For site-specific recombination (see Figure 1, showing the plasmids), the gnd gene (encoding the 6-phosphogluconate dehydrogenase, part of the pentose-phosphate cycle) was selected. This gene (1.444 kb) is located in Salmonella and in E. coli between the His and rfb genes, and DNA sequence analysis has shown that this region is highly conserved in both species (Barcak and Wolf 1988, Reeves and Stevenson 1989). Plasmid pMN6 ( Figure 1) carries the gnd locus together with some flanking DNA. The 741 bp gnd fragment, which was found after partial digestion of PStI / KpnI from pMN6, was purified by gel electrophoresis and subcloned into PstI / KpnI sites of plasmid pUC19, where the plasmid pHB100 was obtained. The gnd segment was further subcloned into the corresponding cleavage site of the suicide vector pGP704 as the EcoRI / SphI fragment, resulting in the plasmid pHB101. The XbaI site of the vector was then removed by XbaI digestion of plasmid pHB101, blunt-filling with the Klenow fragment of DNA polymerase I and religating. The resulting plasmid pHB102 was cleaved with EcoRV, ligated to phosphorylated XbaI linkers, digested with XbaI, and religated to give plasmid pHB103 carrying a single XbaI site on the gnd gene. Then, plasmid pSS37 was cleaved with EcoRV, ligated to the phosphorylated XbaI linker and digested with XbaI, and the rfb rfp cassette was subcloned into Xbal digested plasmid pHB103, where the plasmid pHB107 was produced. Der rfa-Genort von E. coli-K-12 wurde aus einer Genbank von Cos midklonen isoliert, die von Birkenbihl und Kollegen (1989) kon struiert worden war, und auf genetischer Ebene analysiert. Die Gene rfaG, rfaM, rfaN und rfaL wurden durch Kreuz-Komplementa tion identifiziert, wobei die in den entsprechenden Genen des rfa-Bereichs mutierten Salmonella typhimurium-Stämme auf die Produktion von vollständigem LPS analysiert wurden, indem die K- 12 rfa-Analoga in die mutierten Stämme transferiert wurden. Nachdem das rfaL-Gen von K-12 in S. typhi Ty21a, welches die rfb-rfp-Genkassette im Chromosom trägt, transferiert worden war, wurde gefunden, daß der Hybridstamm das O-Polysaccharid von S. dysenteriae 1 mit dem Core/Lipid A von S. typhi verknüpfte. Der Trägerstamm Ty21a exprimierte auch homologes LPS. Es scheint deshalb, daß es nicht notwendig ist, den S. typhi-Kern auf ir gendeine Weise zu modifizieren, und daß ein möglicher Grund, warum heterologe O-Polysaccharide nicht an den Kern von Ty21a gebunden werden, derjenige ist, daß die O-Antigen/Core-Ligase (das rfaL-Genprodukt) eine enge, stringente Spezifität aufweist und demzufolge einzig das homologe O-Antigen an den Kern binden kann. Auf der anderen Seite könnten die O-Antigen/Core-Ligasen von E. Coli-K-12 und S. typhimurium erweiterte, relaxierte Spe zifität zeigen und demzufolge in der Lage sein, eine Vielzahl heterologer O-Polysaccharide an die Kernstrukturen des Wirts zu binden. Diese Eigenschaften sind nicht nur vom Standpunkt der Vakzin-Entwicklung aus von Interesse, sondern auch bezüglich der Evolution der O-Antigen/Core-Ligase. In der Tat scheint auch nach unseren Ergebnissen bezüglich der Kreuz-Komplemententation der äußeren Kernstrukturen eine relaxierte Spezifität bei diesen En zymen vorhanden zu sein, da das Substituieren der rfa-Funktion der Salmonella typhimurium-Mutante durch ein Analogon aus