Description
The present invention relates to a bound enzyme bioreactor, in which an enzyme is used as a biocatalyst for industrially carrying out a biochemical reaction.
Recently, research on bioreactors containing in a reaction column a "bound enzyme" catalyst carrying an enzyme fixed to the carrier surface has made rapid progress. For this purpose, various carriers have been proposed, such as high molecular weight organic substances such as carrageenan or polyacrylamide, etc., or ordinary ceramic substances such as alumina, zirconia, etc. However, in reaction systems using a bioreactor having a "bound enzyme" catalyst, operations at high temperatures are often required to prevent contamination of the bioreactor caused by undesirable microorganisms, and bioreactors using organic substances as the carrier of the "bound enzyme" catalyst have disadvantages in that they lack chemical stability and heat resistance property to withstand operation at high temperatures. Bioreactors using ordinary ceramic substances as the carrier of the "bound enzyme" catalyst, on the other hand, have disadvantages in that the catalyst can only bind a small amount of enzyme, so that the productivity of the bioreactor using such a catalyst becomes poor, and the entire plant must be designed large or the reaction time must be extended.
To overcome these disadvantages, bioreactors using porous glass beads as a carrier of the enzyme-fixed catalyst have been of interest (Sumoi Sakka "Application of Ceramics to Enzyme-fixed Carriers" in "Kagakusohchi" [3], 1983, pp. 52-58). The porous glass beads have many advantages such as small pores with a diameter of a few hundred Å by using phase separation of glass, resulting in a large specific surface area of 60-75m²/g, stable thermal and chemical properties, high mechanical strength and an outstandingly large amount of enzyme that can be fixed to a unit amount of carrier compared to conventional ceramic carriers. However, the porous glass beads have disadvantages in that they have to be produced in a complicated process in which a glass is melted at a high temperature of 1 500ºC, the molten glass is reheated to 500-600ºC and Furthermore, the molten glass is heat treated for a long period of time, so they are very expensive as a support and difficult to use commercially in industrial-scale bioreactors. Thus, no bioreactor has yet been created that has both chemical and thermal stability as well as productivity and economic efficiency.
It is an object of the present invention to overcome the above disadvantages.
According to the present invention, it is possible to provide a "bound enzyme" reactor containing in a reaction column a thermally and chemically stable "bound enzyme" catalyst composed of a carrier and an enzyme supported on the carrier in an outstandingly large amount per unit amount of the carrier as compared with that of the porous glass beads and which can be manufactured inexpensively.
The present invention is an enzyme-bound bioreactor comprising a reaction column and catalyst particles evenly and densely packed or filled in the reaction column, wherein the catalyst particles are composed of supported sepiolite particles consisting of sepiolite and an enzyme fixed to the surface of the supported sepiolite particles.
The carrier sepiolite used is fired sepiolite, which is preferably obtained by pulverizing a raw sepiolite material consisting of sepiolite or raw sepiolite stone to form sepiolite powder, optionally adjusting the chemical composition and/or particle size distribution of the sepiolite powder, molding the sepiolite powder into a desired shape such as particles, and firing the molded powder at a temperature of about 300 to 1100°C. A raw sepiolite stone is a lump of magnesium silicate fibers which has a unique crystal structure of talc fractions stacked in a brick wall shape and has peaks of about 10 Å and 200 Å in the pore size distribution. The inventors have found that when a raw sepiolite stone is fired, crystal water is lost to form a ceramic body, while its crystal structure and pore size distribution are changed to have a large peak of pores in the range of 200 to 400 Å. Carrier sepiolite thus obtained has a specific surface area of 230 m²/g, which is larger than that of porous glass beads, so that it has excellent functionality for fixing enzymes to it, as well as thermal and chemical stability due to its ceramization.
The supporting sepiolite can be silanized on the surface using a known method with a silane coupling agent and then treated to fix enzyme to the silanized surface to obtain "bound enzyme" catalyst, which is then packed into the reaction column of the bioreactor.
Conventional means can be used to fix enzyme to the carrier sepiolite, and enzyme is firmly fixed to the surface of the carrier sepiolite consisting of magnesium silicate by covalent bonding at a high density. Enzymes for fixing to the carrier sepiolite are not limited in any way, and illustrative examples thereof are invertase, lactase, pepsin, trypsin, protease, glucoamylase, glucose isomerase, aminoacylase, etc.
