the obtained wheat flour do not have improved softness and elasticity; or the wrappers of gyoza (ravioli), wang-tang and the like cannot be improved. If the amount exceeds 2.0 g, the noodles and the wrappers of gyoza (ravioli), wang-tang and the like obtained using the obtained wheat protein become disadvantageously brittle and exhibit non-viscous texture. In any case, the objects of the present invention cannot be satisfactorily achieved.
A process for producing the modified cereal flour according to the invention from cereal grains by simultaneously using the transglutaminase described above and, if desired, a foreign protein and/or a partial protein hydrolysate, is described in detail below.
First, the concept of grinding is described for deeper understanding.
It is known that milling is carried out as follows: Grains of cereals with an outer shell composed of soft sugar layers and with an inner hard endosperm, such as rice grains, are peeled from the outside to separate the edible part (peeling of rice). On the other hand, grain flour with a very hard outer layer but with a brittle and easily destructible endosperm and, in addition, with elongated furrows in the center of the grain is pulverized to separate the layered part. Such a process, called milling, is carried out. In order to effectively utilize the gluten specific to wheat, wheat is necessarily prepared as wheat flour. The milling process is summarized below.
A process for removing microfine contaminating materials, such as sand, because it is difficult to remove the impurities from the obtained product of wheat flour, the selection of the raw material wheat should be carried out very carefully.
For the purposes of strengthening the layers, easily separating the endosperm part and softening the endosperm part for easier milling, water is added to wheat, which is then left to soak for 24 to 48 hours. In addition, wheat raw materials that have been separately soaked are mixed together, if necessary, depending on the task.
The layer of dressed wheat is removed as much as possible by means of a roller to obtain raw grains of the endosperm (breaking process). Then the raw grains are sieved and transferred to a grain cleaning device to remove the contaminating solids by a combination of selection by sieving and air flow (cleaning process). In addition, the cleaned raw grain is milled by a smooth roller (smooth surface or rough surface) and then the flour is separated by sieving based on the grain size (pulverizing process).
After sifting, the wheat flours of different sizes (final product flour, agari-ko in Japanese) are mixed together depending on their properties to produce a wheat flour of the desired quality and degree of purification.
A final product is then prepared by mixing. In order to add vitamins and the like to the product, vitamins and the like should then be mixed into the product (see "General Dictionary of Food Industry", new edition, edited by Korin K. K., 1993). Needless to say, such grinding can be applied to other grains (cereals) with the same structure as wheat.
The transglutaminase treatment during the milling process will now be described using wheat as an example. Except for this treatment, modified wheat flour according to the invention can be produced from raw material wheat by known milling processes.
The preferable protein content of wheat flour varies depending on its use. In addition, the protein content of wheat flour varies depending on the type of raw material wheat. Wheat flour is divided into high-gluten wheat flour (level I; Kyoriki-Ko in Japanese), semi-high-gluten wheat flour (level II; Jun-Kyoriki-Ko in Japanese), moderately high-gluten wheat flour (level III; Churiki-Ko in Japanese), low-gluten wheat flour (level IV; Hakuriki-Ko), durum semolina flour, and the like. In the present invention, any wheat flours can be effectively modified.
The transglutaminase treatment during the exposure procedure can be carried out, for example, as follows.
When Standard Canada Western Red Spring wheat is selected as the semi-high gluten wheat flour (Stage II), transglutaminase is added at 0.01 to 100 U, preferably 0.1 to 50 U per 1 g of the protein of the starting material wheat during the water addition in the soaking process together with a foreign protein and/or a partial protein hydrolysate of 0.001 to 2.0 g, preferably 0.01 to 1.0 g total per 1 g of the wheat protein, if desired. The amount of water to be added is not particularly limited, but water is added in such an amount that the final water content in the wheat is about 10 to 20%, preferably about 12 to 16%. Thereafter, the treatment is generally carried out at 0 to 60°C, preferably 10 to 30°C, for 16 to 50 hours, so that transglutaminase penetrates from the surface of the wheat grain through the germ into the interior to cause cross-linking of gluten in the wheat protein. By this treatment, the endosperm is pulverized, while the epidermis moderately absorbs water and also becomes hard due to the action of the transglutaminase, so that the resulting grain becomes so brittle that it can be easily broken.
