Description
Verfahren zur Züchtung von im Menschen und Tierkörper parasitisch vegetierenden Kleinlebewesen Durch die in der Patentschrift 17o 68o niedergelegten grundlegenden Arbeiten von Dr. O t t o S c h m i d t ist ein Verfahren zur Züchtung von im Menschen- und Tierkörper parasitisch lebenden Kleinlebewesen außerhalb des tierischen Körpers bekannt geworden, welches darauf beruht, daß niedere Pflanzen als Nährböden (Zwischenwirte) verwendet werden. Die Durchführung des Züchtungsvorgangs soll nach dieser Patentschrift derart erfolgen, daß eine Reinkultur der niederen Pflanze, z. B. von Mucor, auf einem sterilen Nährboden hergestellt und Sporen dieser Kultur auf in sterilen Kulturgläsern befindliche Tumorstückchen übertragen werden. Hierdurch gelangt man zu Stammkulturen, welche den Mucor und, in ihm vegetierend, den Erreger enthalten. Durch überimpfung dieser Stammkultur auf geeignete Nährsubstrate, kann man dann größere Mengen von aus Mucor und Parasit bestehenden Kulturen züchten: Die nach längerem, z. Zunächst wurde gefunden, daß es von Wichtigkeit ist, die Züchtung des pathogenen Krankheitserregers in niederen Pflanzen bei Temperaturen, welche unterhalb der Bluttemperatur (37° C) liegen, durchzuführen. Temperaturen unterhalb a8°, vorzugsweise solche zwischen io bis i8°, als vorteilhaft erwiesen. Die als Ausgangsmaterial für die Züchtung der pathogenen Erreger in Betracht kommenden Stoffe, wie z. B. Gewebestücke, Ödemflüssigkeit, Blut, Exsudat, enthalten vielfach neben dem zu züchtenden Parasiten noch andere-Mikroorganismen. Durch die Möglichkeit der Weiterzüchtung des erstrebten Parasiten in Symbiose mit niederen Pflanzen unterhalb Bluttemperatur besteht nun die Möglichkeit, andersartige aus den Ausgangsstoffen herrührende Mikroorganismen, welche nur bei Bluttemperatur gut zur Entwicklung kommen, auszuschalten. Dabei werden auch solche Mikroorganismen, welche unterhalb Bluttemperatur an sich zu gedeihen vermögen, in ihrer Entwicklung dadurch geschädigt, daß die bei der angewendeten Temperatur optimal wachsenden niederen Pflanzen den zu schädigenden Mikroorganismen das Nährmedium entziehen. Da die zu züchtenden pathogenen Erreger, insbesondere solche, welche zur Bildung von Geschwülsten (Karzinom, Sarkom usw.) Veranlassung geben, sich sonst nur bei Körperwärme entwickeln, war nicht vorauszusehen, daß sie bei der Symbiose mit niederen Pflanzen, wie Mucor u. dgl., auch bei erheblich,niedrigeren Temperaturen einen vollständigen Entwicklungszyklus durchzumachen vermögen. Weiterhin wurde gefunden, daß der Zuckergehalt der Nährsubstrate, als welche Peptonzuckerbouillon, Erbsenzuckerbouillonagar u. dgl. in Betracht kommen, für die vorliegenden Züchtungszwecke von beträchtlicher Bedeutung ist. Es hat sich z. B. gezeigt, daß der Zuckergehalt von Nährbouillon, Nährgelatine u. dgl. im allgemeinen 6 °/o nicht übersteigen soll. Im übrigen richtet sich die Höhe des Zuckergehaltes nach den sonstigen Bestandteilen des Nährsubstrates, z. B. dem Peptongehalt, und den jeweils angewendeten Arbeitsbedingungen. Außerdem hat sich ergeben, daß die Verwendung reinen Zuckers nicht vorteilhaft ist. Dagegen hat sich ungereinigter Zucker, z. B. die unter der Bezeichnung Traubenzucker im Handel befindliche ungereinigte Glukose, als sehr geeignet für die Herstellung der Nährsubstrate erwiesen. Ein gutes Wachstum wird z. B. bei Anwendung