Description
The invention relates to a method for determining the antioxidative potential in living skin as well as a transdermal preparation for use in such Method according to the claims.
Every cell has an antioxidant potential that through the balance between the factors that affect auto-oxidation exercise and those who produce the antioxidant effect is determined. The antio is present in the skin xidantien a process opposite that under the collective term "oxidative stress" the physiological influences of skin specific metabolic reactions as well as exogenous factors such as ionizing and non-ionizing radiation, photosensitive lization, toxic and allergic contact dermatitis etc. includes, and a shift in balance in favor of the oxidants and thus an occurrence of molecular damage causes.
The antioxidants have the job of being biological reactive oxidants such as B. the superoxide anion radical, Hydrogen peroxide, hydroxyl radicals, singlet oxygen, Transition metals, radical chelates, hydroperoxides, lipid radicals and break down thiyl radicals to oxidative cell and tissue to avoid damage. Targets of the oxidants z. B. in the Skin are lipids, proteins, carbohydrates and nucleic acids.
Because of the low concentration of free radicals in the tissue, the extremely high reactivity, the low biological half-life from a few nano to milliseconds and the complex structure of the skin in several biochemical and morphologically different sections, is the proof free radicals and reactive oxygen species in the skin methodically very difficult and only possible for individual species (Spin trap technique). Another aggravating factor is that Variety of chemically overlapping compounds in the bio logical material. The qualitative and quantitative proof of free radicals and reactive oxygen species in biolo material requires sophisticated analytical techniques. Direct detection of reactive oxidants or their oxi dation products in biological material can be isolated Cases with complicated technical methods are provided. However, direct detection is very often in vitro and in vivo not possible. Indirect methods of determination, such as the measurement to increase the concentration of peroxidation products and Decreases in the concentration of antioxidants are common procedures performed.
Der Thiobarbitursäure-Test, disku tiert von Gutteridge, 1986, JMC, Aspects to condsider when detecting and measuring lipid peroxidation. An indirect method of detecting reactive oxidants tien in vitro is the quantitative determination of specific Reaction products, e.g. B. peroxidation products of the lipids, Nucleic acids and proteins. The thiobarbituric acid test, discu by Gutteridge, 1986, JMC, Aspects to condsider when detecting and measuring lipid peroxidation. Free radical Research Communication, 1: 173-184, in the literature as a Technically and sensitively described method, in addition to lipid peroxidation products also after keto acids and nucleic acids and is not suitable for measuring the lipid peroxidation kines tik. The determination of lipid peroxidation products of the cell membranes with optical spectroscopy of transparent solutions gen, on the other hand, only shows damage to the cell membranes themselves that a qualitative and quantitative analysis of lipid oxida tion in biological material appears difficult.
Molecular oxygen is the substrate for education of the superoxide anion radical, hydrogen peroxide and Hydroxyl radical. The quantitative determination of oxygen with the method oxypolarography indirectly enables the Er capture reactive oxygen species when cellular breathing (4 electron reduction of oxygen) is low and the Oxygen consumption through one-electron transfer (radical reaction) dominates. This usually allows in vitro measurements on cell organelles or isolated cells Inhibition of physiological breathing through cyanide.
Antioxidants like glutathione, ascorbate and tocopherol are caused by reactive oxygen species and free radicals oxidized. Determining the concentration of antioxidants or of their oxidation products with sensitive measuring methods (e.g. HPLC with electrochemical, spectrophotometric or fluorimetric detection) enables isolated detection solution of reactive oxidants in different tissue compartments Disadvantages of the method are that conclusions about the chemical nature of the reactive oxidant not clearly possible the sample collection is invasive and preparation artifacts may occur.
The method for measuring bioluminescence is indeed a highly sensitive measuring method and by selecting special ones The method achieves inhibitors and wavelength ranges high selectivity, but has the disadvantage of direct detection solution of light quanta as a parameter for the radical balance as well as the inapplicability for radika investigations chemical reactions without chemiluminescence and limited applicability for in vivo measurements.
From EP-A-351919 it is known that deuterated Nitroxide radicals in the ESRENRI (Electron Spin Resonance Enhanced Magnetic Resonance Imaging) as a contrast Component (contrast medium) can be used. there however, the influence on the relaxation time is measured of hydrogen nuclei for the purpose of improving the Image contrast in NMR tomography with as long as possible stable nitroxides, which, even after the measurement as such in the Body remain.
In a publication in Magnetic Resonance Micros copy, Verlag Chemie Weinheim 1992, 563, the penetration of spin markers and the spectral change in the lines of Proxylmaleimide examined using a special EPR technique.
Overall, the problem in this area is that the presence of individual antioxidants or of their Oxidation products can be detected in the skin in vitro could or the penetration of radicals into the skin, but not all that is available in the skin antioxidative potential. However, this would be a crucial one Size for comparability of certain anti-agents oxidative stress on the skin.