In addition, gluten is cross-linked in the protein inside the endosperm, so that a wheat flour with properties such as elasticity and the ability to impart flexibility to the resulting noodles can be obtained.
The wheat flour obtained by such transglutaminase treatment is the modified wheat flour with improved performance.
The transglutaminase treatment in the milling process is carried out, for example, as follows.
Water is added to the raw material wheat after soaking and mixing for 1 to 3 hours before the milling process.
Transglutaminase and, if desired, a foreign protein and/or a partial protein hydrolysate are dissolved or dispersed in the water to be added during the water addition. The wheat to which transglutaminase and, if desired, the foreign protein and/or the partial protein hydrolysate are added is subjected to a process for separating the endosperm from the epidermis (breaking process), and thereafter standard milling processes are carried out to obtain the modified wheat flour of the invention. The amount of the transglutaminase to be added is 0.01 to 100 U, preferably 0.1 to 50 U per 1 g of the protein of the raw material wheat, and the amount of the foreign protein and/or the partial protein hydrolysate to be added is 0.001 to 2.0 g, preferably 0.01 to 1.0 g per 1 g of the protein of the raw material wheat in total.
The transglutaminase treatment is carried out either during the mixing process and the milling process as described above, or in some cases the treatment can be carried out both during the mixing process as well as during the grinding process.
In addition, although the transglutaminase treatment can be carried out in a process step after the milling process, the effect of this treatment is weak. For example, the transglutaminase treatment of wheat flour (final product flour) is carried out in mixing or in the final treatment after the milling process as follows.
Transglutaminase is dissolved in an amount of 0.01 to 100 U, preferably 0.1 to 50 U per 1 g of the protein in the wheat flour in a suitable amount of water beforehand. If desired, a foreign protein and/or the partial protein hydrolyzate after the milling process is added in an amount of 0.001 to 2.0 g, preferably 0.01 to 1.0 g per 1 g of the wheat protein together with transglutaminase and then dissolved or dispersed. Thereafter, the resulting solution is sprayed over the wheat flour by a pulverizing process (last stage of the milling process), and the flour is treated at 5 to 35°C for 15 minutes to 48 hours, preferably 1 to 24 hours. During this treatment process, transglutaminase exerts its effect. The wheat flour is then dried in air and finished.
Such a method of modifying wheat flour according to the invention is clearly different from the known methods using transglutaminase, as described, for example, in Japanese Patent Laid-Open (Kokai) No. 2-286031 and USP No. 5,279,839, in that transglutaminase exerts its effect in the milling process for producing wheat flour from wheat as an agricultural product.
A processed food based on cereal flour using the modified cereal flour according to the invention is described below.
The process for producing the cereal flour-based food using the modified cereal flour of the present invention can be carried out according to conventional processes for producing processed foods from cereal flour, except that the modified cereal flour of the present invention is used as a starting material.
For example, a loaf of bread is prepared as follows. Yeast, yeast nutrients and water are kneaded into the modified wheat flour of the present invention as a raw material with a mixer. Thereafter, the kneaded mixture is kept at 20 to 40°C for 20 minutes to 10 hours for a first fermentation to obtain a first dough. Water, table salt, sugar, oil, skim milk powder and the like are then added and mixed with the first dough to prepare the bread dough. The bread dough is appropriately divided into portions, which are then left to stand at 20 to 40°C for a predetermined time to form the network structure of the wheat gluten (fermentation), and they are then put into a baking pan. Thereafter, fermentation is carried out again. The total fermentation time is about 20 minutes to 12 hours. After the fermentation is complete, the dough is baked in an oven at 180 to 250ºC.
The loaf of bread obtained by baking is excellent in that the bread can maintain its good taste and properties (such as the ability to retain its shape) even during long-term storage.
It is understood that a direct kneading process, which involves kneading the modified wheat flour, yeast and other raw materials prior to the above-mentioned first fermentation, besides the preparation of the first dough, is sufficient to produce a loaf of bread. The first fermentation is carried out by keeping the kneaded product at a temperature of 20 to 45ºC for 30 minutes to 10 hours. A subsequent fermentation process is carried out under general fermentation conditions for making bread. The fermentation conditions are not particularly limited. If necessary, the bread dough may be fermented again after being allowed to stand at a temperature of 20 to 45ºC.
