B. extra chromosomal replizierende Vektoren, die aus Pflanzenviren ent wickelt wurden (vergl. J.W. Davies und J. Stanley: "Geminivirus genes and vectors". Trends Genet. 5: 77-81, 1989), benutzt werden, um Fremdgene ohne Integration in Pflanzen zu exprimie ren. ist die Gesamtsequenz das TVMV bekannt. Dazu wurden DNA-Kopien
(cDNA) der genomischen TVMV-RNA hergestellt und deren Sequenz
bestimmt. Eine 4,7 kb große TVMV cDNA im Plasmid pTVBam7 (ent
sprechend der cDNA-Sequenz zwischen den BamHI-Schnittstellen,
Nukleotide 1934 bis 6648; Fig. 1 oben) wurde von Dr. Rhoads zur
Verfügung gestellt, und aus diesem Plasmid ein 3,3 kb großes
DraIII/EcoRV-Fragment isoliert (Fig. 1 unten). Nach den Anga
ben in der zitierten Arbeit ist diese 3,3 kb große cDNA aus dem
Klon pTVBam7 durch eine Nukleinsäure- und eine daraus resultie
rende Aminosäuresequenz näher definiert und enthält ein konti
nuierliches offenes Leseraster (ORF), das für das P3-Protein
(42 kDa), ein 6K1-Protein (6 kDa) sowie das zytoplasmatische
Inklusionsprotein CI (67 kDa) kodiert.From a work by

Desiree) übertragen. Die nach Selektion mit dem Antibiotikum Hygromycin erhaltenen Kalli wurden in Ge webekultur zu Pflanzen regeneriert und in vitro für einen er sten Protein-Test durch hohe Saccharose-Konzentrationen zur Bildung von Knollen angeregt. Obwohl die Ergebnisse der Pro teinbestimmungen an den Knollen von 10 unabhängigen Transfor manden mit dem waxy/CI-Konstrukt sowie von 6 Transformanden mit dem CaMV 35S/CI-Konstrukt einen dramatischen Anstieg der Pro teinkonzentration aufwiesen, zeigten alle Transformanden im Vergleich zur nicht-transformierten Kontrolle Desiree einen ab solut anomalen Phänotyp. Um einen Artefakt auszuschließen, wur den in vitro-Knollen für die wx/CI-Linien 1, 4, 8, 9, 10 im Ge wächshaus in normaler Erde zum Keimen gebracht und die abge reiften Knollen einer weiteren Analyse unterzogen. Als Ausgangsmaterial diente der von Dr. R.E. Rhoads (Universi tät Kentucky) zur Verfügung gestellte Plasmidklon pTVBam7. Die ser enthält ≈4,7 kb (entsprechend den Nukleotiden 1934 bis 6648) der TVMV-Sequenz (Domier u. a., Nucleic Acids Res. (1986), 14: S. 5417-5430, 1986) in der BamHI-Schnittstelle des pUc119. DNA dieses Klons wurde mit den Restriktionsenzymen DraIII und EcoRV geschnitten. Nach Auftrennung dieses Restriktionsansatzes auf einem 1%igen Agarosegel wurde eine DNA-Bande von 3,3 kb ausgeschnitten. Die DNA wurde aus dem Gel elektroeluiert und über DEAE-Cellulose gereinigt. Das 3,3 kb große DraIII/EcoRV- Fragment umfaßt die Sequenz für die Helferkomponente (120 3′- terminale Nukleotide), die vollständige P3-Sequenz und die ge samte für das zytoplasmatische Inklusionsprotein (CI) kodieren de Sequenz bis auf die 3′-terminalen 23 Nukleotide. Zur Subklo nierung wurde das 3,3 kb große DraIII/EcoRV-Fragment mit dem Enzym mung bean nuclease behandelt. Die notwendige Terminatorsequenz wurde aus dem zuvor schon erwähnten pBR-Plasmid (1) entnommen. Zu diesem Zweck wur de das Plasmid (1) mit dem Restriktionsenzym BamHI zwischen der CaNV 35S-Promotor und Terminatorsequenz geöffnet und mit dem Restriktionsenzym EcoRI nachgeschnitten. Die Aufreinigung des 0,25 kb großen BamHI/EcoRI-Terminatorfragments erfolgte wie un ter 2a) beschrieben. Zu je weils 200 µl dieser Suspension wurden 200 ng der unter 2c) be schriebenen Konstrukte hinzugegeben und in flüssigem Stickstoff eingefroren. Nach einem Hitzeschock von 5 min bei 37°C wurde 0,8 ml YEB-Medium zugegeben und 1 h bei 28°C inkubiert. Da die T-DNA des Plasmids pGDW32 ein Hygromycin-Resistenzgen trägt, konnte mit Hygromycin auf solche Pflanzenzellen selektioniert werden, die diese T-DNA aufgenommen hatten. Alle 14 Tage wurden die Blätter auf frische Platten transferiert. Nach der Sproßbildung wurden diese vom kallösen Gewebe abgeschnitten und auf MSIII-Platten zur Bewurzelung gebracht. Zur Induktion von in vitro-Knollen bildung wurden Internodien der bewurzelten Sprößlinge auf MS8- Platten gesetzt. Analog dazu wurden Internodien von nicht transformierten Wildtyp-Kartoffelpflanzen (cv. Transformierte Pflanzen wurden durch DNA-Analyse ("Southern blotting") auf das Vorhandensein der entsprechenden Promo tor/3,3 kb TVMV CI-Fragment/Terminator-Konstrukte überprüft. Hierzu wurden je 400 mg Pflanzenmaterial mit Quarzsand zermörsert (vgl. C. Lawson u. a.: "Engineering resistance to mixed virus infection in a commer cial potato cultivar: resistance to potato virus X and potato virus Y in transgenic Russet Burbank." Bio/Technology 8: 127-134, 1990). Nach Zusatz von 3 ml Laemmli-Puffer (62,5 mM TRIS-HCl pH 8, 2% SDS, 10% Glyzerin, 1% DTT, 5% Merkapto äthanol) wurden die Proben 15 Minuten bei 100°C erhitzt und da nach Quarzsand und feste Pflanzenrückstände durch Zentrifuga tion (15 Minuten, 4000 Upm) entfernt. Die Proteinbestimmung erfolgte nach Bradford (vgl. M.M. Bradford: "A rapid and sensitive method for quantitation of microgram quantities of proteins using the principle of protein-dye bin ding." After adding 3 ml of Laemmli buffer (62.5 mM TRIS-HCl pH 8, 2% SDS, 10% glycerin, 1% DTT, 5% mercapto ethanol), the samples were heated at 100 ° C for 15 minutes and then after quartz sand and solid plant residues removed by centrifugation (15 minutes, 4000 rpm). For the gel electrophoretic detection of the expressed TVMV proteins (P3 + CI, CI, P3; note: the P3 protein is obviously a protease and cleaves from the P3 + CI precursor), part of the extract was precipitated again, in Laemmli Buffer denatured by heating and separated on a 10% polyacrylamide gel (cf. UK Laemmli: "Cleavage of structural proteins during the assembly of the head of bacteriophage T4." Nature 227: 681-685, 1970). The separated proteins were made visible by staining with Coomassie blue.
