The Dictyostelium discoideum transformation was carried out as published (Gene 59, 99; Gene 39, 155; Mol. Cell. Biol. 4, 2890). Transformants were in the presence of Klebsiella aerogenes cloned (DNA 8, 193; Gene 85, 353). The plasmid used for transformation DNA was stably integrated into the genome in transformants and was amplified 10 to 100 times.

Durch Mißpaarung im 5′-Primer an den Positionen 7, 8, 20, 26 und 32 wurden die Codons innerhalb der ersten 8 Aminosäuren an den Codon-Gebrauch von Dictyostelium discoideum angepaßt. Ferner wurde durch die Sequenz des Primers am unmittelbaren 5′-Ende der aufamplifizierten cDNA eine EcoRI-Restriktionsschnittstelle eingeführt. Durch die Sequenz des 3′-Primers wurde am 3′-Ende der aufamplifizierten cDNA eine XbaI-Restriktionsschnittstelle eingeführt.The physical map of the plasmid pDNeo2.hm2 is shown in Fig. 1.