In addition, instead of baking, steamed bread can be obtained by heating in steam. It goes without saying that the heating conditions for heating in steam are according to conventional methods.
Zum Herstellen eines Laibes Brot werden andere Bestandteile routinemäßig eingesetzt, beispielsweise Trockenmilchpulver, Eier, Polysaccharid, Früchte, Kaffee-Extraktkomponenten, Gewürze, Aromastoffe, Zusätze, wie Ascorbinsäure, Quellmittel (Ammoniumhydrogencarbonat, Natriumhydrogencarbonat usw.), Bleichmittel (Ammoniumperschwefelsäure, Kaliumbromat usw.), Qualitätsmodifikationsmittel (Calciumstearoyllactat, L-Cysteinhydrochlorid usw.), Emulgiermittel (Glycerinfettsäureester, Saccharosefettsäureester usw.), welche zusätzlich zu modifiziertem Weizenmehl, Hefe, Hefenährstoffe, Wasser, Kochsalz, Zucker und Fett und dergleichen als Ausgangsmaterial eingesetzt werden können. In making a loaf of bread, other ingredients are routinely used, such as dry milk powder, eggs, polysaccharide, fruits, coffee extract components, spices, flavorings, additives such as ascorbic acid, swelling agents (ammonium hydrogen carbonate, sodium hydrogen carbonate, etc.), bleaching agents (ammonium persulfuric acid, potassium bromate, etc.), quality modifiers (calcium stearoyl lactate, L-cysteine hydrochloride, etc.), emulsifiers (glycerol fatty acid esters, sucrose fatty acid esters, etc.), which may be used in addition to modified wheat flour, yeast, yeast nutrients, water, table salt, sugar and fat, and the like as raw materials.
It goes without saying that baked quick breads such as doughnuts are prepared by conventional methods using the modified wheat flour of the present invention as the main component. Secondary raw materials are used without specific limitation, and therefore Sugar, egg, egg white, cream, butter, milk, flavorings, table salt, spices and the like are used as appropriate if necessary. The baked quick breads such as doughnuts produced using the modified wheat flour of the present invention taste very good.
It goes without saying that the production of cakes such as sponge cakes using the modified cereal flour of the present invention is carried out by conventional methods. Secondary raw materials are used without any specific limitation, and therefore egg, sugar, milk, butter and the like which are usually used are suitably used. It goes without saying that the production of the sponge cake can be carried out using general equipment.
For example, foamed meringue is prepared by foaming the mixture obtained by adding sugar to egg yolks using a Hobart mixer at low speed for good mixing, by simultaneously adding egg whites and sugar to the resulting mixture.
Then, the modified wheat flour of the invention is added and mixed with the meringue, and then melted butter is added if necessary, and then stirring is continued at low speed to mix the components. The dough is put into a pan, then baked at 160 to 180°C for about 30 minutes, thereby producing a sponge cake. The sponge cake obtained using the modified wheat flour of the invention tastes good and is elastic.
It goes without saying that the production of Japanese-style pastries using the modified wheat flour according to the invention can be carried out according to general procedures When the Japanese-style pastry is a bean pastry covered with a cherry blossom petal (sakura mochi), water is added to the modified wheat flour along with sugar and rice flour as secondary raw materials, followed by mixing and kneading to make a dough. Then, boiled, mashed and pressed azuki beans in jam (koshi-an Japanese) are covered with a coating made from the dough, and the resulting stuffed material is rolled with a salted cherry blossom petal to make sakura mochi. The sakura mochi that uses the modified wheat flour as the main raw material can maintain its softness even after a long time, and it tastes good.
It goes without saying that the production of pasta, such as spaghetti and macaroni, Japanese noodles, such as udon and soba, Chinese noodles (including wrappers of gyoza, wang-tang and the like) can be satisfactorily carried out by conventional methods, except that the modified grain flour of the present invention is to be used instead of conventional grain flour.
For pasta, for example, water is added to modified wheat flour and egg powder for routine use as a second raw material and kneaded to make pasta dough. The dough is then kept at a predetermined temperature (so-called "ripening process"), after which a first rolling, assembling and pressure rolling are carried out, and the obtained dough is finally cut into the desired width and the desired length to make pasta strips. The pasta strips are cooked in water together with a small amount of table salt, and the obtained pasta can have the desired hardness, the desired crispness and the desired elasticity. This is due to the formation a network structure in the gluten of wheat through the action of transglutaminase possible.
