Description
- Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von 2-Alkoxyisobutylisonitril, Kupferisonitriladdukten und die Markierung mit radioaktiven Isotopen, z.B. 99mTc.
- Mit 99mTc markierte Isonitrilverbindungen haben sich als myokardiale Perfusionsmittel erwiesen. Die Herstellung von Etherisonitrilliganden wurde von Bergstein et al. im europäischen Patent 233368 vom 26. Okt. 1987 beschrieben. Die zweckmäßigste dieser Etherisonitrilverbindungen ist 2-Methoxyisobutylisonitril, dessen Herstellung von Bergstein folgendermaßen beschrieben wird:
- Bei den zwei obengenannten Synthesewegen (A) und (B), die 4 bzw. 5 Schritte umfassen, beträgt die Gesamtausbeute lediglich 5,9 - 8,1%. Van Wyk et al. entwickelten ein Verfahren für die Herstellung von 2- Methoxyisobutylisonitril sowie 2-Ethoxyisobutylisonitril, das unter dem Titel "Synthesis and 99mTc Labelling of MMI(MIBI) and its Ethyl Analogue EMI" [Herstellung und 99mTc-Markierung von MMI(MIBI) und seinem Ethylanalogen EMI], Appl. Radiat. Isot. 1991, 42:687
- veröffentlicht wurde.
- Ein Nachteil des oben beschriebenen Verfahrens ist die mögliche Verunreinigung des Produkts mit Quecksilber. Es ist da eine Frage, ob das Reagenz Quecksilberacetat die Produkte im zweiten Schritt beeinflußt oder ob das nicht der Fall ist. Demgemäß wurde ein neues, einfaches und wirksames Verfahren für die Herstellung von 2- Alkoxyisobutylisonitrilverbindungen erfunden.
- Hauptaufgabe der vorliegenden Erfindung ist es, ein neues Verfahren für die Herstellung und 99mTc- Markierung von 2-Alkoxyisobutylisonitril bereitzustellen. Isobutylen wird mit N-Halogensuccinimid in der Gegenwart einer Alkohollösung zu 2-Alkoxyisobutylhalogenid haloalkoxyliert. Im Anschluß hieran erfolgt eine Reaktion von Kaliumphthalimid mit 2-Alkoxyisobutylhalogenid, und im Anschluß hieran eine Hydrazinolyse mit Hydrazin, wodurch man 2-Alkoxyisobutylamin erhält. Schließlich setzt man 2- Alkoxyisobutylamin mit Chloroform unter basischen Bedingungen in der Anwesenheit von Benzyltriethylammoniumchlorid als Katalysator zu 2-Alkoxyisobutylisonitril um.
- Das neue Verfahren zur Herstellung dieser Isonitrile hat sich daher als sauberer, wirkungsvoller und zweckmäßiger als aus der Literatur bekannte Verfahren erwiesen.
- Die 99mTc-Markierung von 2-Alkoxyisobutylisonitril wird vorgenommen, indem man Kupferisonitriladdukte mit dem radioaktiven Isotop 99mTc mischt. Solch ein Addukt läßt sich leicht und rasch mit 99mTc markieren und ergibt gute Ausbeuten. Tetrakis(2-alkoxyisobutylisonitril)kupfer(I)tetrafluorborate können dadurch hergestellt werden, daß man Acetonitrilmoleküle in Tetrakis(acetonitril)kupfer(I)tetrafluorborat gegen einen 2-Alkoxyisobutylisonitrilliganden bei Raumtemperatur austauscht.
- Die Haloalkoxylierung von Isobutylen gelingt mittels Halogen in Alkohol, wie in Gleichung (1) dargestellt. Haloalkoxylierung
- Von den Olefinen erhält man mit diesem Verfahren zweckmäßige, hohe Ausbeuten an 2. Läßt man Kaliumphthalimid mit 2 reagieren, so erhält man das N-Alkylphthalimid. N-substituierte Phthalimide können durch Hydrolyse oder Hydrazinolyse in die entsprechenden Verbindungen 3 umgewandelt werden. Die Herstellung von 3 läßt sich schematisch als Gleichung (2) zusammenfassen.
- Es ist offensichtlich, daß die in dieser Reaktion gebildete Verbindung 3 nicht durch sekundäre oder tertiäre Amine verunreinigt sein wird.
- Für die Herstellung von Isonitrilen wurde das Phasentransferkatalyseverfahren auf wirksame Weise eingesetzt.
- Wie in Gleichung (3) dargestellt, erhält man 4 dadurch, daß man 3 mit Chloroform in einer NaOH-Lösung mit dem Benzyltriethylammoniumchlorid als Katalysator reagieren läßt. Die vorliegende Erfindung läßt sich durch die folgenden Beispiele genauer beschreiben; sofern nicht anders angegeben, handelt es sich bei den Prozentzahlen um Gewichtsprozent.
- N-Bromsuccinimid (3,56 g, 0,02 Mol) wurde in Methanol gelöst und die Lösung in einem Eis/Acetonbad auf -10ºC gekühlt. Die Lösung wurde langsam mit Isobutylen versetzt, 5 Stunden rühren gelassen und in einen Scheidetrichter gegossen, der mit NaCl gesättigtes Wasser enthielt. Die organische Phase wurde entfernt und die wäßrige Phase dreimal mit je 100 ml Dichlormethan extrahiert. Die vereinigten organischen Phasen wurden über wasserfreiem Magnesiumsulfat getrocknet und filtriert und das Lösungsmittel größtenteils abrotiert. Die erhaltene Lösung wurde bei Atmosphärendruck destilliert und das Produkt bei 140ºC aufgefangen (3,14 g, Ausbeute 94%).