For a better understanding of the present invention, reference is made to the accompanying drawings in which:

Referring to Figure 1, an embodiment of the bioreactor according to the invention is shown, wherein the "bound enzyme" catalyst 1 is introduced into the reaction column. The "bound enzyme" catalyst 1 using the supported sepiolite has a disadvantage in that it is fragile when rubbed against itself as compared with the conventional catalyst using the porous glass beads or alumina ceramics. Therefore, the "bound enzyme" catalyst 1 according to the present invention should be uniformly and densely packed in the reaction column 2 to avoid movement relative to itself. For this purpose, the "bound enzyme" bioreactor according to the present invention preferably has a movable plate 3 at the bottom and/or the top of the reaction column 2, and the plate 3 is movable by a screw 4, etc. to pack the "bound enzyme" catalyst 1 uniformly and densely in the reaction column 2 as shown in Figure 1. In the embodiment shown in Fig. 1, a crude feed solution is introduced upward into the reaction column 2 through the inside of the screw 4, and a waste solution containing reaction products is discharged from a discharge port 5 at the top of the reaction column 5. The uniformly and densely packed "bound enzyme" catalyst particles do not move in the reaction column 2 even if they are rinsed to prevent the decrease of enzyme activity, to reabsorb enzyme with reduced enzyme activity and to re-fix new enzymes on the catalyst, etc., so that they are not broken or destroyed and do not cause troubles such as clogging or shortcuts in the reaction column 2.
In the following, the present invention will be explained in more detail with reference to an example.
Burnt sepiolite particles having an average particle diameter of 0.3 mm prepared by pulverizing, molding and firing as above are densely packed into a reaction column having an inner diameter of 20 cm and a height of 8 cm and pressed in the reaction column by a movable plate arranged on the reaction column. The volume of the layer of packed catalyst particles at this point is about 11. For comparison, another reaction column is prepared which is filled with carrier cordierite particles having the same average particle size is filled.
Then, 70% aqueous ethanol solution is supplied to the two reaction columns at a flow rate of 30 ml/min and the bioreactors are drained. Then, distilled water is supplied to the reaction columns at a flow rate of 100 ml/min to wash out ethanol, and then 50 mA of aqueous acetic acid buffer solution (pH 4.0) is supplied at a flow rate of 100 ml/min to exchange the solution in the reaction columns. Then, 2% commercial invertase/50 mA aqueous acetic acid buffer solution is circulated in the reaction columns at a flow rate of 30 ml/min for 3 hours to fix invertase to the support surface. Excess invertase is recovered and then 30% sucrose aqueous solution/50 mA acetic acid aqueous buffer solution (pH 4.0) is supplied to the reaction columns as invertase substrate at such flow rates that the average residence times of the solution in the column are 5 min, 10 min, and 15 min, respectively, to measure the glucose concentration of the solution at the outlet of the bioreactor, while the reaction columns are kept warm by passing 37ºC water through jackets of the reaction columns.
The result is shown in Figure 2. As can be seen from Figure 2, the bioreactor of the present invention has a sucrose decomposition rate that is about 30 times faster than that of the conventional bioreactor using the carrier cordierite particles. Thus, the bioreactor of the present invention has a productivity that is 30 times higher than that of the conventional bioreactor having an equal volume. When the productivity of the bioreactor of the present invention is kept constant, i.e., the same as that of the conventional bioreactor, the average residence time of the substrate in the reaction column or the volume of the bioreactor of the present invention can be reduced to 1/30 of that of the conventional bioreactor.
As is clear from the foregoing explanations, the bioreactor according to the present invention using "bonded enzyme" catalyst particles consisting of supported sepiolite particles and enzyme fixed to the surface of the supported sepiolite particles uniformly and densely packed or filled in the reaction column has far superior productivity of the intended product compared with a conventional bioreactor using a ceramic catalyst carrier. and excellent heat resistance and chemical resistance. Even when the bioreactor of the present invention is compared with the conventional bioreactor using porous glass beads, the bioreactor of the present invention has excellent economy because the carrier sepiolite particles can be manufactured very cheaply and economically, so that even a large bioreactor can be manufactured at a low cost. Moreover, when the "bound enzyme" catalyst particles are uniformly and densely packed in the reaction column with a movable plate, etc., breakage of the "bound enzyme" catalyst particles, shortcuts and clogging in the reaction column are prevented. Therefore, the "bound enzyme" bioreactor of the present invention overcomes all the problems existing in the prior art and is highly industrially useful.
Although the present invention has been described with reference to specific values and embodiments, it will be understood by those skilled in the art that various variations and modifications are possible.