It goes without saying that the modified wheat flour of the present invention can also be used for the production of noodles other than pasta, for example, Japanese noodles, including udon and soba, Chinese noodles, and the like. The production of these noodles can be carried out by conventional methods, except that the modified grain flour is used instead of the conventional grain flour.
The present invention is described in detail below using the examples.
Es wurden Versuche unternommen, ein modifiziertes Weizenmehl aus kanadischem Weizen als Ausgangsmaterial herzustellen (Canada Western Red Spring Species; 9 kg).Attempts were made to produce a modified wheat flour using Canadian wheat as the starting material (Canada Western Red Spring Species; 9 kg).
First, microfine sand particles, microfine iron particles and the like were removed from the raw material wheat (selection method). Then, the wheat thus selected was placed in a container to which water was added until the final water content in the wheat was about 14.7%. Then, the wheat was kept at 25ºC for 24 hours (mixing method). Transglutaminase was previously added to the water to be added and dissolved therein to a final concentration of 5 U per 1 g of the wheat protein.
After dressing, the wheat husk was removed to obtain raw endosperm grains (breaking process). Then, the contaminating husk material was separated from the raw endosperm grain by a combination of sieving and selection (cleaning process). In addition, the cleaned raw grain was ground with a smooth roller and sieved according to the size of the grain (pulverization process), obtaining 7 fractions depending on the size.
Among the 7 fractions thus obtained, wheat flour of the second largest size and wheat flour of the fourth largest size were mixed together (mixing process), and then sufficiently mixed (finishing treatment). The modified wheat flour thus obtained was defined as Invention Product 1.
A modified wheat flour obtained by the same milling process except that 10 U of transglutaminase was added instead of 5 U per 1 g of the wheat protein in the milling process was defined as Inventive Product 2. Similarly, a modified wheat flour obtained by a milling process comprising the transglutaminase treatment by adding 5 U of transglutaminase per 1 g of the wheat protein and the addition of 0.1 g of the partial protein hydrolysate of the wheat protein per 1 g of the wheat protein was defined as Inventive Product 3, and the modified wheat flour obtained by a milling process comprising the transglutaminase treatment by adding 5 U of transglutaminase per 1 g of the wheat protein and the addition of 1.0 g of the partial protein hydrolysate of the wheat protein per 1 g of the wheat protein was defined as Inventive Product 4. A partially hydrolyzed product of the protein portion of wheat with an average molecular weight of about 7,000 was used as the partial protein hydrolyzate (trademark: Glutamine peptide, manufactured by "Canpina Milk Uni- Japan")
Tabelle 1 Weizenmehl Transglutaminase Glutaminpeptid As a control product, general wheat flour was used which was grown without the addition of any transglutaminase and any Partial protein hydrolysate, namely general wheat flour produced by a milling process without any transglutaminase treatment as described above. Table 1 below shows a summary. Table 1 Wheat flour Transglutaminase Glutamine peptide
The four types of modified wheat flour and the control thus obtained were individually weighed to obtain samples of 2,000 g each, which were mixed with common salt (20 g), flavoring for Chinese noodles (Kansui; 20 g) and water (800 g), and then kneaded at 500 mmHg with a vacuum mixer (TVM 03-0028 type vacuum mixer manufactured by Tokyo Noodle Machine, Co.) for 10 minutes. Thereafter, the obtained kneaded product was loosely rolled with a noodle machine manufactured by Shinagawa Noodle Machine, K.K., and thereafter the dough was lumped twice and pressure-rolled four times, and the obtained product was cut, thereby producing four types of Chinese noodles.
Tabelle 2 Chinesische Nudeln The sensory evaluation of the Chinese noodle samples by 10 experts was carried out using a 10-point scale, with the control product being rated at 5 points. The mean scores are shown in Table 2 below. In addition, the cutting energy was measured using a cutting test method with a rheometer, and the results are shown in the same table. In the table, further, the respective individual types of Chinese noodles correspond to the wheat flours as raw materials. (For example, the Chinese noodles of the invention product 2 are made from the modified grain flour of the invention product 2). Table 2 Chinese noodles
Table 2 above shows that the Chinese noodles prepared using any of the modified wheat flour treated with transglutaminase alone and with a combination of transglutaminase and the partial protein hydrolysate as raw materials were not only excellent in terms of sensory evaluation but also better in terms of the objective criterion of cutting energy. The data show that the Chinese noodles obtained using the modified wheat flour exhibited the best noodle flavor.