- N-Jodsuccinimid (4,50 g, 0,02 Mol) wurde in Methanol gelöst und in einem Eis/Acetonbad auf -15ºC gekühlt. Die Lösung wurde langsam mit Isobutylen versetzt, 4 Stunden rühren gelassen und in einen Scheidetrichter gegossen, der mit NaCl gesättigtes Wasser enthielt. Die organische Phase wurde entfernt und die wäßrige Phase dreimal mit je 100 ml Dichlormethan extrahiert. Die vereinigten organischen Phasen wurden über wasserfreiem Magnesiumsulfat getrocknet und filtriert und das Lösungsmittel größtenteils abrotiert. Die erhaltene Lösung wurde unter verringertem Druck bei 10 mm Hg-Druck und einer Temperatur von 50ºC eingeengt (4.07 g, Ausbeute 95%).

- Eine Lösung von 2-Methoxyisobutylbromid (3,34 g, 0,02 Mol) in 100 ml Dimethylformamid wurde mit Kaliumphthalimid (3,70 g, 0,02 Mol) versetzt. Die Mischung wurde weitere 4 Stunden bei Rückflußtemperatur gerührt, wobei die Temperatur langsam auf 25ºC absank. Man gab 200 ml Chloroform zu und goß die Mischung auf 500 ml kaltes Wasser. Die wäßrige Phase wurde abgetrennt und mit zweimal je 50 ml Chloroform extrahiert. Die vereinigten Chloroformphasen wurden mit 100 ml 0,2 N Natriumhydroxid und 100 ml Wasser gewaschen. Nach dem Trocknen wurde das Chloroform entfernt. Mit diesem Rückstand wurde Hydrazin (2 g, 0,04 Mol) in 100 ml Methanol versetzt und die Mischung eine Stunde auf Rückflußtemperatur erhitzt. Das Methanol wurde durch Einengen unter verringertem Druck entfernt. Die verbleibende wäßrige Lösung wurde mit konzentrierter Salzsäure versetzt und die Mischung eine Stunde auf Rückflußtemperatur erhitzt. Anschließend wurde die Lösung unter vermindertem Druck eingeengt, um die Salzsäure größtenteils zu entfernen. Der pH des feuchten Rückstands wurde mit Natriumhydroxid auf 14 gebracht und die Mischung in einen Scheidetrichter gegossen, der gesättigte K&sub2;CO&sub3;-Lösung enthielt. Die erhaltene Lösung wurde bei Atmosphärendruck destilliert und das Produkt bei 125ºC aufgefangen (1,6 g, Ausbeute 78%).

- Ein Kolben mit Natriumhydroxidlösung (3,2 g NaOH und 5 ml H&sub2;O) wurde tropfenweise mit einer Mischung aus 2-Methoxyisobutylamin (2,06 g, 0,02 Mol), Chloroform (4,80 g, 0,04 Mol) und Benzyltriethylammoniumchlorid (40 mg, 0,17 mmol) in 50 ml Dichlormethan versetzt. Der Ansatz wurde zwei Stunden auf Rückflußtemperatur erhitzt. Anschließend verdünnte man mit 100 ml Eiswasser, die organische Phase wurde abgetrennt und aufbewahrt und die wäßrige Phase mit 50 ml Dichlormethan extrahiert. Die Dichlormethanlösungen wurden vereinigt und über wasserfreiem Magnesiumsulfat getrocknet. Die erhaltene Lösung wurde unter vermindertem Druck bei 25 mm Hg-Druck eingeengt und das Produkt bei 55º-60ºC aufgefangen (1,42 g, Ausbeute 63%).

- N-Bromsuccinimid (3,56 g, 0,02 Mol) wurde in Ethanol gelöst. Das Reaktionsprodukt wurde nach Beispiel 1 entsprechenden Verfahren erhalten (Ausbeute 95%).

- Tetrakis(acetonitril)kupfer(I)tetrafluorborat (0,50 g, 1,6 x 10&supmin;³ Mol) wurde in 100 ml Ethanol suspendiert. Zu dieser Suspension wurde 2-Methoxyisobutylisonitril (0,72 g, 6,4 x 10&supmin;³ Mol) langsam zugegeben und die Mischung bei Raumtemperatur eine Stunde gerührt. Das Lösungsmittel wurde anschließend unter vermindertem Druck abgedampft. Das Produkt wurde aus Ethanol/Ether umkristallisiert (0,91 g, Ausbeute 95%), Smp. 100 -101ºC. Analyse: Berechnet für C&sub2;&sub4;H&sub4;&sub4;N&sub4;O&sub4;CuBF&sub4;: C 47,80; H 7,30; N 9,29; Cu 10,54; B 1,79; F 12,62. Gefunden: C 47,69; H 7,40; N 9,05; Cu 10,52; B 1,80; F 12,71.