E. coli-K-12 zur Vervollständigung der Kernstruktur führt, wobei im Wesentlichen ein Enzym aus E. coli-K-12 beteiligt ist, während der Rest aus S. typhimurium stammt. Dies ist vom Evolutions standpunkt aus von Interesse, da es gut bekannt ist, daß die Kohlehydratkette von LPS eine enorme Heterogenität aufweist. Ein weiteres Problem, das bei der Konstruktion einer bivalenten S. dysenteriae 1/S. typhi-Vakzine auftrat, war, daß das Plasmid pSS37, das die für die O-Polysaccharide von S. dysenteriae 1 ko dierenden rfb-rfp-Gene trägt, in S. typhi Ty21a instabil ist. Dieses Problem wurde vorliegend dadurch gelöst, daß ein chromo somales Integrations-Vektorsystem auf Transposonbasis verwendet wurde, um die rfb-rfp-Gene in das Chromoson von Ty21a zu inser tieren. Fig. 2 zeigt eine physikalische Karte des E. coli-K-12 rfa-Gen ortes und Plasmid-Konstruktionen innerhalb dieser Region. Die oberste Reihe zeigt die Chromosomen-Karte von E. coli-K-12 im Bereich der 82. Minute (ein Teil davon wurde bereits von Austin et al., 1990, gezeigt). Der rfg-Genort wird vom tdh-Gen (welches für die 2-Amino-3-ketobutyrat-Coenzym A Ligase kodiert; Aronson et al. 1988) und das fpg-Gen (welches für die Formamidopyrimi din-DNA-Glycosylase kodiert; Boiteux et al. 1987) flankiert. Die Lokalisierung des fpg-Gens ist in der achten Ausgabe der E. coli-K-12 Linkage Map (Bachmann 1990) nicht dargestellt; wenn man jedoch die Kartierungs- und die DNA-Sequenzierungsdaten von Boiteux und Kollegen (1987) auswertet, so kann man klar erken nen, daß das fpg-Gen die BamHI-Schnittstelle in der 5′-proxima len Region des rfa-Genortes überspannt, wie oben dargestellt. Die Auswertung der DNA-Sequenz des fpg-Gens und die Annahme, daß das 5′-Ende von rfa direkt in 3′-Richtung neben dem fpg-Gen be ginnt, sollte nahelegen, daß das 5′-Ende von rfa mindestens 300 Bp links von der BamHI-Schnittstelle (Position 0) liegt. Die Re striktionsschnittstellen sind die folgenden: (A) ApaI, (Ac) AccI, (Av) AvaI, (B) BstEII, (Bg) BglII, (Bin) BamHI, (C) ClaI, (E) EcoRI, (EV) EcoRV, (H) HindIII, (Hc) HincII, (Hp) HpaI, (M) MluI, (N) NcoI, (P) PstI, (Pv) PvuII, (S) SalI, (Sc) ScaI, (X) XbaI, (Xh) XhoI. Die Positionen (Werte in kB) der wesentlichen Restriktionsenzym-Schnittstellen (auf die im Text Bezug genommen wird) sind unterhalb der jeweiligen Enzyme angegeben, wobei die BamHI-Schnittstelle im fpg-Gen mit der Position "0" und die EcoRI-Schnittstelle zwischen tdh und rfa mit 18,4 bezeichnet werden. Die Restriktionsenzym-Schnittstellen, die zwischen BamHI (0) und BglII (1,1) dargestellt sind, wurden von Austin und Kol legen (1990) erhalten. In der Region zwischen HindIII (9,7) und EcoRI (18,4) wurden keine Restriktionsenzyme-Schnittstellen für BamHI, KpnI, NcoI, PstI, SacI, SalI und XbaI gefunden. Die En zyme AccI, AvaI, BstEII, HincII, NcoI und PvuII, die in der frü her veröffentlichten Karte verwendet wurden (Austin et al. 1990), wurden nicht für die Kartierung von Schnittstellen zwi schen HindIII (9,7) und EcoRI (18,4) verwendet. Figure 2 shows a physical map of the E. coli K-12 rfa gene locus and plasmid constructions within this region. The top row shows the chromosomal map of E. coli K-12 in the 82th minute range (part of which has already been shown by Austin et al., 1990). The rfg locus is derived from the tdh gene (which encodes the 2-amino-3-ketobutyrate