Attempts were made to prepare four types of modified wheat flours and a control product in the same manner as in Example 1 except that the partial protein hydrolysate of a wheat protein having an average molecular weight of about 10,000 (trademark: MA-Z, manufactured by Morinaga Milk Industry, Co., Ltd.) was used instead of the partial protein hydrolysate of a wheat protein having a molecular weight of about 7,000 (trademark: Glutamine Peptide, manufactured by Campina Milk Uni-Japan).
Beim Abschluß der Fermentation war der Teig bei 28ºC und hatte einen pH-Wert von 5,3.1,400 g of each of the four types of modified wheat flours and the control product thus obtained were weighed, followed by adding yeast (40 g), yeast nutrients (2.5 g) and water (750 g), followed by mixing at low speed for 2 minutes, then at medium speed for 4 minutes and then at high speed for 1 minute with a Hobart mixer. Thereafter, processed fat (50 g) was added to the obtained dough, followed by further mixing at medium speed for 3 minutes and high speed for 1 minute. Then, the dough was kept at 27ºC and 75% RH (Relative Humidity) for 4 hours as a first fermentation to obtain a starting dough. At the completion of fermentation, the dough was at 28ºC and had a pH of 5.3.
Other ingredients (40 g of table salt, 60 g of sugar, 60 g of glucose, 60 g of shortening, 40 g of dry milk powder and 440 g of water) were added to the initial dough and then mixed with a mixer to prepare bread dough. The bread dough was kept at the above temperature for 10 minutes (second fermentation) and then equally divided into 6 portions and kept at 28ºC for another 10 minutes. ten (third fermentation). Then, the obtained dough was put into a baking pan. The dough was kept at 37ºC and 75% RH for 50 minutes for further fermentation (fourth fermentation). Then, the fermented bread dough was put into an oven and baked at 220ºC for 40 minutes to form 5 types of bread loaves.
Tabelle 3 Tabelle 4 The obtained 5 types of bread loaves were organoleptically examined according to the following method. Specifically, the bread loaves were cut into 1.5 cm thick pieces 4 days after baking, and then they were examined by 10 specialists. In addition, the evaluation was carried out using a 10-point scale based on the evaluation in Table 3 below, with each property of the control product being evaluated with 5 points. The mean score of each result of the 5 evaluation criteria, ie, surface color, surface quality, acid expansion, inner color phase and texture, are shown in Table 4 below. Table 3 Table 4
Table 4 shows that all loaves of bread (product of the invention) produced using the modified wheat flour containing transglutaminase alone or a combination of transglutaminase and the partial protein hydrolysate were excellent in terms of sensory evaluation compared to the control product.
A modified wheat flour was produced using American wheat (Standard White Species; 10 kg) as starting material.
First, the microfine sand and the like were removed from the raw material wheat (selection method). Then, the wheat thus selected was put into a container, after which water was added until the water content in the wheat was about 14.3%, and then the obtained wheat was kept at 25°C for 36 hours (soaking method). Then, transglutaminase was added to water to be added in an amount of 1 U per 1 g of the wheat protein and dissolved therein.
After dressing, the wheat husk was separated to obtain a raw endosperm grain (breaking process). Then, the contaminating husk material was removed from the raw endosperm grain by a combination of sieving and selection (cleaning process). In addition, the cleaned raw grain was ground with a smooth roller (pulverizing process). Then, the obtained flour was sieved to form 7 fractions depending on the size of the flour.
Among the 7 fractions thus obtained, the wheat flour of the second largest size and the wheat flour of the fourth largest size were mixed together (mixing process), then sufficiently mixed (final treatment). The modified wheat flour thus obtained was defined as Invention Product 1.