- Eine gerührte Suspension von Tetrakis(acetonitril)kupfer(I)tetrafluorborat (0,50 g, 1,6 x 10&supmin;³ Mol) in 100 ml Ethanol wurde bei Raumtemperatur langsam mit 2- Ethoxyisobutylisonitril (0,81 g, 6,4 x 10&supmin;³ Mol) versetzt. Die Reaktionsmischung wurde 30 Minuten gerührt, um eine klare Lösung zu erhalten, und das Lösungsmittel wurde anschließend unter vermindertem Druck zur Trockene verdampft. Das Reaktionsprodukt wurde aus Ethanol/n-Hexan umkristallisiert und die erhaltenen weißen Feststoffe mit n- Hexan gewaschen und im Vakuum getrocknet; (1,01 g, Ausbeute 96%), Smp. 76º-77ºC. Analyse: Berechnet für C&sub2;&sub8;H&sub5;&sub2;N&sub4;O&sub4;CuBF&sub4;:
- C 51,04; H 7,90; N 8,51; Cu 9,65; B 1,64; F 11,54. Gefunden:
- C 51,10; H 7,85; H 8,59; Cu 9,61; B 1,70; F 11,61.

- In einem 8 ml-Probenröhrchen werden Tetrakis(2-methoxyisobutylisonitril)kupfer(I)tetrafluorborat (1-2 mg) oder Tetrakis(2-ethoxyisobutylisonitril)kupfer(I)tetrafluorborat (1-2 mg), Natriumcitratdihydrat (2,2-3,2 mg), Mannit (16-26 mg), Cysteinhydrochlorid (1-3 mg) und Zinndichlorid (0,05-0,09 mg) vermischt. Die Röhrchen wurden verschlossen, und 25-40 mCi (1-2 ml) 99mTcO&sub4;&supmin;, das durch Elution eines &sup9;&sup9;Mo/99mTc-Radionuklidgenerators erhalten worden war, zugegeben. Die Röhrchen wurden bei 95º - 100ºC 10-15 Minuten im Wasserbad erhitzt und anschließend auf Raumtemperatur abkühlen gelassen. Eine Qualitätskontrolle der in-vitro-Stabilität wurde mittels ITLC(Silicagel) unter Verwendung von Kochsalzlösung und Methylethylketon (MEK) durchgeführt, und so 99mTcO&sub2; (Rf:0), 99mTcO&sub4;&supmin; (Rf:0,9 - 1,0) und der 99mTc-Isonitrilkomplex bestimmt (MEK:Rf:MMI 0,45, EMI 0,8; Kochsalzlösung:0).
Claims (5)
1. Synthesemethode von 2-Alkoxyisobutylisonitril
mit Isobutylen als Ausgangsmaterial, dadurch
gekennzeichnet, dass durch Haloalkoxylation des genannten
Isobutylens 2-Alkoxyisobutylhalid erhalten wird,
welches dann zu 2-Alkoxyisobutylamin versetzt wird,
wobei das genannte 2-Alkoxyisobutylamin, unter
basischen Bedingungen, mit Chloroform reagiert und 2-
Alkoxyisobutylisonitril ergibt, wobei das genannte 2-
Alkoxyisobutylisonitril-Produkt die allgemeine Formel
aufweist, wobei R eine Alkylgruppe mit 1 bis 4
Kohlenstoffatomen darstellt.
2. Synthesemethode gemäss Anspruch 1, in der
das genannte 2-Alkoxyisobutylhalid durch Reaktion von
Isobutylen mit N-Halosuccinimid, bei Temperaturen
zwischen -50ºC bis 0ºC, in einem entsprechenden
alkoholischen Lösungsmittel, synthetisiert wird.
3. Synthesemethode gemäss Anspruch 1, bei der
das genannte 2-Alkoxyisobutylamin durch Reaktion von
2-Alkoxyisobutylhalid mit Kaliumphthalimid und Hydrazin
bei einer Temperatur zwischen 100ºC bis 200ºC
synthetisiert wird.
4. Synthesemethode gemäss Anspruch 1, in der das
genannte 2-Alkoxyisobutylisonitril durch Reaktion von
2-Alkoxyisobutylamin mit Chloroform, Natriumhydroxid
und dem Katalisator Benzyltriethylammoniumchlorid bei
einer Temperatur von 50ºC bis 100ºC synthetisiert wird.
5. Synthesemethode gemäss Anspruch 1, in der die
genannte Struktur 1 mit einem Kupfersalz einen Komplex
ergibt, welches mit Technetium-99m in Anwesenheit von
Sn(II) markiert wird und die Markierungsausbeute
grösser ist als 90%.