The invention has for its object a method to determine the antioxidative potential on the skin and to provide a transdermal preparation therefor.
Erfindungsgemäß besteht das Verfahren zur Messung des
antioxidativen Potentials darin, daß man
According to the invention, the method for measuring the antioxidative potential is that
- a) eine transdermale Zubereitung auf die Haut aufträgt, bestehend aus einem physiologisch annehmbaren, in die Haut eindringenden Trägerstoff und einer physiologisch annehmbaren, paramagnetischen Modellsubstanz in Form eines freie Radikale tragenden Nitroxids oder Nitroxidgemisches, die in der Haut durch Antioxidantien innerhalb von 5 bis 60 Minuten abgebaut werden, wobei die Konzentration der Modellsubstanz im Bereich von 1 bis 100 mMol liegt;a) applies a transdermal preparation to the skin, consisting of a physiologically acceptable, in the skin penetrating carrier and a physiologically acceptable paramagnetic model substance in the form of a free radical carrying nitroxide or mixture of nitroxides in the skin degraded by antioxidants within 5 to 60 minutes the concentration of the model substance in the range from 1 to 100 mmol;
- b) eine gemäß (a) behandelte Haut einem oxidativen Stress unterzieht;b) a skin treated according to (a) an oxidative stress subjects;
- c) die magnetischen Momente der freien Elektronen der Nitroxidradikale mittels der paramagnetischen Elektronenreso nanz (EPR) nicht-invasiv unter den Bedingungen Mikrowellenleistung 1 bis 20 mM, Frequenzbereich 1 bis 10 GHz, Einsatz einer Oberflächenspule, Linienbreite der Modellsubstanz 0,1 bis 5 Gauss (0,1 bis 5 T) misst; und (d) die unter (c) erhaltenen Meßwerte der Intensität des low- field-Peaks oder des mid-field-Peaks des EPR-Spektrums als zeitabhängige Intensität erfaßt und im Vergleich mit nur gemäß (a) behandelter und danach gemäß (c) gemessener Haut als rela tive Intensität ausweist.c) the magnetic moments of the free electrons of the Nitroxide radicals using the paramagnetic electron resonance nance (EPR) non-invasive under the conditions Microwave power 1 to 20 mM, frequency range 1 to 10 GHz, Use of a surface coil, line width of the model substance Measures 0.1 to 5 Gauss (0.1 to 5 T); and (d) the measured values of the intensity of the low- field peaks or the mid-field peak of the EPR spectrum as time-dependent intensity recorded and compared with only according (a) treated and then measured according to (c) skin as rela tive intensity.
With the EPR the detection of the unpaired electron of the radical of the model substance (nitroxide).
Starting from a relatively quick degradability in the Periods of 5 to 60 minutes are suitable as model radicals 5- and 6-ring systems, which include the following nitroxides Proxo (2,2,5,5-tetramethyl-1-dihydropyrrolinoxy-nitroxide), Proxad (2,2,5,5-tetramethyl-1-dihydropyrroloxy-nitroxide), Doxo (2,2,5,5-tetramethyl-3-oxazolidinoxy-nitroxide), Tempo (2,2,6,6-tetramethyl-1-piperidinoxy-nitroxide), Tempol (2,2,6,6-tetramethyl-1-piperidinoxy-4-ol-nitroxide), CAT 1 (2,2,6,6-tetramethyl-1-piperidinoxy-4-trimethylammo bromide-nitroxide). These connections as well as the connection DTBN (di-tert-butyl nitroxide) with a particularly small molecule have low stability, i. H. they are easily degradable in the range of the intended measuring time, they have a good dif fusion behavior when penetrating into the object to be examined jekt, and they are physiologically compatible.
The breakdown time of a model radical is in the range of 10 to 60 minutes, preferably 10 to 30 minutes. Under the The term "dismantling time" used here means the time in which the radical is neutralized (reduction).
The solubility of the model radicals depends on the Area of application (target). For dermatological use are transport systems based on liposomes or micro emulsions possible. Also the use of with fluorocarbons asymmetric lamellar aggregates produced (DE-A-42 21 255) as a carrier system is possible.
stellt:
When the method is carried out in practice, a model radical is added to the system to be examined in a concentration of 1 to 100 mmol, for example 10 mmol, and the EPR signal intensity (low field peak or mid field peak of the spectrum) to determine the antioxidative potential of a biological system ) measured after a defined time t. This relative intensity I r is set as "0". To determine the relative antioxidative potential, the size A p is defined, which is a measure of the relative AOP:
Ap(t, k) = 1 - Ir(t, k)
A p (t, k) = 1 - I r (t, k)
Zur Bestimmung der relativen Signalintensität Ir werden die Para meter Meßzeitpunkt t und Modellradikalkonzentration k für eine Meßreihe konstant gehalten.where k is a measure of the concentration of the model used radical. Corresponding to I r = 0, A p = 1 results. To determine the relative signal intensity I r , the parameters measuring time t and model radical concentration k are kept constant for a series of measurements.