Tabelle 5 zeigt die Produkte Tabelle 5 By the same milling procedure, the product 2 of the present invention was obtained except that 1 U of transglutaminase per 1 g of the wheat protein and 0.1 g of sodium caseinate (manufactured by Nissei Kyoeki, Kabushiki Kaisha) per 1 g of the protein in the wheat were used. Table 5 shows the products Table 5
By using the two types of modified wheat flour and the control wheat flour (wheat flour processed to the same manner except that the transglutaminase treatment was not carried out), sponge cakes were obtained by routine procedures having the compositions shown in Table 6.

Tabelle 7 The egg was placed in a bowl, then water (45 ml) was added and thoroughly mixed with a Hobart mixer, and sugar was added to the resulting mixture. Then, the mixture was adjusted to 30ºC. After that, the mixture was foamed at a high speed for 8 to 10 minutes. After adding the remaining water (40 ml) to the mixture, the mixture was foamed at a high speed for 2 minutes and then at a low speed for 1 minute, thereby preparing the cake batter. The modified wheat flours or the control wheat flour were added to the cake batter, and then sufficiently mixed. The resulting batter was put into a baking pan, after which the surface was smoothed, and the resulting batter was then baked in an oven at 180ºC for 30 minutes. After baking, the product was taken out of the baking pan and cooled to room temperature, and then it was sensorially evaluated by 10 experts. The evaluation results are shown in Table 7 below. In addition, the appearance and internal phase were observed, and the volume of the cake was measured. The results are also shown in this table. The overall assessment based on all these results is also shown in this table. The symbols X, Δ, 0 and indicate not good, normal, relatively good and very good results respectively. Table 7
The above table shows that sponge cakes (the inventive products) were produced with larger volume without cracks in the middle and preferably with soft and smooth texture as compared to the control product.
By the same procedure as in Example 1, 4 types of modified wheat flours and a control product were prepared.
The extensograms of the 4 types of modified wheat flours and the control wheat flour were measured by the following method. The secondary processing properties of wheat flour were measured herein by such an extensogram.
Genauer gesagt wurde bei der Teigherstellung die Menge des reinen Wassers so eingestellt, daß die Spitze der Rundung des Teiges nach 1-minütigem Kneten, 5-minütigem Halten und einem weiteren 2-minütigen Kneten 500 B. U. war.300 g of each of the 4 types of modified wheat flour and the control wheat flour were placed in a Farinogram mixer (trademark: "Farinogram", manufactured by Brabender, Co.), and then 6 g of common salt and an appropriate amount of pure water were added to form a dough. More specifically, in the dough preparation, the amount of pure water was adjusted so that the peak of the roundness of the dough after kneading for 1 minute, holding for 5 minutes and kneading for another 2 minutes was 500 BU.
From the dough thus prepared, 150 g was drawn out and molded in a molding machine, which was then kept in a thermostat at 30ºC for 45 minutes. After this, a first measurement of the extensogram was taken. 45 minutes after further molding (namely, 90 minutes after the start of the first standing) a second measurement was taken. 45 minutes after further molding (namely, 135 minutes after the start) a third measurement was taken. Here, the measurement was carried out with a machine called "Extensigraph" manufactured by Barbender, Co.
Tabelle 8 The data obtained after 135 minutes are shown in Table 8, which serves as an optimal indicator of the elasticity of wheat. In this table, R represents the tensile strength, which is the peak height of the curve of a measured extensigram (the unit is BU). In addition, E represents the elongation, which is the length of the curve of the measured extensigram (the unit is mm). In addition, R/E represents the shape coefficient, which is a value of the tensile strength divided by the elongation. A larger value of R/E represents a higher elasticity of the wheat flour. Table 8
Table 8 shows that all the products of the invention were wheat flours with higher elasticity than that of the control product.
According to the present invention, modified grain flour having excellent processability can be easily produced, and thus processed foods based on grain flour having high quality can be easily produced. Furthermore, according to the present invention, transglutaminase is added to exert its effect during the process of producing wheat flour from wheat as an agricultural product, thereby producing wheat flour having excellent processability. Therefore, the flour industry can easily produce and sell such high quality wheat flour, and thus the advantages of the present invention for the flour industry are enormous.
The modified wheat flour according to the invention has a lower allergen content and is hardly affected by storage over a long period of time compared to conventional products. In particular, the transglutaminase treatment in combination with a foreign protein and/or a partial protein hydrolysate causes much fewer allergies.