von Zuckermengen zwischen 0,5 und 8 °i" erzielt; das Wachstumoptimum liegt im allgemeinen zwischen o,8 bis 2 °/a Zuckergehalt. Weitere Untersuchungen haben ergeben, daß die in Betracht kommenden Erreger vielfach schon im lebenden Körper mit niederen Pflanzen oder pflanzlichen Kleinlebewesen in Gemeinschaft leben können. Auf Grund dieser Erkenntnis hat es sich als möglich erwiesen, aus Körperbestandteilen erkrankter Tiere oder Menschen die gesuchten Parasiten zusammen mit den niederen Pflanzen auf verhältnismäßig einfachem Wege zu gewinnen und zu züchten und hierdurch das eingangs erwähnte Züchtungsverfahren erheblich zu vereinfachen. Es bleibt dann auch außerhalb des lebenden Körpers die Symbiose bestehen. Parasit und niedere Pflanze vermögen sich ,auf geeigneten Nährböden, und zwar auch unterhalb Körpertemperatur zusammen weiterzuentwickeln, derart, daß die als Zwischenwirt fungierende niedere Pflanze sich von dem künstlichen Nährboden nährt, während die in dem Zwischenwirt lebenden, .ihre Virulenz behaltenden Erreger ihre Nahrung aus der Wirtspflanze schöpfen. In Ausübung des Verfahrens wird z. B. so vorgegangen, daß in einem keimfreien Raum steril herausgenommene Geschwulststückchen oder steril entnommenes Blut oder Ödemflüssigkeit von erkrankten Tieren oder Menschen in :geeignete flüssige oder feste Nährsubstrate, z. B. Pepton-, »Liebig«-, Zuckerbouillon oder Erbsenzuckerbouillonagar, überimpft werden. Aufschwemmungen von Tumorgewebe, Blut, Exsudat usw., von an Tumoren erkrankten Menschen oder Tieren und außerdem noch Kulturender niederen Pflanze, z. B. Wenn die Entwicklung ihren Höhepunkt erreicht hat, z. B. nach q. bis 6 Wochen, wird die einen üppigen Rasen bildende Nährpflanze mit den darin lebenden Erregern herausgenommen und zerkleinert. Weiterhin wird der Bodensatz, welcher ebenfalls Teile der niederen Pflanze und Erreger enthält, z. B. durch Abhebern oder Abgießen der Hauptflüssigkeitsmenge und Zentrifugieren der restlichen Menge gewonnen. Hierauf werden die gesamten .festen Bestandteile getrennt oder zusammen einer Behandlung unterworfen, welche eine möglichst weitgehende Trennung des Erregers von der niederen Pflanze gestattet. Man verfährt z. B. derart, daß man das Produkt in einer geeigneten Flüssigkeit, z. B. Wasser, oder ihrer eigenen Kulturflüssigkeit, welche geringe Mengen von quellend wirkenden Substanzen, z. B. geringe von Salzen oder alkalischen Stoffen, z. B. o,5 g auf roo ccm. Flüssigkeit enthalten, erwärmt, z. B. auf Temperaturen von 3o bis q.2° C, und das Gemisch dann einer mechanischen Behandlung, z. B. in einem Schüttelapparat, unterwirft. Zweckmäßig wird Erwärmung und Schütteln mehrmals abwechselnd vorgenommen. Durch diese Behandlung werden die Bestandteile der niederen Pflanze zum Schwellen und Platzen gebracht, so daß die Erreger austreten und in die Flüssigkeit gelangen können, woselbst sie zum Teil- zur Auflösung gebracht, zum Teil. erhalten bleiben. Nach dieser Behandlung wird die Flüssigkeit z. B. durch Zentrifugieren, Filtrieren und ähnliche Behandlungsmethoden von den festen Bestandteilen getrennt und als Vaccine verwendet. Gegebenenfalls kann man die Flüssigkeit auf gewünschte Stärke einstellen, z. B. durch Zusatz passender Mengen der aus den Kulturkolben gewonnenen, von festen Bestandteilen befreiten, ebenfalls noch Erreger und Abbauprodukte derselben enthaltenden Flüssigkeit.