In a second step, the biological system is exposed to the oxidative stress to be investigated, which can be, for example, an endogenous inflammatory reaction or exogenous influence by electromagnetic radiation (UV radiation, radioactive radiation, thermal radiation) or mechanical influences (impact, pressure) , The increased oxidative stress leads to a slowed down degradation of the model radical subsequently supplied. Depending on the magnitude of the oxidative stress, the measured intensity I r will be greater than 0. Accordingly, the antioxidative potential of the investigated biological system is less than 1.
Ap steigt an.If antioxidants such as ascorbate and / or tocopherol are added to the biological system under consideration, the AOP is increased again, ie A p increases.
The diversity of the physiologically occurring Antioxidants, which are also caused by a variety of reactive oxidants, leads to different reactions speed with the model radicals. So reduced at for example, ascorbate prefers piperidine nitroxides, whereas the reaction rate with pyrrolidine nitroxides is slow is. In contrast to pyrrolidine, piperidine nitroxides are Nitroxides NADPN-dependent through thioredoxin reductase redu ed. Thioredoxin reductase complements the ascorbate as Reducing agent for nitroxide radicals. The nitroxide reduction is stimulated for example by NADPH, NADP and NADH. Liposomally bound nitroxide can be caused by ascorbate, but not can be reduced by thioredoxin reductase.
The relatively high reduction rate of pyrrolidine nitroxides in the skin suggests that there are other Reduk tantien as ascorbate and thioredoxin are present.
The model radical used contains semi-stable nitro xid radicals, their high chemical selectivity a reduction through enzymes and specific antioxidants like glutathione, Ascorbate and tocopherol favored. By selecting individual Test systems such as piperidine nitroxide can make the AOP more appropriate Antioxidants such as B. ascorbate can be measured selectively.
By using perdeuterated and 15 N-labeled nitroxide radicals, the signal intensity and thus the sensitivity of the process can be increased many times over.
Another improvement of the process is that with the help of EPR tomography the antioxidative potential can be spatially resolved. A particularly cheap ver The driving variant is the one with modulated gradients. This The variant enables the antioxidati to be measured in layers potential in the skin, for example, which corresponds to appropriate conclusions on the efficiency of medical or cosmetic metallic treatment of the skin can be drawn. A further explanation of this procedure is already in the cited literature Magnetic Resonance Microscopy ent to which expressly refer in this context is taken.
With the present invention, the term becomes the first of the antioxidative potential introduced and at the same time simple and well comparable method offering measurement results provided. The model substances used are in the applied concentration is skin-friendly and becomes easily broken down by antioxidants. The procedure is repeatable, while previous in vitro procedures always only the individual detection of an antioxidant within a be agreed tissue sample and thus a condition documented "to". The new process therefore offers one hitherto unique possibility of antioxidation on living things potentials not only of the skin itself, but also potentials in different layer depths and thus the Depth of penetration of cosmetically or medically applied Antioxidants.
In der dazugehörigen Zeichnung istThe invention is intended to be explained in more detail below by examples to be refined. In the accompanying drawing is
Fig. 1 eine grafische Darstellung des in den Beispielen 1, 2 und 3 mit der dazugehörigen Kurve 1, 2 und 3 gemesse nen AOP über die Gesamtzeit Fig. 1 is a graphical representation of the AOP measured in Examples 1, 2 and 3 with the associated curve 1, 2 and 3 over the total time
To measure the AOP on the skin, the AOP was determined using the Decrease in the radical concentration of the model radical system CAT 1 measured for three different exogenous influencing factors.
The skin was irradiated with UVA / B. After a triple MED (minimal erythema dose), the model substance CAT 1 was applied transdermally in a microemulsion with PEG 80 as a carrier substance in a concentration of 10 mmol. The concentration of the model radical was measured every 5 minutes; Microwave power 20 mM; Frequency 3.5 GHz. After t = 25 minutes, the radical concentration was still Ir = 0.23. Then in this case an A p = 0.77 was obtained after 25 minutes.
In this example, a substance was added to the skin with 8-methoxy-psoralen, which favors the formation of reactive oxidants under UV radiation. After the subsequent application of the model radical as in Example 1 and measurements at intervals of 5 minutes, the radical concentration after 25 minutes was still Ir = 0.36, so that A p = 0.64 resulted.
In this example, the skin was treated with an antioxidant composition (containing, for example, tocopherol, ascorbate) before the radiation. The measurement carried out with the model substance as in Example 1 after irradiation at the same dose as in Example 1 showed the radical signal to drop to 6% after 25 minutes. This resulted in an antioxidative potential of A p = 0.94. The antioxidative potential measured on the skin before the radiation had a value A p = 0.86. The comparison of the A p values clearly shows an increase in the AOP in the skin after the external use of antioxidants and thus an exact reflection of the change in the AOP, which could be determined by the method according to the invention.