coenzyme A ligase, Aronson et al., 1988) and the fpg gene (which encodes the formamidopyrimidine din glycosylase, Boiteux flanked et al., 1987). The location of the fpg gene is not shown in the eighth edition of the E. coli K-12 Linkage Map (Bachmann 1990); however, when evaluating the mapping and DNA sequencing data of Boiteux and colleagues (1987), it can be clearly seen that the fpg gene spans the BamHI site in the 5 'proximal region of the rfa locus, as shown above. The evaluation of the DNA sequence of the fpg gene and the assumption that the 5 'end of rfa starts directly in the 3' direction next to the fpg gene should suggest that the 5 'end of rfa is at least 300 bp to the left of the BamHI interface (position 0). The restriction sites are as follows: (A) ApaI, (Ac) AccI, (Av) AvaI, (B) BstEII, (Bg) BglII, (Bin) BamHI, (C) ClaI, (E) EcoRI, (EV) EcoRV, (H) HindIII, (Hc) HincII, (Hp) HpaI, (M) MluI, (N) NcoI, (P) PstI, (Pv) PvuII, (S) SalI, (Sc) ScaI, (X) XbaI, (Xh) XhoI. The positions (values in kb) of the major restriction enzyme sites (referred to in the text) are indicated below the respective enzymes, with the BamHI site in the fpg gene at position "0" and the EcoRI site between tdh and rfa be designated 18.4. The restriction enzyme sites shown between BamHI (0) and BglII (1,1) were obtained from Austin and Kolsche (1990). In the region between HindIII (9.7) and EcoRI (18.4), no restriction enzyme sites were found for BamHI, KpnI, NcoI, PstI, SacI, SalI and XbaI. The enzymes AccI, AvaI, BstEII, HincII, NcoI and PvuII used in the earlier published map (Austin et al., 1990) were not used for the mapping of HindIII (9.7) and EcoRI (FIG. 18.4). The enzymes Apal, ClaI, EcoRV, ScaI and XhoI have not been mapped in the region between BamHI (0) and BglII (11,1), and there are no MluI sites in the same region. The presence or absence of the rfaG-I, J and L enzymes (marked + and -, respectively) encoded by the various clones is shown.
In Fig. 3 sind die chemischen Strukturen der LPS-Kerne von E.
coli-K-12 und Salmonella typhimurium zusammen mit den jeweiligen
rfa-Genen gezeigt, welche für die Enzyme kodieren, die am jewei
ligen Schritt der Kern-Biosynthese beteiligt sind. Die Ligierung
von O-Antigen an den Kern erfolgt mit Hilfe von 2 Enzymen, von
denen eines durch rfaL und das andere durch ein rfb-Gen, rfp-T,
kodiert wird. Gestrichelte Bindungen stellen unvollständige Sub
stitutionen dar.

Nummer 1 zeigt Bakterien, die ohne Galactose und doppelt mar

waren. Die Bakterien der Nummer 2 wurden in Anwesenheit von Ga
lactose und wie oben beschrieben doppelt markiert vermehrt. Zu
erst wurde das O-Antiserum von S. dysenteriae verwendet, gefolgt
vom O-Antiserum vom S. typhi.
Nummer 3 unterscheidet sich von Nummer 2 nur dadurch, daß die
Reihenfolge der Antiseren umgedreht wurde. Fig. 5C Here, strain H4098 / pHB139 is shown. Number 1 shows bacteria that had been bred without galactose and doubly labeled with S. typhi O antiserum (10 nm gold particles) and S. dysenteriae O antiserum (5 nm gold particles). The number 2 bacteria were propagated in the presence of galactose and double labeled as described above. First, the O antiserum of S. dysenteriae was used, followed by the O antiserum of S. typhi.
Number 3 differs from number 2 only in that the order of the antisera was reversed.
